The sexually dimorphic expression of glutamate transporters and their implication in pain after spinal cord injury

Long-term survivor of metastatic squamous-cell head and neck carcinoma with occult primary after cetuximab-based chemotherapy:A case report

BACKGROUND Cancer of unknown primary(CUP)is a histological proven malignant tumor whose origin cannot be detected despite careful *** cervical lymph node metastases in CUP(80%)will originate from head and neck sites,and 15%show infiltration of squamous carcinoma *** survival rates of CUP are poor:The 5-year-survival rate ranges from 10%to 15%.First-line treatment recommendation for advanced,inoperable squamous cell carcinoma of head/neck(HNSCC)was cetuximab plus platinum-fluorouracil chemotherapy until recently,when checkpoint inhibitors proved clinically beneficial *** SUmmARY Here,we report a case of a 42-year-old female patient with cervical and abdominal lymph node and distant bone metastases of an occult primary of the head and neck(squamous cell carcinoma,human papillomavirus positive).The cancer was diagnosed during pregnancy 10 years ago,and after giving birth,the patient was treated with cetuximab plus platinum-fluorouracil chemotherapy achieving complete remission(CR).CR lasted 26 mo when new metastases(abdominal lymph node,lumbar vertebral body)*** manifestations were *** then on,the patient has not received any further treatment,and her disease has remained *** years after the initial cancer diagnosis,the patient is still alive and in good health,representing an exceptional case of *** This case illustrates the exceptional clinical course and benefits of combined therapy approaches in advanced metastatic HNSCC with occult primary.

Tolerance to individual and joint effects of arsenic and Bacillus thuringiensis subsp, israelensis or Lysinibacillus sphaericus in Culex mosquitoes

Arsenic contamination of global water supplies has come to the forefront in policy decisions in recent decades. However, the effects of arsenic on lower trophic levels of insects inhabiting contaminated ecosystems are not well understood. One approach to document both acute and sublethal effects of toxicants like arsenic is to assay them in combination with microbial pathogens to evaluate shifts in survival curves of the test organisms. Larvae of Culex quinquefasciatus and Culex tarsalis were reared in water containing 0 or 1 000μg/L of arsenate or arsenite. Fourth instars were then exposed to a range of doses of Bacillus thuringiensis subsp, israelensis （Bti） or Lysinibacillus sphaericus （Ls）, with shifts in lethal concentrations determined. Arsenic accumulation in 4th instars was also quantified, and a relative growth index （RGI） calculated for the treatments and compared to controls. Larvae of both species accumulated between 4 447 ± 169 ng As/g and 6 983 4- 367 ng As/g, though RGI values indicated accumulation did not affect growth and development. In all cases, the LC50＇s and LC90＇s of Cx. quinquefasciatus exposed jointly with arsenic and Bti/Ls were higher than Cx. tarsafis. Cx. tarsafis reared in arsenite showed a significant reduction in their Bti LC90 values compared to the control, indicating a sublethal effect of Bti. When exposed jointly with Ls, arsenite was more toxic than arsenate in Cx. tarsalis. Overall, these results indicate tolerance of these Culex species to arsenic exposures, and why this may occur is discussed.

magmatic Ni-Cu-(PGe) sulfide and Fe-Ti oxide deposits in plume-related large igneous provinces(LIPs)are commonly related to low-Ti and high-Ti series magmas, respectively, but the major factors that control such a relationship of metallogenic types and magma compositions are unclear. magma fOcontrols sulfur status and relative timing of Fe-Ti oxide saturation in mafic magmas, which may help clarify this issue. Taking the emeishan LIP as a case, we calculated the magma fOof the high-Ti and low-Ti picrites based on the olivine-spinel oxygen barometer, and the partitioning of V in olivine. The obtained fOof the high-Ti series magma(FmQ + 1.1 to FmQ + 2.6) is higher than that of the low-Ti series magma(FmQ-0.5to FmQ + 0.5). The magma fOof the high-Ti and low-Ti picrites containing Fo > 90 olivine reveals that the mantle source of the high-Ti series is likely more oxidized than that of the low-Ti series. The results using the ’lambda Ree’ approach show that the high-Ti series may have been derived from relatively oxidized mantle with garnet pyroxenite component. The S contents at sulfide saturation(SCSS) of the two series magmas were calculated based on liquid compositions obtained from the alpha melts modeling, and the results show that the low-Ti series magma could easily attain the sulfide saturation as it has low fOwith S being dominantly as S. In contrast, the oxidized high-Ti series magma is difficult to attain the sulfide saturation, but could crystallize Fe-Ti oxides at magma mgO content of ~7.0 wt.%. Thus, contrasting magma fOof low-Ti and high-Ti series in plume-related LIPs may play an important role in producing two different styles of metallogeny.

β-defensins and the epididymis： contrasting influences of prenatal, postnatal, and adult scenarios

β-defensins are components of host defense, with antimicrobial and pleiotropic immuno-modulatory properties. Research over the last 15 years has demonstrated abundant expression of a variety of β-defensins in the postnatal epididymis of different species. A gradient of region- and cell-specific expression of these proteins is observed in the epithelium of the postnatal epididymis. Their secretion into the luminal fluid and binding to spermatozoa as they travel along the epididymis has suggested their involvement in reproduction-specific tasks. Therefore, continuous attention has been given to various β-defensins for their role in sperm function and fertility. Although β-defensins are largely dependent on androgens, the underlying mechanisms regulating their expression and function in the epididymis are not well understood. Recent investigation has pointed out to a new and interesting scenario where β-defensins emerge with a different expression pattern in the Wolffian duct, the embryonic precursor of the epididymis, as opposed to the adult epididymis, thereby redefining the concept concerning the multifunctional roles of β-defensins in the developing epididymis. In this review, we summarize some current views of β-defensins in the epididymis highlighting our most recent data and speculations on their role in the developing epididymis during the prenatal-to-postnatal transition, bringing attention to the many unanswered questions in this research area that may contribute to a better understanding of epididymal biology and male fertility.

Background:The breakdown of dead organic matter is driven by a diverse array of organisms and is an important process increasingly impacted by a range of *** many studies have documented how contaminants affect food webs that are fueled by decaying plant litters,much less is known about how contaminants affect organisms that rely on dead animal ***,we begin to explore the effects of food contamination—using silver nanoparticles(AgNPs)as a model contaminant—on the carrion beetle Nicrophorus vespilloides that buries carcasses of small vertebrates in soils as food source and larval nursing ***:Our data show that a single ingestion of a non-lethal dose of 1μg mL^(−1) AgNPs by adult female beetles does not affect overall gut microbial activity but results in shifts in the gut microbial community composition towards pathogens including Alcaligenes,morganella,and *** no effects were observed in offspring clutch size,some reductions were visible in clutch weight,number of larvae,and number of eclosing pupae in exposed *** in comparison with *** ingestion of AgNPs over several weeks led to a decrease in survival of adult beetles,suggesting that more environmentally realistic exposure scenarios can directly affect the success of carcass-feeding ***:Sub-lethal carcass contamination with a model pollutant can affect the gut microbial composition in female beetles and reduce offspring *** encourages consideration of currently overlooked propagation routes of contaminants through necrophagous food webs and inherent consequences for ecological and evolutionary processes.

Inflammatory bowel diseases and spondyloarthropathies: From pathogenesis to treatment

Spondyloarthropathies(SpA) include many different forms of inflammatory arthritis and can affect the spine(axial SpA) and/or peripheral joints(peripheral SpA) with Ankylosing spondylitis(AS) being the prototype of the former. extraarticular manifestations, like uveitis, psoriasis and inflammatory bowel disease(IBD) are frequently observed in the setting of SpA and are, in fact, part of the SpA classification criteria. Bowel involvement seems to be the most common of these manifestations. Clinically evident IBD is observed in 6%-14% of AS patients, which is significantly more frequent compared to the general population. Besides, it seems that silent microscopic gut inflammation, is evident in around 60% in AS patients. Interestingly, occurrence of IBD has been associated with AS disease activity. For peripheral SpA, two different forms have been proposed with diverse characteristics. Of note, SpA(axial or peripheral) is more commonly observed in Crohn's disease than in ulcerative colitis. The common pathogenetic mechanisms that explain the link between IBD and SpA are still ill-defined. The role of dysregulated microbiome along with migration of T lymphocytes and other cells from gut to the joint("gut-joint" axis) has been recognized, in the context of a genetic background including associations with alleles inside or outside the human leukocyte antigen system. Various therapeutic modalities are available with monoclonal antibodies against tumour necrosis factor, interleukin-23 and interleukin-17, being the most effective. Both gastroenterologists and rheumatologists should be alert to identify the coexistence of these conditions and ideally follow-up these patients in combined clinics.
