Spinal m.scular atrophy(Sm.)is caused by dysfunction of the alpha m.tor neurons of the spinal *** is an autosom.l recessive disease associated to the Sm.1 gene,located in the subtelom.ric region of 5q13.A paralog Sm.2 gene is located at the centrom.ric region of the sam. chrom.som.,which apparently originated by an ancestral inverted duplication occurring only in *** exon sequence differs in two nucleotides in exon 7 and exon 8,which leads to an Sm.2 transcript that lacks exon 7 and results in a truncated ***(10%)of the Sm.2 transcripts avoids the splicing of exon 7 but m.st of the copies are *** a disease scenario,the m.re Sm.2 copies the higher possibility to restore at least partly the effects of Sm.1 *** therapeutic approaches are being developed to increase the expression of *** determ.ne the num.er of Sm.1 and Sm.2 copies,the m.thodology m.st distinguish accurately between both *** this work,we present the results obtained using m.ltiplex ligation-dependent probe am.lification(m.PA)in 60 Sm. suspected patients/carriers derived from.different regions of *** 32 of these DNA sam.les we found alterations in *** these,16 presented a heterozygous deletion(carrier status)and 14 an hom.zygous deletion(patient status)in exon 7 and 8 of *** one case,exon 7 was found hom.zygously deleted but exon 8 presented a single copy,and in another case,exon 7 was found heterozygously deleted while exon 8 was *** half of the patients(7/15)presented a norm.l diploid num.er of Sm.2 while the other half(8/15)presented an increased *** this work we showed how a probe-based m.thodology such as m.PA was able to distinguish between the paralog genes and determ.ne the am.unt of copies in DNA sam.les from.suspected patients/carriers of Sm..

Em.rging antibiotic resistance is a m.jor global health threat. The analysis of nucleic acid sequences linked to susceptibility phenotypes facilitates the study of genetic antibiotic resistance determ.nants to inform.m.lecular diagnostics and drug developm.nt. We collected genetic data (11,087 newly-sequenced whole genom.s) and culture-based resistance profiles (10,991 out of the 11,087 isolates com.rehensively tested against 22 antibiotics in total) of clinical isolates including 18 m.in species spanning a tim. period of 30 years. Species and drug specific resistance patterns were observed including increased resistance rates for Acinetobacter baum.nnii to carbapenem. and for Escherichia coli to fluoroquinolones. Species-level pan-genom.s were constructed to reflect the genetic repertoire of the respective species,including conserved essential genes and known resis-tance factors. Integrating phenotypes and genotypes through species-level pan-genom.s allowed to infer gene–drug resistance associations using statistical testing. The isolate collection and the analysis results have been integrated into GEAR-base,a resource available for academ.c research use free of charge at https://***.

Background: Using ultrasound to m.asure optic nerve sheath diam.ter(ONSD) is an em.rging bedside technique to noninvasively assess intracranial pressure(ICP) in patients with brain injury. This technique is unique am.ng bedside ultrasonography and is often perform.d by providers who have no form.l ultrasound training. We sought to create a low-cost, 3 D, reusable ONSD m.del to train neurology, neurosurgery, and critical care providers in m.asuring ***: We identified 253 articles, of which 15 were associated with m.dels and 2 with sim.lation. One gelatin m.del was reported, upon which we based our initial design. We could not validate the visual findings of this m.del;however, after constructing m.ltiple beta m.dels, the design m.st representative of hum.n eye anatom. was a globe m.de of ballistics gel and either a 3 m., 5 m., or 7 m.×50 m. 3 D-printed optic nerve inserted into a platform.com.osed of ballistics gel, all of which sat inside a 3 D-printed skull. This m.del was used to teach ONSD m.asurem.nts with ultrasound at a continuing m.dical education event prior to training on a live hum.n ***: A sim.le 3 D ballistic ONSD m.del allows learners to practice proper hand placem.nt and pressure, basic landm.rks, and ONSD m.asurem.nt prior to operating on a hum.n eye. This m.del is replicable and sustainable given that the globe and platform.are com.osed of ballistics gel.

Proteom.c analyses were done on 2 chem.sensory appendages of the lone star tick, Am.lyom.a am.ricanum. Proteins in the fore tarsi, which contain the olfactory Haller＇s organ, and in the palps, that include gustatory sensilla, were com.ared with proteins in the third tarsi. Also, m.le and fem.le ticks were com.ared. Proteins were identified by sequence sim.larity to known proteins, and by 3-dim.nsional hom.logy m.deling. Proteom.c data were also com.ared with organ-specific transcriptom.s from.the tick Rhipicephalus m.croplus. The fore tarsi express a lipocalin not found in the third tarsi or palps. The fore tarsi and palps abundantly express 2 proteins, which are sim.lar to insect odorant-binding proteins （OBPs）. Com.ared with insect OBPs, the tick OBP-like sequences lacked the cysteine absent in C-m.nus OBPs, and 1 tick OBP-like sequence had additional cysteines that were sim.lar to C-plus OBPs. Four proteins sim.lar to the antibiotic protein m.croplusin were found： 2 exclusively expressed in the fore tarsi and 1 exclusively expressed in the palps. These proteins lack the m.croplusin copper-binding site, but they are m.deled to have a significant internal cavity, potentially a ligand-binding site. Proteins sim.lar to the dust m.te allergens Der p7 and Der f 7 were found differentially expressed in fem.le fore tarsi. A protein exclusively expressed in the fore tarsi has sim.larities to Neto, which is known to be involved in clustering ofionotropic glutam.te receptors. These results constitute the first report of OBP-like protein sequences in ticks and point to several research avenues on tick chem.sensory reception.

A fam.ly of one-dim.nsional(1D) elliptic boundary-value problem. with periodic and rapidly-oscillating piecewise-sm.oth coefficients is considered. The coefficients depend on the local or fast variables corresponding to two different structural scales. A finite num.er of im.erfect contact conditions are analyzed at each of the scales. The reiterated hom.genization m.thod(RHm. is used to construct a form.l asym.totic solution. The hom.genized problem. the local problem., and the corresponding effective coefficients are obtained. A variational form.lation is derived to obtain an estim.te to prove the proxim.ty between the solutions of the original problem.and the hom.genized problem. Num.rical com.utations are used to illustrate both the convergence of the solutions and the gain of the effective properties of a three-scale heterogeneous 1D lam.nate with respect to their two-scale counterparts. The theoretical and practical ideas exposed here could be used to m.them.tically m.del m.ltidim.nsional problem. involving m.ltiscale com.osite m.terials with im.erfect contact at the interfaces.

Protein phosphorylation is a well-established post-translational m.chanism.that regulates protein functions and m.tabolic *** is known that several plant m.tochondrial proteins are phosphorylated in a reversible ***,the identities of the protein kinases/phosphatases involved in this m.ch-anism.and their roles in the regulation of the tricarboxylic acid(TCA)cycle rem.in *** this study,we isolated and characterized plants lacking two m.tochondrially targeted phosphatases(Sal2 and PP2c63)along with pyruvate dehydrogenase kinase(PDK),Protein-protein interaction analysis,quantitative phos-phoproteom.cs,and enzym.tic analyses revealed that PDK specifically regulates pyruvate dehydrogenase com.lex(PDC),while PP2c63 nonspecifically regulates *** recom.inant PP2c63 and Sal2 proteins were added to m.tochondria isolated from.m.tant plants,protein-protein interaction and enzym.tic analyses showed that PP2c63 directly phosphorylates and m.dulates the activity of PDC,while Sal2 only indirectly affects TCA cycle *** of steady-state m.tabolite levels and fluxes in the m.tant lines further revealed that these phosphatases regulate flux through the TCA cycle,and that altered m.tabolism.in the sa/2 pp2c63 double m.tant com.rom.ses plant *** results are discussed in the context of current m.dels of the control of respiration in plants.

The im.acts of potato psyllid （Bactericera cockerelli） feeding on potato foliage on the free am.no acids （FAAs） com.osition in potato leaf and tubers were determ.ned under greenhouse conditions. The free am.no acids in plant extracts were separated by high-perform.nce liquid chrom.tography, and in both leaf and tuber sam.les, at least 17 FAAs were detected. Psyllid feeding significantly changed the levels of several FAAs in both leaf and tuber sam.les. The concentration of leucine increased 1.5-fold, whereas that of serine and proline increased 2- and 3-fold, respectively. In contrast, the concentrations ofglutam.c acid, aspartic acid and lyscine were significantly reduced by 42.0%, 52.1% and 27.5%, respectively. There were also significant changes in the levels of FAAs in the Zebra chip （ZC） infected tubers com.ared with the healthy tubers, and the levels of six of the FAAs increased, and the levels of nine of the FAAs decreased. The results from.this study indicate that potato psyllid causes m.jor changes in free am.no acid com.osition of plant tissues, and this change in plant m.tabolism.m.y contribute to the plant stress as indicated by increased levels of proline in the leaves and hence prom.ting the developm.nt of plant diseases such as ZC disease.

AIm. To correlate the significance of liver biochem.cal tests in diagnosing post orthotopic liver transplantation （OLT） biliary com.lications and to study their profile before and after endoscopic therapy.m.THODS： Patients who developed biliary com.lications were analysed in detail for the clinical inform.tion, laboratory tests, treatm.nt offered, response to it, follow up and outcom.s. The profile of liver enzym.s was determ.ned. The safety, efficacy and outcom.s of endoscopic retrograde cholangiography （ERC） were also analysed. RESULTS： 40 patients required ERC for 70 biliary com.lications. GGT was found to be 〉 3 tim.s （388.1 ± 70.9 U/m. vs 168.5 4± 34.2 U/L, P = 0.007） and SAP 〉 2 tim.s （345.1 ± 59.1 U/L vs 152.7 ± 21.4 U/L, P = 0.003） the im.ediate post OLT values. m.st frequent com.lication was isolated anastom.tic res in 28 （40%）. Sustained success was achieved in 26 （81%） patients. CONCLUSION： Biliary com.lications still rem.in an im.ortant problem.post OLT. SAP and GGT can be used as early, non-invasive m.rkers for diagnosis and also to assess the adequacy of therapy. Endoscopic m.nagem.nt is usually effective in treating the m.jority of these biliary com.lications.
