Novel rapid tissue lysis method to evaluate cancer proteins: Correlation between elevated Bcl-X_L expression and colorectal cancer cell proliferation

AIM： We optimized a rapid and efficient tissue lysis method using the MagNA Lyser （Roche, Germany）. Using this novel method combined with immunoblot analysis, we investigated the correlation between abnormal Bcl-XL expression and clinicopathological characteristics in colorectal cancer. METHODS： Tissue samples from Sprague-Dawley rats were tested to determine optimal lysis conditions for use with MagNA Lyser. We next used the new method to extract tissue proteins from the tumor tissue of a colorectal cancer patient. The availability of extractable tissue proteins for proteomic study was demonstrated by two-dimensional （2D） gel electrophoresis and subsequent matrix-assisted laser desorption/ionization time-of-flight （MALDI-TOF） mass spectrometry. In addition, we prepared tissue lysates from paired tumor tissues and adjacent nontumor tissues of 50 colorectal carcinoma patients. Ensuing immunoblot analyses were performed to detect the level of Bcl-X, expression. RESULTS： The optimal sample sizes processed were found to be around 200 mg, with oscillation frequency of 6 500 r/rain for 80 s. Test of the first human tissue lysate confirmed that the MagNA Lyser method was adequate for protein extraction and subsequent identification by current proteomic protocols. The method was also applicable to immunoblot analysis. Thirty of 50 （60%） colorectal patients exhibited higher level of Bcl-XL expression in their tumor tissues. Raised level of Bcl-XL expression correlated with patients＇ gender and tumor cell proliferation index （P= 0.037 and P〈0.001, respectively）, but was independent of clinicopathological characteristics and overall survival. CONCLUSION： We report a novel tissue lysis method applicable to proteomic and immunoblot analyses, which can facilitate the discovery and detection of cancer protein alterations.

Intestinal lymphangiectasia, characterized by dilatation of intestinal lacteals, is rare. The major treatment for primary intestinal lymphangiectasia is dietary modification. Surgery to relieve symptoms and to clarify the etiology should be considered when medical treatment failed. This article reports a 49-year-old woman of solitary duodenal lymphangiectasia, who presented with epigastralgia and anemia. Her symptoms persisted with medical *** was finally performed to relieve the symptoms and to exclude the existence of underlying etiologies, with satisfactory effect. In conclusion, duodenal lymphangiectasia can present clinically as epigastralgia and chronic blood loss. Surgical resection may be resorted to relieve pain,control bleeding, and exclude underlying diseases in some patients.

Human Action Recognition Based on Dense Trajectories Analysis and Random Forest

This paper presents a human action recognition method. It analyzes the spatio-temporal grids along the dense trajectories and generates the histogram of oriented gradients （HOG） and histogram of optical flow （HOF） to describe the appearance and motion of the human object. Then, HOG combined with HOF is converted to bag-of-words （BoWs） by the vocabulary tree. Finally, it applies random forest to recognize the type of human action. In the experiments, KTH database and URADL database are tested for the performance evaluation. Comparing with the other approaches, we show that our approach has a better performance for the action videos with high inter-class and low inter-class variabilities.

旨在研究日粮中添加不同类型油脂对蛋鸡生产性能、蛋品质及血浆生化指标的影响。选取252只330日龄海兰褐蛋鸡,采用单因子设计,根据实际产蛋率近似原则随机分为大豆油组(对照组)、猪油组和棕榈油组3组,每组7个重复,每个重复12只,不同油脂添加量均为2%。试验周期为8周,记录采食量、蛋重、产蛋数,计算料蛋比,检测鸡蛋品质、蛋黄脂肪酸组成以及血浆生化指标。结果:在生产性能方面,与对照组相比,添加猪油、棕榈油均极显著降低平均蛋重(P0.05)。在蛋黄脂肪酸方面,与对照组相比,添加猪油、棕榈油都显著降低蛋黄花生烯酸(C20:1)、二十一烷酸(C21:0)含量(P<0.05),极显著降低多不饱和脂肪酸(PUFA)含量(P<0.01),极显著增加单不饱和脂肪酸(MUFA)含量(P<0.01);添加猪油极显著增加蛋黄油酸(C18:1n-9(t9)、C18:1n-9(c9))含量(P<0.01),显著增加花生四烯酸(C20:4n-6)含量(P<0.05);添加棕榈油极显著增加蛋黄饱和脂肪酸(SFA)(P<0.01),显著增加肉豆蔻酸(C14:0)含量(P<0.05),显著降低不饱和脂肪酸(USFA)含量(P<0.05)。在血浆生化指标方面,与对照组相比,添加猪油和棕榈油极显著降低血浆碱性磷酸酶浓度(P<0.01),显著降低血浆葡萄糖浓度(P<0.05);添加棕榈油极显著升高血浆谷丙转氨酶浓度(P<0.01)。综上,日粮添加猪油和棕榈油可显著降低平均蛋重,显著增加料蛋比,添加棕榈油可显著提高平均产蛋率;添加猪油和棕榈油可显著改善蛋黄脂肪酸组成并显著降低血浆碱性磷酸酶和葡萄糖浓度,添加棕榈油可显著升高血浆谷丙转氨酶浓度。

Risk assessment of gas control and spontaneous combustion of coal under gas drainage of an upper tunnel

The adjustment of the gas drainage rate has an immediate impact on air leakage in gob,thus resulting in the change of self-heating of *** regulating the gas drainage parameters,the risk of spontaneous combustion of coal should be *** risk assessment of gas control and spontaneous combustion of coal under gas drainage in a tunnel was investigated at different gas drainage *** distributions of the air volume along the working face,the gas management effects and the width of the oxidation zone were subjected to risk *** the simulation results showed,with increasing gas drainage rate,although the safety of gas dilution by ventilation was assured,the intensifying air leakage caused the oxidation zone to move into the deeper gob and led to an increase in the width of the oxidation zone.A risk assessment method was proposed to determine a suitable gas drainage rate for the upper *** correctness of the risk assessment and the validity of the numerical modelling were confirmed by the field measurements.

Lavatories are frequently used facilities,especially on long-haul *** a vacuum toilet in a lavatory can induce strong airflow,produce aerosols in the toilet bowl,and resuspend deposited particles from the ***,the exact particle transport routes and the fates of particle after toilet flushing are unclear so *** investigation used computational fluid dynamics(CFD)to model the transient airflow and pollutant transport after a toilet flushing process in a lavatory of a commercial *** time-varying pressure profile measured in a laboratory was assigned to the drainage valve as boundary *** aerosols generated inside the toilet bowl during flushing and the particles resuspended from the lavatory floor were used as particle *** tracking of airborne particles in the lavatory was *** addition,ammonia gas was used to examine odor *** multi-physics software program COMSOL 5.4 was employed for numerical solution after being *** results revealed that more than 70%of the generated particles in the toilet bowl are drained into sewage.A few particles may leak out of the toilet bowl and remain suspended in the air for more than five minutes when the toilet lid is open during *** the toilet with a closed lid can effectively reduce the particle leakage and the spread of odor gas,but it leads to greater deposition of particles on both the lid and *** is a slight inhalation exposure risk in the initial three minutes after flushing with a closed lid.

Modeling of laser adjustment for large diameter tubes using robotic kinematic theories

Laser adjustment is a new technique that employs laser forming processes to align components along specific directions. To develop a laser adjustment solution with a minimal number of bends, herein, a new modeling and calculation method that uses robotic kinematic theories is introduced. A numerical example and the corresponding laser adjustment solution are presented. The simulation results suggest that a straight tube with a flange connector can be adjusted to match the corresponding connector with three bends. Finally, the simulation results were verified using experiments. The maximum deviation was approximately 0.4 mm and the deviation of the flange connector was smaller, showing that a good adjustment quality could be achieved using the specific adjustment solution.

Quantum-chemical Study on the Structures and Properties of Uracil-BX_3 (X = F,Cl) Complexes

For the uracil-BX3 （X = F, Cl） systems, geometries and binding energies have been calculated by using the Lee-Young-Parr correlation functionals （B3LYP） method of density functional theory （DFT） and the second-order Moller-Plesset （MP2） method of ab initio at the 6- 311 ＋G^＊ or 6-311 ＋＋G^＊ basis set. Four isomers were found for each system, and then the single-point energy evaluations were performed using the larger basis sets of （6-311 ＋G（2df, p） and aug-cc-pVDZ with DFF method. In the most stable isomer of uracil-BF3 or uracil-BCl3, the boron atom of BX3 （X = F, Cl） connects to the carbonyl oxygen O7 of uracil with a stabilization energy of -46.56 or -31.10 kJ/mol at the B3LYP/6-31 1＋G^＊ level （BSSE corrected）. The analyses for combining interaction between BX3 and uracil with the atom-in-molecule theory （AIM） and natural bond orbital method （NBO） have been performed. The results indicate that all isomers were formed with σ-p type interactions between uracil and BX3, in which the carbonyl oxygen offers its lone pair electrons to the empty p orbital of boron atom and the concomitances of charge transfer from uracil to BX3 occur. Moreover, there exists one or two hydrogen bonds in most isomers of uracil-BX3 system and these hydrogen bonds contribute to the stability of the complex systems. Frequency analysis suggests that the stretching vibration of BX3 undergoes a red shift in complexes. Uracil-BF3 complex is more stable than uracil-BCl3 although the distance of B-O is shorter in the latter. Besides, the conversion mechanisms between different isomers of uracil-BF3 have been obtained.

Impact Performance Research of Re-Entrant Octagonal Negative Poisson’s Ratio Honeycomb with Gradient Design

Based on the traditional re-entrant honeycomb,a novel re-entrant octagon honeycomb(ROH)is *** deformation mode of the honeycomb under quasi-static compression is analyzed by numerical simulation,and the results are in good agreement with the experimental *** deformation modes,mechanical properties,and energy absorption characteristics of ROH along the impact and perpendicular directions gradient design are investigated under different *** results indicated that the deformation mode of ROH is affected by gradient design along the direction of impact and impact *** addition,gradient design along the direction of impact can increase the initial peak stress of ROH and accelerate its densification *** design perpendicular to the impact direction can enhance the energy absorption performance of ROH,especially for ROH,with wall thickness increasing from the inside *** to ROH with uniform wall thickness at the same relative density,ROH with a gradient design can increase the plateau stress by over *** the elevation of impact velocity,the plateau stress and specific energy absorption exhibit an upward trend,aligning with the dynamic performance pattern observed in conventional *** results can be used as a reference for the design and application of honeycomb and provide a new idea for developing more efficient and reliable energy-absorbing materials.
