Identification and profile of phenolamides with anthracnose resistance potential in tea(Camellia sinensis)

Tea anthracnose is a prevalent disease in China that can lead to reduced tea production and lower quality,yet there is currently a lack of effective means for controlling this disease.In this study,we identified 46 phenolamides(including 27 isomers)in different tissues and organs of tea plants based on a developed workflow,and the secondary mass spectra of all these compounds have been documented.It was revealed that tea plants predominantly accumulate protonated aliphatic phenolamides,rather than aromatic phenolamides.The profile of phenolamides indicate that their buildup in tea plants is specific to certain tissues and acyl-acceptors,and this distribution is associated with the extent of phenolamide acyl-modification.Additionally,it was observed that N-Feruloylputrescine(Fer-Put,a type of phenolamides)was responsive to the stimulated accumulation of the tea anthracnose pathogen.The findings of anti-anthracnose experiments in vitro and on tea leaf demonstrated that Fer-Put was capable of significantly inhibiting the growth of anthracnose pathogen colony,effectively prevented tea leaf disease.Furthermore,it was observed that Fer-Put treatment can enhance the antioxidant enzyme activity of tea leaves.TEA002780.1 and TEA013165.1 gene may be responsible for the biosynthesis of Fer-Put in the disease resistance process in tea plants.Through these studies,the types and distribution of phenolamides in tea plants have been elucidated,and Fer-Put’s ability to resist anthracnose has been established,providing new insights into the resistance of tea anthracnose.

Interactive response of photosynthetic characteristics in Haloxylon ammodendron and Hedysarum scoparium exposed to soil water and air vapor pressure deficits

C4 plants possess better drought tolerance than C3 plants. However, Hedysarum scoparium, a C3 species, is dominant and widely distributed in the desert areas of northwestern China due to its strong drought tolerance. This study compared it with Haloxylon ammodendron, a C4 species, regarding the interactive effects of drought stress and different leaf–air vapor pressure deficits. Variables of interest included gas exchange, the activity levels of key C4 photosynthetic enzymes, and cellular anatomy. In both species, gas exchange parameters were more sensitive to high vapor pressure deficit than to strong water stress, and the net CO2 assimilation rate（A n） was enhanced as vapor pressure deficits increased. A close relationship between A n and stomatal conductance（g s） suggested that the species shared a similar response mechanism. In H. ammodendron, the activity levels of key C4 enzymes were higher, including those of phosphoenolpyruvate carboxylase（PEPC） and nicotinamide adenine dinucleotide phosphate-malate enzyme（NADP-ME）, whereas in H. scoparium, the activity level of nicotinamide adenine dinucleotide-malate enzyme（NAD-ME） was higher.Meanwhile, H. scoparium utilized adaptive structural features, including a larger relative vessel area and a shorter distance from vein to stomata, which facilitated the movement of water. These findings implied that some C4 biochemical pathways were present in H. scoparium to respond to environmental challenges.

Effects of soil water and nitrogen availability on photosynthesis and water use efficiency of Robinia pseudoacacia seedlings

The efficient use of water and nitrogen （N） to promote growth and increase yield of fruit trees and crops is well studied.However,little is known about their effects on woody plants growing in arid and semiarid areas with limited water and N availability.To examine the effects of water and N supply on early growth and water use efficiency （WUE） of trees on dry soils,one-year-old seedlings of Robinia pseudoacacia were exposed to three soil water contents （non-limiting,medium drought,and severe drought） as well as to low and high N levels,for four months.Photosynthetic parameters,leaf instantaneous WUE （WUEi） and whole tree WUE （WUEb） were determined.Results showed that,independent of N levels,increasing soil water content enhanced the tree transpiration rate （Tr）,stomatal conductance （Gs）,intercellular CO2 concentration （Ci）,maximum net assimilation rate （Amax）,apparent quantum yield （AQY）,the range of photosynthetically active radiation （PAR） due to both reduced light compensation point and enhanced light saturation point,and dark respiration rate （Rd）,resulting in a higher net photosynthetic rate （Pn） and a significantly increased whole tree biomass.Consequently,WUEi and WUEb were reduced at low N,whereas WUE i was enhanced at high N levels.Irrespective of soil water availability,N supply enhanced Pn in association with an increase of Gs and Ci and a decrease of the stomatal limitation value （Ls）,while Tr remained unchanged.Biomass and WUEi increased under non-limiting water conditions and medium drought,as well as WUEb under all water conditions;but under severe drought,WUEi and biomass were not affected by N application.In conclusion,increasing soil water availability improves photosynthetic capacity and biomass accumulation under low and high N levels,but its effects on WUE vary with soil N levels.N supply increased Pn and WUE,but under severe drought,N supply did not enhance WUEi and biomass.

为研究幼龄茶园中间作药用植物对茶园小气候和土壤状况的影响,以单作茶园为对照,间作元胡、紫云英和菘蓝3种药用植物为处理,测定茶园温湿度、土壤养分含量和土壤酶活性,并进行相关性分析。结果表明:间作药用植物可有效降低茶园空气温度,增加茶园空气湿度,其中菘蓝间作效果最佳;间作茶园土壤元素含量在不同发育时期均有所提高,土壤有效氮在成熟采收期含量高,有效磷在不同发育期含量都高,有效钾在发育中期含量高,且均高于单作茶园,其中菘蓝间作茶园土壤元素含量最高;在发育中期,菘蓝间作茶园的土壤脲酶和蔗糖酶活性显著高于其他间作类型。相关性分析表明土壤脲酶、土壤养分含量呈正相关,土壤蔗糖酶、土壤有效钾呈正相关,过氧化氢酶与土壤pH和有效钾呈正相关。茶园间作药用植物能明显改善幼龄茶园土壤和小气候环境,保障幼龄茶树生长,尽快达到采摘标准,使茶农及时获得收益。

Adhesion molecules mediate cell-to-cell and cell-to-extracellular matrix interactions and transmit mechanical and chemical signals among them.Various mechanisms deregulate adhesion molecules in cancer,enabling tumor cells to proliferate without restraint,invade through tissue boundaries,escape from immune surveillance,and survive in the tumor microenvironment.Recent studies have revealed that adhesion molecules also drive angiogenesis,reshape metabolism,and are involved in stem cell self-renewal.In this review,we summarize the functions and mechanisms of adhesion molecules in cancer and the tumor microenvironment,as well as the therapeutic strategies targeting adhesion molecules.These studies have implications for furthering our understanding of adhesion molecules in cancer and providing a paradigm for exploring novel therapeutic approaches.

Nano silicon dioxide(nano-SiO) is becoming more and more widely applied in the fields of industry. The potential toxic effects of nano-SiO and its hazard to human health are drawing more attention.The mRNA expression of poly(ADP-ribose) polymerases-1(PARP-1),a pivotal repair gene,has been decreased by nano-SiO exposure.However,the effect of epigenetic modification on nano-SiO-induced low PARP1 expression has not been reported.In this study,HaCaT cells with or without DNA methyitransferase 1(DNMT1) knock down were incubated with nano-SiO and then further treated with DNMT inhibitor, 5-aza-2-deoxycytidine(DAC),which is a kind of key epigenetic modification reagents.Real-time Q-PCR and western blotting were used to examine the mRNA and protein expression of PARP-1.For promoter methylation status of PARP-1,methylation-specific PCR(MSP) and Bisulfite sequencing assay were performed. Results showed a dramatic decrease of PARP-1 expression on mRNA and protein level and a simultaneously obvious increase in the level of PARP-1 methylation in nano-SiO-treated cells compared to the control group.Further,the expression and promoter methylation of PARP-1 in HaCaT cells were restored following DNMT1 knock down,suggesting that the effects of PARP-1 promoter hypermethylation are mediated at least in part by DNMT1.Taken together,methylation of PARP-1 promoter might be involved in the regulation of nano-SiO-induced decrease of PARP-1 expression.
