Colorectal cancer(CRC),a widespread malignancy,is closely associated with tumor microenvironmental hydrogen peroxide(H_(2)O_(2))*** clinical trials targeting H_(2)O_(2)for cancer treatment have revealed its paradoxical role as a promoter of cancer *** the dynamics of cancer cell H_(2)O_(2)eustress at the single–cell level is *** this study,non–contact hopping probe mode scanning ion conductance microscopy(HPICM)with high-sensitive Pt–functionalized nanoelectrodes was employed to measure dynamic extracellular to intracellular H_(2)O_(2)gradients in individual colorectal cancer Caco–*** explored the relationship between cellular mechanical properties and H_(2)O_(2)*** to 0.1 or 1 mmol/L H_(2)O_(2)eustress increased the extracellular to intracellular H_(2)O_(2)gradient from 0.3 to 1.91 or 3.04,***,cellular F–actin–dependent stiffness increased at 0.1 mmol/L but decreased at 1 mmol/L H_(2)O_(2)*** H_(2)O_(2)–induced stiffness modulated AKT activation positively and glutathione peroxidase 2(GPX2)expression *** findings unveil the failure of some H_(2)O_(2)-targeted therapies due to their ineffectiveness in generating H_(2)O_(2),which instead acts eustress to promote cancer cell *** research also reveals the complex interplay between physical properties and biochemical signaling in cancer cells'antioxidant defense,illuminating the exploitation of H_(2)O_(2)eustress for survival at the single–cell *** GPX and/or catalase(CAT)enhances the cytotoxic activity of H_(2)O_(2)eustress against CRC cells,which holds significant promise for developing innovative therapies targeting cancer and other H_(2)O_(2)-related inflammatory diseases.

Carboxylic acid-functionalized single walled carbon nanotubes(SWNTs)prepared via the reaction of an amino acid,NH2(CH2)nCO2H where n=1(glycine,GLY),5(6-aminohexanoic acid,AHA),10(11-aminoundecanoic acid,AUDA),with fl uorinated single walled carbon nanotubes(F-SWNTs)have been characterized by MAS 13C NMR *** ease of observing the aliphatic CH2 groups and the resolution of the signal are dependent on the length of the amino acid’s aliphatic *** have proposed that where substituent chains are short(making NMR data collection difficult)chemical modification to extend the chain length should alleviate analysis *** this regard,we have investigated the esterifi cation of the carboxylic acid *** amino acid-functionalized SWNTs were esterified with an appropriate alcohol to ensure parity of the overall substituent length,i.e.,GLY-SWNT(C1)+1-dodecanol(C_(12))=DOD-GLY-SWNT(1),AHA-SWNT(C_(5))+1-octanol(C_(8))=OCT-AHA-SWNT(2),and AUDA-SWNT(C_(10))+1-propanol(C_(3))=PRO-AUDA-SWNT(3).The 13C NMR shift for the sp3 nitrogen-substituted carbon atoms of the SWNT sidewall is observed atδ≈75 *** the length of SWNT sidewall functional groups enhances the ability to observe the sidewall sp^(3)*** methylene carbon signal intensity is less attenuated in the dipolar dephasing spectrum of the ester-functionalized SWNTs than their associated amino acid derivatives,suggesting more motional freedom of the side chain in the solid *** confi rmation of the dipolar dephasing spectral effects was assisted by the characterization of the ester of AUDA-SWNT with 1,3-propanediol:PPD-AUDA-SWNT(4).

Prospects for sociogenomics in avian cooperative breeding and parental care

For the last 40 years,the study of cooperative breeding(CB)in birds has proceeded primarily in the context of discovering the ecological,geographical,and behavioral drivers of *** advent of molecular tools in the early 1990s assisted in clarifying the relatedness of helpers to those helped,in some cases,confirming predictions of kin selection *** for genome-wide analysis of sequence variation,gene expression,and epigenetics promise to add new dimensions to our understanding of avian CB,primarily in the area of molecular and developmental correlates of delayed breeding and dispersal,as well as the ontogeny of achieving parental status in ***,we outline key ways in which modern-omics approaches,in particular genome sequencing,transcriptomics,and epigenetic profiling such as ATAC-seq,can be used to add a new level of ana-lysis of avian *** on recent and ongoing studies of avian social behavior and sociogenom-ics,we review how high-throughput sequencing of a focal species or clade can provide a robust foundation for downstream,context-dependent destructive and non-destructive sampling of spe-cific tissues or physiological states in the field for analysis of gene expression and epigenetics.-Omics approaches have the potential to inform not only studies of the diversification of CB over evolutionary time,but real-time analyses of behavioral interactions in the field or *** of birds represents a new branch in the network of methods used to study CB,and can help clarify ways in which the different levels of analysis of CB ultimately interact in novel and unexpected ways.

Hypertriglyceridemia-induced pancreatitis: A case-based review

Hypertriglyceridemia is an established cause of pancreatitis. In a case-based approach, we present a review of hypertriglyceridemJa and how it can cause pancreatitis. We outline how to investigate and manage such patients. A 35 year old man presented to the emergency department with abdominal pain and biochemical evidence of acute pancreatitis. There was no history of alcohol consumption and biliary imaging was normal. The only relevant past medical history was that of mild hyperlipidemia, treated with diet alone. Physical exam revealed epigastric tenderness, right lateral rectus palsy, lipemia retinalis, bitemporal hemianopsia and a delay in the relaxation phase of his ankle reflexes. Subsequent laboratory investigation revealed marked hypertriglyceridemia and panhypopituarism. An enhanced CT scan of the head revealed a large suprasellar mass impinging on the optic chiasm and hypothalamus. The patient was treated supportively; thyroid replacement and lipid lowering agents were started. He underwent a successful resection of a craniopharyngioma. Postoperatively, the patient did well on hormone replacement therapy. He has had no further attacks of pancreatitis. This case highlights many of the factors involved in the regulation of triglyceride metabolism. We review the common causes of hypertriglyceridemia and the proposed mechanisms resulting in pancreatitis. The incidence and management of hypertriglyceridemiainduced pancreatitis are also discussed.

The Utility of Voided Urine Samples as a Proxy for the Vaginal Microbiome and for the Prediction of Bacterial Vaginosis

Recent work has shown that the vaginal microbiome exerts a strong impact on women's gynecological ***,collection of vaginal specimens is invasive and requires previous clinical training or the involvement of a trained *** contrast,urine sample collection is routine and noninvasive and does not require involvement of a *** sought to compare the vaginal and urogenital microbiomes to assess the utility of voided urine samples as a proxy for the vaginal *** urogenital and vaginal samples were collected from pregnant women and characterized by 16S rRNA taxonomic *** examined diversities and compositions of paired urogenital and vaginal microbiomes using five discrete strategies to explore the similarity between the vaginal and urogenital microbiomes.A strategy comparing the paired urogenital and vaginal microbiomes in which taxa were assigned using the STIRRUPS database and urine-specific taxa were removed showed no significant difference in diversity and composition between the paired urogenital and vaginal ***,the relative abundances of common vaginal taxa were linearly correlated with those in the paired *** similarities suggest that voided urine samples could represent a noninvasive protocol for accurate profiling of the vaginalmicrobiome with likely clinical ***,a machine learning model was established in which the voided urine microbiome was compared favorably to the vaginal microbiome in predicting bacterial vaginosis.

目的：运用外固定架治疗肢体不等长或畸形，矫正具有潜在的并发症。现报道一组因治疗过程中或结束后该延长肢体出现并发症而需再次运用外固定架的病例。方法：1991年2月-1999年7月，共有261例（297肢体）用Or-thofix或Ilizarov外固定进行肢体延长或畸形矫正。其中儿童228肢体，成人69肢体。结果：儿童再次运用外固定架治疗的有22例次（9.65％），成人有4例次（5.8％0.总的再次运用外固定架率达8.75％。再次运用外固定架的原因为延长区再生骨的骨折和成角畸形。有4例患者最后接受截肢术。结论：对因先天性疾患致肢体严重残疾的患儿，再次运用外固定架治疗仍具有临床价值。

Diet-Induced Alteration of the Murine Intestinal Microbiome Following Antibiotic Ablation

Mouse models of antibiotic-induced ablation of the intestinal microbiome have been used to study the microbiome in health and disease. The fecal microbiomes of mice treated with broad-spectrum antibiotics while being fed different laboratory chows were analyzed by Gram stain, quantitative flow cytometry, bacterial cell culture, next generation sequencing of the V3 regions of the 16S ribosomal RNA (rRNA) gene, microscopy, and sequence analysis of the tuf gene. Noncultivatable gram-positive cocci and cultivatable yeast were the microorganisms most readily detected in feces of antibiotic-treated mice fed a defined diet that utilizes casein as a protein source, maltodextrin 10 and sucrose as sources of carbohydrates, and lard as the major source of fat. High-throughput sequencing of the variable regions of the 16S rRNA gene and tuf gene sequencing identified the major bacterial phylotype as Lactococcus. The mouse chow was heavily laden with noncultivatable Lactococcus, which dominated the intestinal flora after consumption. The microbiome of antibiotic-treated mice fed a grain-based diet (mainly wheat, corn and alfalfa) consisted predominantly of a member of the Enterobacteriaceae identified as Escherichia coli, and yeast was not detected by culture or Gram stain. Appearance of intestinal yeast by culture and Gram stain was dependent on the specific chow, although yeast was not detected by culture or Gram stain in the chow. We conclude that bacteria found in food sources can influence qualitative and quantitative assessments of the fecal microbiome, at least in the context of antibiotic therapy, and potentially confound molecular studies that assess the effects of diet on the intestinal ecology. Not surprisingly, different food sources can influence the microbiome, particularly in the context of antibiotic-mediated ablation of the intestinal microbiome. Whether and how the food-derived dead bacteria alter intestinal physiology needs to be determined.
