Three-microRNA signature identified by bioinformatics analysis predicts prognosis of gastric cancer patients

AIM To identify multiple micro RNAs(mi RNAs) for predicting the prognosis of gastric cancer(GC) patients by bioinformatics *** The original microarray dataset GSE93415,which included 20 GC and 20 tumor adjacent normal gastric mucosal tissues,was downloaded from the Gene Expression Omnibus database and used for screening differentially expressed mi RNAs(DEMs).The cutoff criteria were P *** addition,we acquired the mi RNA expression profiles and clinical information of 361 GC patients from The Cancer Genome Atlas database to assess the prognostic role of the *** target genes of mi RNAs were predicted using Target Scan,mi RDB,mi RWalk,and DIANA,and then the common target genes were selected for functional enrichment *** A total of 110 DEMs including 19 up-regulated and 91 down-regulated mi RNAs were identified between 20 pairs of GC and tumor adjacent normal tissues,and the Kaplan-Meier survival analysis found that a threemi RNA signature(mi R-145-3 p,mi R-125 b-5 p,and mi R-99 a-5 p) had an obvious correlation with the survival of GC ***,univariate and multivariate Cox regression analyses indicated that the three-mi RNA signature could be a significant prognostic marker in GC *** common target genes of the three mi RNAs are added up to 108 and used for Gene Functional Enrichment *** Process and Molecular Function analyses showed that the target genes are involved in cell recognition,gene silencing and nucleic acid binding,transcription factor activity,and transmembrane receptor *** Component analysis revealed that the genes are portion of nucleus,chromatin silencing complex,and TORC1/2 *** Pathway analysis indicated that the genes participate in several cancer-related pathways,such as the focal adhesion,PI3 K,and m TOR signaling *** This study justified that a three-mi RNA signature could play a role in predicting the surviv

BACKGROUND The effect of low ligation(LL)vs high ligation(HL)of the inferior mesenteric artery(IMA)on functional outcomes during sigmoid colon and rectal cancer surgery,including urinary,sexual,and bowel function,is still *** To assess the effect of LL of the IMA on genitourinary function and defecation after colorectal cancer(CRC)*** EMBASE,PubMed,Web of Science,and the Cochrane Library were systematically searched to retrieve studies describing sigmoid colon and rectal cancer surgery in order to compare outcomes following LL and HL.A total of 14 articles,including 4750 patients,were analyzed using Review Manager 5.3 *** results are expressed as odds ratios(ORs)with 95%confidence intervals(CIs)and continuous outcomes are expressed as weighted mean differences(WMDs)with 95%*** LL resulted in a significantly lower incidence of nocturnal bowel movement(OR=0.73,95%CI:0.55 to 0.97,P=0.03)and anastomotic stenosis(OR=0.31,95%CI:0.16 to 0.62,P=0.0009)compared with *** risk of postoperative urinary dysfunction,however,did not differ significantly between the two *** meta-analysis also showed no significant differences between LL and HL in terms of anastomotic leakage,postoperative complications,total lymph nodes harvested,blood loss,operation time,tumor recurrence,mortality,5-year overall survival rate,or 5-year disease-free survival *** Since LL may result in better bowel function and a reduced rate of anastomotic stenosis following CRC surgeries,we suggest that LL be preferred over HL.

Extended antimicrobial prophylaxis after gastric cancer surgery: A systematic review and meta-analysis

AIM: To investigate the efficacy of extended antimicrobial prophylaxis (EAP) after gastrectomy by systematic review of literature and meta-analysis. METHODS: Electronic databases of PubMed, Embase, CINAHL, the Cochrane Database of Systematic Reviews, the Cochrane Controlled Trials Register and the China National Knowledge Infrastructure were searched systematically from January 1980 to October 2012. Strict literature retrieval and data extraction were carried out independently by two reviewers and meta-analyses were conducted using RevMan 5.0.2 with statistics tools risk ratios (RRs) and intention-to-treat analyses to evaluate the items of total complications, surgical site infection, incision infection, organ (or space) infection, remote site infection, anastomotic leakage (or dehiscence) and mortality. Fixed model or random model was selected accordingly and forest plot was conducted to display RR. Likewise, Cochrane Risk of Bias Tool was applied to evaluate the quality of randomized controlled trials (RCTs) included in this meta-analysis. RESULTS: A total of 1095 patients with gastric cancer were enrolled in four RCTs. No statistically significant differences were detected between EAP and intraoperative antimicrobial prophylaxis (IAP) in total complications (RR of 0.86, 95%CI: 0.63-1.16, P = 0.32), surgical site infection (RR of 1.97, 95%CI: 0.86-4.48, P = 0.11), incision infection (RR of 4.92, 95%CI: 0.58-41.66, P = 0.14), organ or space infection (RR of 1.55, 95%CI: 0.61-3.89, P = 0.36), anastomotic leakage or dehiscence (RR of 3.85, 95%CI: 0.64-23.17, P = 0.14) and mortality (RR of 1.14, 95%CI: 0.10-13.12; P = 0.92). Likewise, multiple-dose antimicrobial prophylaxis showed no difference compared with single-dose antimicrobial prophylaxis in surgical site infection (RR of 1.10, 95%CI: 0.62-1.93, P = 0.75). The effect of dopant valence on oxygen desorption and oxygen permeability of SrCo0.4Fe0.5M0.1O3-δ （M = Ni, Al and Zr） mixed-conducting oxides were investigated in detail by O2-TPD and oxygen permeation measurement. The SrCo0.4Fe0.5M0.1O3-δ for M = Fe, Ni, Al and Zr were denoted as SCF, SCFN, SCFA and SCFZ, respectively. O2-TPD analysis revealed that the amount of α oxygen desorption decreased with increasing the valance of doped metal elements （SCFN 〉 SCFA SCF 〉 SCFZ）. The oxygen permeation flux at the temperature ≈igher than 1148 K decreased in the order of SCFN 〉 SCF 〉 SCFZ 〉 SCFA. Single activation for oxygen permeation was observed for SCFZ oxide and the activation energies of SCF and SCFA change at around 1073 K, while the change temoerature of SCFN was about 1173 K.

Effects of starvation and hormones on DNA synthesis in silk gland cells of the silkworm, Bombyx moil

Silk gland cells of silkworm larvae undergo multiple cycles of endomitosis for the synthesis of silk proteins during the spinning phase. In this paper, we analyzed the endomitotic DNA synthesis of silk gland cells during larval development, and found that it was a periodic fluctuation, increasing during the vigorous feeding phase and being gradually inhibited in the next molting phase. That means it might be activated by a self- regulating process after molting. The expression levels of cyclin E, cdtl and pcna were consistent with these developmental changes. Moreover, we further examined whether these changes in endomitotic DNA synthesis resulted from feeding or hormonal stimulation. The results showed that DNA synthesis could be inhibited by starvation and re-activated by re-feeding, and therefore appears to be dependent on nutrition. DNA synthesis was suppressed by in vivo treatment with 20-hydroxyecdysone （20E）. However, there was no effect on DNA synthesis by in vitro 20E treatment or by either in vivo or in vitro juvenile hormone treatment. The levels of Akt and 4E-BP phosphorylation in the silk glands were also reduced by starvation and in vivo treatment with 20E. These results indicate that the activation of endomitotic DNA synthesis during the intermolt stages is related to feeding and DNA synthesis is inhibited indirectly by 20E.

Propagating subduction initiation of the Caroline Plate at the Mussau Trench

New seismic imaging from the Mussau Trench confirms that, under horizontal compression on an oceanic plate boundary, it is within the young and weak oceanic plate that a trench will develop, and it is the young plate, not so long after its birth, that starts to subduct toward the old plate. The Mesozoic Pacific Plate cannot easily bend, under the joint horizontal compression and vertical gravity pull, even with a pre-existing weak zone in place and with a large age offset across the plate boundary. The bending of the Caroline Plate near the northern end of the Mussau Trench started only after a long-time span of trench migration, and/or after a prolonged transition from initial compression to trench development. Highly contrasting trench deformation and sedimentary accretion between two parallel seismic sections support an evident migration of underthrusting or significant longitudinal heterogeneity along the trench. The swift transition from underthrusting to non-thrusting can be explained by a counter-clockwise rotation of the Caroline Plate. That miniature accretionary wedges develop even before the subduction initiates imply strong compression and uplifting before the underthrusting can kick-start. Rotation of the Caroline Plate have been halted as the Mussau Trench is seismically rather quiet and the Lyra Trough to the east shows relaxational extension.
