An analytical biomarker for treatment of patients with recurrent B-ALL after remission induced by infusion of anti-CD19 chimeric antigen receptor T(CAR-T) cells

Anti-CD19 chimeric antigen receptor-modified T(CAR-T-19) cells have emerged as a powerful targeted immunotherapy for B-cell lineage acute lymphoblastic leukemia with a remarkable clinical response in recent trials. Nonetheless, few data are available on the subsequent clinical monitoring and treatment of the patients, especially those with disease recurrence after CAR-T-19 cell infusion. Here, we analyzed three patients who survived after our phase I clinical trial and who were studied by means of biomarkers reflecting persistence of CAR-T-19 cells in vivo and predictive factors directing further treatment. One patient achieved 9-week sustained complete remission and subsequently received an allogeneic hematopoietic stem cell transplant. Another patient who showed relapse after 20 weeks without detectable leukemia in the cerebrospinal fluid after CAR-T-19 cell treatment was able to achieve a morphological remission under the influence of stand-alone low-dose chemotherapeutic agents. The third patient gradually developed extensive extramedullary involvement in tissues with scarce immune-cell infiltration during a long period of hematopoietic remission after CAR-T-19 cell therapy. Long-term and discontinuous increases in serum cytokines(mainly interleukin 6 and C-reactive protein) were identified in two patients(Nos. 1 and 6) even though only a low copy number of CAR molecules could be detected in their peripheral blood. This finding was suggestive of persistent functional activity of CAR-T-19 cells. Combined analyses of laboratory biomarkers with their clinical manifestations before and after salvage treatment showed that the persistent immunosurveillance mediated by CAR-T-19 cells would inevitably potentiate the leukemia-killing effectiveness of subsequent chemotherapy in patients who showed relapse after CAR-T-19-induced remission.

Enhanced autophagy protects hepatic cells from radiation injury

ObjectiveTo study the influence of radiation on autophagy and its protective effect on radiation injury of hepatic cells.MethodsAutophagy in mouse liver tissues was examined by GFP-LC3 staining and Western blot. Radiation-induced hepatic injury was evaluated by ALT and AST in mouse serum, protein expressions, and H & E and TUNEL staining of liver tissue. L02 cells were used for in vitro study. Chloroquine and rapamycin were used to manipulate the level of autophagy. ResultsTotal body irradiation (TBI) of 8 Gy caused an increase of autophagy in mouse liver tissue and AST level in serum( t＝－7.47, P<0.05) at 12 h after irradiation. Irradiation significantly increased the apoptotic level in liver tissue as well. Inhibition of autophagy by chloroquine caused a further increases of AST [IR: (345.42±35.25)U/L vs. IR＋CQ: (433.42±40.07)U/L, t＝－2.86, P<0.05] and ALT [IR: (35.67±8.08)U/L vs. IR＋CQ: (98.5±26.67)U/L, t＝－3.09, P<0.05] in the serum, and it also promoted apoptosis in live tissue. However, rapamycin as an autophagy promoter showed protective effect for radiation-induced hepatic injury [AST: IR: (345.42±35.25)U/L vs. IR＋Rap: (278.42±20.09)U/L, t＝－2.86, P<0.05]. Similar changes of autophagy and apoptosis in L02 cells were also observed in the cells treated with chloroquine and rapamycin. Inhibition of autophagy by CQ caused an increase of ROS in vitro and in vivo and further increased ALT and AST levels in serum, reduced L02 cell viability. Activation of autophagy by Rap effectively reversed those changes. ConclusionsAutophagy protects hepatic cells from radiation injury by decreasing ROS induction, which provides a potential target for the development of new clinical regimens against radiation induced liver injury.

两级式光伏并网系统将光伏电能汇集至低压直流母线,通过并网逆变器送入交流电网,低压直流母线能满足本地负载需求,实现光伏就地消纳,而且相比传统分散逆变、就地并网的方案具有更高的发电能效。两级式光伏并网系统在交流电网电压发生不平衡跌落时,存在低压直流母线电压上升以及母线电压二倍频波动问题。电压上升可能导致光伏脱网,不满足光伏并网的低电压穿越要求;电压上升及二倍频波动也不利于直流负载的稳定工作。为了稳定电网电压不平衡跌落下直流母线电压,针对前级DC/DC(direct current to direct current)变换器采用了最大功率点跟踪与定低压直流母线电压控制模式切换策略,并在此基础上增加了比例谐振控制,解决了低压直流母线电压上升及二倍频波动问题,保证了电压的稳定,提高了电能质量。以电网电压发生单相跌落为例,进行了仿真和试验验证,结果证明了该控制策略的有效性。

环境中微量新污染物的存在及风险已受到广泛关注。城镇污水处理厂作为微量新污染物的重要“汇”,是阻断微量新污染物向环境中排放的关键屏障。依托“十二五”和“十三五”国家水体污染控制与治理科技重大专项,在全国不同地域的城镇污水处理厂开展了较系统、全面的试验研究与统计分析,明确了我国城镇污水处理厂中典型微量新污染物的赋存特征,并在解析其在全工艺流程中的迁移转化规律的基础上,尝试提出了城镇污水处理厂微量新污染物的去除途径。研究成果为城镇污水处理厂微量新污染物控制提供了重要的科学基础,为城镇污水净化处理目标从常规指标升级到微量新污染物提供了可能性和全过程控制技术路径。

为提高空间调制系统信号检测算法的性能,提出基于粒子群的智能信号检测算法及其改进算法.利用粒子智能化搜索,实现信号高效检测;设计权重系数对传统速度更新公式进行修改,避免粒子群陷入局部收敛从而进一步提高算法的检测性能.通过对改进算法的收敛性和复杂度进行理论分析,并在不同天线数目和不同调制方式下对其误码性能进行仿真,仿真结果表明:与传统的粒子群算法相比,本文提出的改进算法具有计算复杂度低、误码率低、收敛快的优点,可作为空间调制接收机的有效备选算法.

Umbilical cord blood (UCB) is a current major source of hematopoietic stem cells (HSCs) for cell transplantation therapy. Cell transplantation with HSCs derived from UCB is advantageous over transplantation with HSCs from adult tissues. However, the low number of HSC derived from a single unit of UCB limits its application. Thus, ex vivo expansion is a good option to create more UCB HSCs for clinical application. The strategies for HSC expansion in vitro focus on mimicking the composition and structure of HSC natural niche by enhancing self-renewal and inhibiting lineage differentiation of HSCs. In the past decade, the mechanisms of the interaction between HSC and the natural niche have been deeply investigated. This great progress in basic research has led to advancements in UCB HSC ex vivo expansion. In addition, the biological characteristics of the originally isolated UCB HSCs correlate with outcome of subsequent ex vivo expansion. In this paper, we summarize the late progress achieved in isolation and ex vivo expansion of UCB HSCs. Importantly, we attempt to provide an impact and practicable procedure to expand UCB HSC in vitro from isolation of original HSCs to identification of expanded HSCs.

基于核密度函数的多尺度北京市休闲农业空间分布分析

核密度函数估计法是常用的分析城市点要素空间分布模式的方法。使用核密度函数估计法的关键是最佳带宽的确定。目前,大多数研究使用单纯基于数学的方法或目视判读法确定核密度函数的最适带宽,但是针对同一地理实体在不同分析尺度上所对应的核密度函数适用带宽确定方法问题研究相对不足。该文考虑了北京市休闲农业本身所具有的尺度特征,基于北京市休闲农业POI数据,使用核密度函数评估方法,识别并分析其多尺度空间分布模式,并使用文献求证法进行了验证。使用Moran’I、HH个数和Comprehensive I指数曲线相结合的方法确定了适合分析北京市休闲农业区域尺度和局部尺度空间分布模式的核密度函数所对应的带宽,分别为9 km和3 km。进而分析了北京市休闲农业多尺度空间分布模式。北京市休闲农业空间分布模式在区域尺度上形成了2个圈层及多个聚集中心的结构。距离城市中心约30～50 km的圈层为1日游圈层。距离城市中心约50～90 km的圈层为2日游或多日游圈层。1个主中心位于昌平区东部、怀柔区东南部、密云区西南部的山前平原地区。2个副中心分别位于密云区东北部和房山区西南部;在局部尺度上形成了3个等级的26个小的聚集中心,第一等级2个,第二等级3个,第三等级21个,主要位于中北部的山前平原地区,东南部城乡交错带的平原地区及西部山区分布相对较少,中心城区分布最少。该研究可为北京市休闲农业空间规划提供重要参考依据。

基于DSSAT作物模型的中美大豆主产区单产模拟与验证

开展基于作物模型的大面积作物产量估测研究,可以为及时掌握全球重点地区农作物的生产情况提供数据支撑。该研究以大豆为监测作物,选取中国吉林省和美国爱荷华州作为研究区域,基于DSSAT作物估产模型中的SOYGRO大豆模型,利用分辨率为0.5°×0.5°的生育期气象要素以及500 m×500 m绿色叶绿素植被指数,进行遥感数据融合作物模型估测大豆单位产量的模拟与验证研究。结果显示,2008-2017年,美国爱荷华州大豆单位产量模拟值的平均误差为16.8%,均方根误差为762.8 kg/hm^(2),平均偏差为107.2 kg/hm^(2);中国吉林省大豆单位产量估测的平均误差为36.3%,均方根误差为1088.4 kg/hm^(2),平均偏差为-237.9 kg/hm^(2)。在县域尺度下,大豆单位产量模拟值与调查值的拟合度较好,尤其在产量较低的年份,其中美国爱荷华州的产量相关系数最高可达0.78,中国吉林省的相关系数偏小,为0.59,表明对美国爱荷华州大豆单位产量的估测精度优于中国吉林省。研究所建立的大豆单位产量估测技术路线,可以为中美两国主产区作物单位产量的大面积有效估测提供参考。

针对异源图像配准中传感器物理特性差异造成的待匹配特征维度较高、稳定性较弱、配准质量较差等问题,提出一种基于尺度不变特征变换(SIFT)的异源图像配准方法。该方法首先结合相位一致性和改进的SIFT算法获得稳定的特征,然后利用最近邻距离比方法进行初匹配,接着提出了一种联合误差与欧氏距离(JEED)方法进行再匹配,最后采用模式搜索尺度不变特征变换(MS-SIFT)方法优化匹配点对以提高图像配准质量。实验结果表明,相比于现有方法,所提方法能够提取可靠稳定的特征,获得了较高配准质量,同时提高了配准算法的实时性。
