Targeting new ways for large-scale,high-speed surface functionalization using direct laser interference patterning in a roll-to-roll process

New GATEWAY vectors for High Throughput Analyses of Protein-Protein Interactions by Bimolecular Fluorescence Complementation

Complex protein interaction networks constitute plant metabolic and signaling systems. Bimolecular fluorescence complementation （BiFC） is a suitable technique to investigate the formation of protein complexes and the localization of protein-protein interactions in planta. However, the generation of large plasmid collections to facilitate the exploration of complex interaction networks is often limited by the need for conventional cloning techniques. Here, we report the implementation of a GATEWAY vector system enabling large-scale combination and investigation of candidate proteins in BiFC studies. We describe a set of 12 GATEWAY-compatible BiFC vectors that efficiently permit the combination of candidate protein pairs with every possible N-or C-terminal sub-fragment of S（CFP）3A or Venus, respectively, and enable the performance of multicolor BiFC （mcBiFC）. We used proteins of the plant molybdenum metabolism, in that more than 20 potentially interacting proteins are assumed to form the cellular molybdenum network, as a case study to establish the functionality of the new vectors. Using these vectors, we report the formation of the molybdopterin synthase complex by interaction of Arabidopsis proteins Cnx6 and Cnx7 detected by BiFC as well as the simultaneous formation of Cnx6/Cnx6 and Cnx6/Cnx7 complexes revealed by mcBiFC. Consequently, these GATEWAY-based BiFC vector systems should significantly facilitate the large-scale investigation of complex regulatory networks in plant cells.

Restoration of the Western Himalaya： implications for metamorphic protoliths, thrust and normal faulting, and channel flow models

The Greater Himalayan Sequence (GHS) is composed of a sequence of Barrovian facies metamorphic rocks up to kyanite or sillimanite+K-feldspar grade, migmatites, layered stromatic migmatites and leucogranite sheets. These rocks were metamorphosed during the late Eocene to early miocene, and are bounded below by a large-scale SW-vergent ductile shear zone - thrust fault (main Central Thrust; mCT), and above by a NE-dipping low-angle normal sense shear zone and fault (Zanskar Shear Zone; ZSZ), part of the South Tibetan Detachment (STD) system. Restoration of the high-grade metamorphic rocks of the GHS reveals that protoliths are Proterozoic shales (Haimanta Group), Cambrian-Ordovician orthogneisses, Permian Panjal volcanics and Palaeozoic-Triassic/Jurassic sedimentary rocks, lateral equivalent rocks of the adjacent unmetamorphosed Tethyan zone. Structural outliers of low-grade or unmetamorphosed rocks (e.g., Chamba syncline) have been mapped overlying high-grade (usually sillimanite grade) rocks of the GHS. The low-grade klippen are underlain by low-angle north-directed ductile shear zones and normal faults, that are interpreted here as earlier equivalents of the ZSZ. These low-angle normal faults formed during continuous NE-SW compression, crustal thickening and southwestward extrusion of the GHS slab along the footwall. STD normal faults propagated structurally upward (northeast) with time, whereas mCT reverse faults along the base of the GHS propagated structurally downward (southwest) with time. Inverted metamorphic isograds along the base of the slab (mCT zone) can be linked on the map with right way-up isograds along the footwall of the ZSZ normal fault at the top of the slab, demonstrating that the recumbently folded isograd model is a viable geometrical representation of the metamorphic structure of the GHS. Post-metamorphic shearing along the ZSZ and mCT has condensed the isograds by a combination of pure shear and simple shear. These observations support the C

Carbon dioxide exchange processes over the grassland ecosystems in semiarid areas of China

We present results of a high resolution study of the filamentary infrared dark cloud G192.76＋00.10 in the S254-S258 OB complex in several molecular species tracing different physical conditions. These include three isotopologues of carbon monoxide （CO）, ammonia （NH3） and carbon monosulfide （CS）. The aim of this work is to study the general structure and kinematics of the filamen- tary cloud, and its fragmentation and physical parameters. The gas temperature is derived from the NH3 （J, K） = （1, 1）, （2, 2） and 12CO（2-1） lines, and the 13CO（1-0）, 13CO（2-1） emission is used to inves- tigate the overall gas distribution and kinematics. Several dense clumps are identified from the CS（2-1） data. Values of the gas temperature lie in the range 10 - 35 K, and column density N（H2） reaches the value 5.1 ×1022 cm-2. The width of the filament is of order 1 pc. The masses of the dense clumps range from ×30 m⊙ to - 160 m⊙. They appear to be gravitationally unstable. The molecular emission shows a gas dynamical coherence along the filament, The velocity pattern may indicate longitudinal collapse.

Comparison of high-resolution ultrasound and mR-enterography in patients with inflammatory bowel disease

AIm:To compare the results of high-resolution ultrasound(HR-US) and magnetic resonance enterography(mRE) examinations in patients with inflammatory bowel disease(IBD).mETHODS:The reports of 250 consecutive cases with known IBD,who had an mRE and HR-US examination,were retrospectively *** a patient-based approach we evaluated morphological disease features such as affected bowel wall,stenosis,abscess and *** comparison between the two modalities was based on the hypothesis,that any pathological change described in any imaging modality was a true finding,as no further standard of reference was available for complete ***:Two hundred and fifty examinations representing 207 different patients were *** modalities assessed similar bowel wall changes in 65% of the examinations,with more US findings in 11% and more mRE findings in 15%.When the reports were analyzed with regard to "bowel wall inflammation",US reported more findings in 2%,while mRE reported more findings in 53%.Stenoses were assessed to be identical in 8%,while US found more in 3% and mRE in 29%(P more findings in 2%(n = 4) while mRE detected more in 6%(n = 16).US detected more fistulas in 1%(n = 2),while mRE detected more in 13%(n = 32)(P most common reason for no detected pathology by US was a difficult to assess anatomical region(lesser pelvis,n = 72).CONCLUSION:US can miss clinically relevant pathological changes in patients with IBD mostly due to difficulty in assessing certain anatomical regions.

Positive association between forest management, environmental change, and forest bird abundance

Background: The global decrease in wildlife populations, especially birds, is mainly due to land use change and increasing intensity of land use(Parmesan and Yohe 2003). However, impacts of management tools to mitigate biodiversity loss at regional and global scales are less apparent in forest regions that have a constant forest area,and which did not suffer from habitat degradation, and where forests are sustainably managed, such as in Central Europe or the northeastern USA. A biodiversity assessment for Germany suggested, for example, that bird populations were constant(Bundesamt für Naturschutz 2015).Results: This study shows that changes in the environment and in forest management over the past 45 years have had a significant, positive effect on the abundance of non-migratory forest bird species in Central Europe. Economy(timber prices and GDP), forest management(timber harvest and mixed forest area), and environmental factors(atmospheric CO_2 concentration and nitrogen deposition) were investigated together with changes in abundances of migratory and non-migratory forest birds using partial least squares path modeling. Climate change, resulting in longer seasons and milder winters, and forest management, promoting tree diversity, were significantly positively related to the abundance of non-migratory forest birds and explained 92% of the variation in their abundance in Europe. Regionally-migrating forest birds had stable populations with large variation, while birds migrating across continents declined in recent decades, suggesting significant, contrasting changes in bird populations in Europe. In northeastern North America we also found evidence that non-migratory forests have experienced long-term increases in abundance, and this increase was related to management.
