Molecular Cloning and Preliminary Analysis of a Fragile Site Associated Gene

Objective To analyze the molecular coining of a fragile site-associated gene. Methods Genomic Chinese hamster ovary （CHO） DNA library was constructed using high molecular weight CHO DNA partially digested with MboI restriction enzyme from cultured CHO cells. Screening of genomic DNA library followed the established procedures. Genomic CHO in the positive clones was sequenced. Appropriate primers were designed for the reverse transcriptase-polymerase chain reactions （RT-PCR）. The RT-PCR products were cloned into a pCRII TOPO vector and confirmed by DNA sequencing. Antibodies were prepared using synthetic peptides as antigens by immunizing the rabbits. Immunohistochemical analyses were performed to evaluate the expression of the novel gene in different tissues. Results To investigate the molecular mechanism underlying the initial events of mdrla amplification, we cloned lq31 fragile site DNA. Strikingly, we found that this fragile site contained a novel gene which was designated as a fragile site-associated （FSA） gene. FSA encoded an unusually large mRNA of - 16 kb. Full-length human FSA eDNA was cloned. FSA mRNA was expressed in many cultured cells and tissue types. Immunohistochemical analyses also revealed an expression pattern of the encoded proteins in postmitotic, well-differentiated epithelial compartments of many organs, including colon, mammary glands, ovary, prostate, and bladder. Conclusion FSA plays an important role in regulating mammalian epithelial cell growth and differentiation.

No-go guide for the Hubble tension:Late-time or local-scale new physics

The standard model of modern cosmology might be cracked by the recent persistent hot debate on the Hubble-constant(H_(0))tension,which manifests itself as the sound-horizon(r_(s))tension or absolute-magnitude(M_(B))tension if deeming the origin of the Hubble tension from modifying the early or late Universe,*** this study,we achieve a fully model-independent constraint(fitting a model-independent global parameterization to a model-independent inverse distant ladder with a modelindependent high-redshift calibration)on late-time models with strong evidence against homogeneous new physics over theΛ-cold-dark-matter(ΛCDM)*** using this model-independent constraint to calibrate sufficiently local supernovae with corresponding late-time models extrapolated below the homogeneity scale,we find surprisingly that,although both H_(0)tension and MB tension are absent in our local Universe,a combination of H_(0)and M_(B)as the intercept a B of the magnitude-redshift relation exhibits 3~7σtension even for theΛCDM *** aBtension seems to call for local-scale inhomogeneous new physics disguised as local observational systematics.

On-line determination of selenium compounds in tea infusion by extractive electrospray ionization mass spectrometry combined with a heating reaction device

Selenium is one of the important trace elements in the human *** deficiency will directly affect human *** people's attention to health,the content of selenium in food has gradually attracted ***,detecting selenium compounds in complex samples remains a *** this work,we built an online heating-reaction *** device combines the electrospray extraction ionization mass spectrometry(EESI-MS)with the heating reaction device,which can simultaneously detect various selenium compounds in complex liquid *** acidic conditions,the sample was heated and catalyzed by a heating reaction device,so that the SeO~(2-)_(3)and O-phenylenediamine(OPD)could generate 1,3-dihydro-2,1,*** on the above reactions,we can detect organic selenium,inorganic selenium and other compounds in liquid samples by organic mass *** this experiment,we determined the content of three forms of selenium:selenomethionine(SeMet),l-selenocystine(SeCys(2)),and sodium *** calibration curves for SeMet,SeCys(2),and sodium selenite showed strong linearity within a range of 0.50-50.00μg/*** limits of detection(LOD)for the three compounds were 0.22,0.27,and 0.41μg/L,*** limits of quantification(LOQ)were 0.68,0.81,and 1.23μg/L,*** recoveries at three levels ranged from 98.8%to 106.1%.In addition,this method can simultaneously detect three selenium compounds and three other specific chemical components in tea infusion samples,providing a rapid and efficient method for identifying tea quality.

Mechanism of extracellular electron transport and reactive oxygen mediated Sb(Ⅲ)oxidation by Klebsiella aerogenes HC10

Microbial oxidation and the mechanism of Sb(Ⅲ)are key governing elements in biogeochemical cycling.A novel Sb oxidizing bacterium,Klebsiella aerogenes HC10,was attracted early and revealed that extracellular metabolites were the main fractions driving Sb ***,linkages between the extracellular metabolite driven Sb oxidation process and mechanism remain ***,model phenolic and quinone compounds,i.e.,anthraquinone-2,6-disulfonate(AQDS)and hydroquinone(HYD),representing extracellular oxidants secreted by *** HC10,were chosen to further study the Sb(Ⅲ)oxidation mechanism.N_(2)purging and free radical quenching showed that oxygen-induced oxidation accounted for 36.78%of Sb(Ⅲ)in the metabolite reaction system,while hydroxyl free radicals(·OH)accounted for 15.52%.·OH and H_(2)O_(2)are the main driving factors for Sb *** quenching,methanol purification and electron paramagnetic resonance(EPR)analysis revealed that·OH,superoxide radical(O_(2)^(•-))and semiquinone(SQ-•)were reactive intermediates of the phenolic induced oxidation ***-induced ROS are one of the main oxidants in *** voltammetry(CV)showed that electron transfer of quinone also mediated Sb(Ⅲ)*** of Sb(V)was scavenged by the formation of the secondary Sb(V)-bearing mineral mopungite[NaSb(OH)6]in the incubation *** study demonstrates the microbial role of oxidation detoxification and mineralization of Sb and provides scientific references for the biochemical remediation of Sb-contaminated soil.

Accurate measurements of the bond dissociation energies of ¹⁴N₂, ¹⁴N¹⁵N and ¹⁵N₂

Accurate determinations of bond dissociation energies (BDEs) and the corresponding enthalpies of formation derived from them are of fundamental importance, while on the other hand also very challenging in chemistry. In this study, we demonstrate a direct experimental scheme for measuring the BDEs of 14N2, 14N15N and 15N2 to an accuracy of a few wavenumbers. The high-resolution quantum-state-selected velocity-mapped ion images for several rotational levels with accurately known spectroscopic term energies slightly above the dissociation limit N(2D5/2, 3/2) + N(2D5/2, 3/2) were obtained for each of the dinitrogen isotopologue molecules. A linear fit between squares of the radii of the quantum-state-selected images and the spectroscopic term energies gives out an accurate measurement of the threshold D0 for dissociating into the channel N(2D5/2, 3/2) + N(2D5/2, 3/2). The BDEs of 4N2, 14N15N and 15N2 into the lowest channel N(4S) + N(4S) are independently determined to be 78688 ± 3, 78705 ± 4 and 78728 ± 4 cm−1, respectively. These independently determined BDEs are found to give a consistent value of De after considering the zero-point energies for each of the isotopologues, implying the reliability of the present measurement. Comparisons with several previous measurements and theoretical calculations are discussed. The experimental scheme demonstrated in this study should be generally applicable for precisely determining the BDEs of many other diatomic molecules. © 2022

Construction of an artificial zinc alloy layer toward stable zinc-metal anode

Aqueous zinc-ion batteries(AZIBs)present a promising option for next-generation batteries given their high safety,eco-friendliness,and resource ***,the practical application of zinc anodes is hindered by inevitable parasitic reactions and dendrite ***,zinc alloy layers(i.e.,ZnCo and ZnFe alloys)were rationally constructed on the zinc surface by chemical displacement *** alloying process exposes more(002)planes of the ZnCo anode to guide the preferential and dendrite-free zinc ***,the ZnCo alloy layer not only effectively inhibits water-induced side reactions but also accelerates electrode kinetics,enabling highly reversible zinc plating/*** a result,the ZnCo anode achieves a Coulombic efficiency of 99.2%over 1300 cycles,and the ZnCo symmetric cell exhibits a long cycle life of over 2000 h at 4.4 mA cm^(-2).Importantly,the ZnCo//NH_(4)V_(4)O_(10)full cell retains a high discharge capacity of 218.4 mAh g^(-1)after 800 ***,the ZnFe-based symmetric cell also displays excellent cycling stability over 2500 h at 1.77 mA cm^(-2).This strategy provides a facile anode modification approach toward high-performance AZIBs.

Evaluation of the malignant potential of gliomas using diffusion-weighted and gadolinium-enhanced magnetic resonance imaging

Controlling segregation and hardenability of gear steel based on as-cast structure

Hardenability significantly impacts the distortion of gear during heat treatment,correlated to the uniformity of solute distribution in steel *** experimental analysis was conducted on the macrostructure,solute distribution,dendrite structure,and rod hardenability of 20CrMnTiH gear steel in continuously cast blooms and hot roller *** evaluation approach by the standards for the hardenability of gear steel rods and the corresponding blooms was analyzed,and the inheritance mechanism from solidification segregation to hardenability fluctuation of gear steel was *** results indicate that semi-macroscopic spot segregation located in the equiaxed zone exhibits larger size,higher solute enrichment,and worse solute homogeneity,leading to significant solute fluctuations in the blooms and hardenability fluctuation in the *** increasing the liquid steel superheat from 35 to 40℃,reducing the mold electromagnetic stirring from 300 to 100 A,and implementing the soft reduction(SR)of 7 mm at the solidification end,the equiaxed ratio of the strand decreased from 26.42%to 6.69%.Consequently,the solute fluctuation range and standard deviation decrease significantly in the transverse section,while the maximum segregation ratio,average fluctuation range,and average standard deviation of solutes C,Cr,and Mn in the spot segregation decrease at the same *** the meanwhile,the equiaxed ratio of the rod decreased from 24.89%to 4.09%,and the structure of the hardenability detection zone was transformed from equiaxed crystals to columnar ***,the solute fluctuation range and standard deviation in the transverse section decreased,while the homogeneity in spot segregation was also *** hardness difference of A and B surfaces at J9 and J15 positions was smaller than 2 HRC,meeting the qualification standard for hardenability.

The effect of fine grits and fine debris concentrations on the MLVSS/MLSS ratio of an activated sludge system

This study embarks on an explorative investigation into the effects of typical concentrations and varying particle sizes of fine grits(FG,the involatile portion of suspended solids)and fine debris(FD,the volatile yet unbiodegradable fraction of suspended solids)within the influent on themixed liquor volatile suspended solids(MLVSS)/mixed liquor suspended solids(MLSS)ratio of an activated sludge *** meticulous experimentation,it was discerned that the addition of FG or FD,the particle size of FG,and the concentration of FD bore no substantial impact on the pollutant removal efficiency(denoted by the removal rate of COD and ammonia nitrogen)under constant operational ***,a notable decrease in the MLVSS/MLSS ratio was observed with a typical FG concentration of 20 mg/L,with smaller FG particle sizes exacerbating this ***,variations in FD concentrations influenced both MLSS andMLVSS/MLSS ratios;a higher FD concentration led to an increased MLSS and a reduced MLVSS/MLSS ratio,indicating FD accumulation in the system.A predictive model for MLVSS/MLSS was constructed based on quality balance calculations,offering a tool for foreseeing the MLVSS/MLSS ratio under stable long-term influent conditions of FG and *** model,validated using data from the BXH wastewater treatment plant(WWTP),showcased remarkable accuracy.
