The present study aimed to explore the potential of artificial intelligence(AI)methodology based on magnetic resonance(MR)images to aid in the management of prostate cancer(PCa).To this end,we reviewed and summarized the studies comparing the diagnostic and predictive performance for PCa between AI and common clinical assessment methods based on MR images and/or clinical characteristics,thereby investigating whether AI methods are generally superior to common clinical assessment methods for the diagnosis and prediction fields of ***,we found that,in the included studies of the present study,AI methods were generally equal to or better than the clinical assessment methods for the risk assessment of PCa,such as risk stratification of prostate lesions and the prediction of therapeutic outcomes or PCa *** particular,for the diagnosis of clinically significant PCa,the AI methods achieved a higher summary receiver operator characteristic curve(SROC-AUC)than that of the clinical assessment methods(0.87 vs.0.82).For the prediction of adverse pathology,the AI methods also achieved a higher SROC-AUC than that of the clinical assessment methods(0.86 vs.0.75).Second,as revealed by the radiomics quality score(RQS),the studies included in the present study presented a relatively high total average RQS of 15.2(11.0–20.0).Further,the scores of the individual RQS elements implied that the AI models in these studies were constructed with relatively perfect and standard radiomics processes,but the exact generalizability and clinical practicality of the AI models should be further validated using higher levels of evidence,such as prospective studies and open-testing datasets.

Background:Prostate cancer is the second leading cause of cancer deaths in the western *** detection and effective therapy is essential for reducing mortality and increasing ***19 is a unique subunit and member of the Mediator super-family of proteins that plays a key role in the activation and repression of signal transduction or the regulation of transcription in ***,no functions of Med19 in prostate cancer cells have yet been reported. Methodology/Principal Findings:For the first time,we demonstrated that the expression of Med19 was specifically enhanced in prostate cancer patients' *** determine the effect of Med19 on human prostate cancer,lentivirus-mediated RNA interference(RNAi) was used to knock down the Med19 in two prostate cancer cell lines,PC-3 and *** was revealed that the in vitro cell proliferation and colonies formation ability,as well as in vivo tumor growth in nude mice xenografts significantly reduced after the expression level of Med19 was down-regulated,suggesting that Med19 can serve as a significant predictor of prostate ***,flow cytometry assay proved that Med19 affected different phases of the cell cycle in prostate cancer cell lines in vitro. Conclusions/Significance:Med19 may play a positive role in the progression of prostate cancer,providing a new approach to molecularly targeted RNAi gene therapy for prostate cancer.

Objective. To evaluate the efficiency and safety of a short-term low-dose tamsulosin treatment for ureteral calculi after single extracorporeal shock wave lithotripsy (ESWL). Methods. A 2-week-period randomized and controlled clinical trial was conducted in 108 patients with single ureteral calculi, who accepted ESWL as the initial interventional therapy. After ESWL, the patients in Group 1 (controls, n=55) received conservative therapy, such as hydration, antibiotics and acetaminophen on demand. But in Group 2 (n=53), patients received tamsulosin (0.2mg daily) in addition to conservative therapy. All patients were questioned about the number and intensity of post-discharge pain and evaluated by plain abdominal radiography at the end of the follow-up. Results. The rate of calculi clearance in the Group 2 (84.91%, 45/53) was higher than that Group 1 (65.45%, 36/55) (P=0.020), evidently for the larger (≥10mm) calculi (83.33%, 20/24 versus 48.48%, 16/33) (P=0.007) and the proximal ones (86.67%, 26/30 versus 62.16%, 23/37) (P=0.024). Moreover, Group 1 reported higher mean visual analog scores of colic intensity (3.11±2.60) than Group 2 did (1.98±2.08) (P=0.015). And 36.36% (20/55) patients in Group 1 used acetaminophen to reduce post-discharge pain, higher than that of Group 2 (18.87%, 10/53) (P=0.042). No side effects of tamsulosin were observed in either group. Conclusion. The addition of low-dose tamsulosin after single ESWL can enhance the clearance of ureteral calculi successfully in a 2-week term, evidently for those larger and proximal ones, and reduce the intensity of post-discharge pain, with low risk of side effects.

Prostate cancer(PCa)is a common malignant tumor with high morbidity and mortality *** prostate cancer stem cell(PCSC)model provides novel insights into the pathogenesis of PCa and its therapeutic ***,the roles and molecular mechanisms of specific genes in mediating fate decisions of PCSCs and carcinogenesis of PCa remain to be *** this study,we have explored the expression,function,and mechanism of AZGP1P2,a pseudogene of AZGP1,in regulating the stemness and apoptosis of PCSCs and treatment resistance of docetaxel in castration-resistant prostate cancer(CRPC).We revealed that AZGP1P2 was downregulated in CRPC cell lines and PCSCs,while it was positively associated with progression-free *** of the AZGP1P2 enhanced the sensitivity of docetaxel treatment in CRPCs via inhibiting their *** pull-down associated with mass spectrometry analysis,co-immunoprecipitation assay,and RNA immunoprecipitation assay demonstrated that AZGP1P2 could bind to UBA1 and RBM15 as a“writer”of methyltransferase to form a ***1,an E1 ubiquitin-activating enzyme,contributed to RBM15 protein degradation via ubiquitination *** RNA immunoprecipitation assay displayed that RBM15 controlled the mRNA decay of TPM1 in m6A ***,a xenograft mouse model and patient-derived organoids showed that the therapeutic effect of docetaxel in CRPC was increased by AZGP1P2 in ***,these results imply that AZGP1P2 mediates the stemness and apoptosis of PCSCs and promotes docetaxel therapeutic effect by suppressing tumor growth and metastasis via UBA1/RBM15-mediated TPM1 mRNA decay in CRPC.
