Antiseptic Efficacy of A Soap Made from Biosurfactants Isolated from Bacillus and Lactobacillus against Pathogenic Bacteria

The aim of our study was to use a biosurfactant produced by Bacillus and Lactobacillus isolates as an antiseptic in the formulation of local soap. A total of 60 isolates were characterized by microbiological techniques (30 Bacillus and 30 Lactobacillus) and the ability to produce biosurfactants was demonstrated by a hydrocarbon emulsification index (E24). The emulsification indexes (E24) varied from 9% to 100% for Bacillus and from 33% to 100% for Lactobacillus as well. The antagonistic assay showed that biosurfactants were able to inhibit the formation of biofilms and growth of pathogens such as Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus cereus, Salmonella typhirium, Shigella boydii and Proteus mirabilis. The biosurfactant consortium (BioC) from Bacillus consortium and from Lactobacillus was able to inhibit biofilm formation and the pathogens growth. The BioC was stable to alkaline pH and the temperatures stability of Biosurfactant was ranging from 50°C to 90°C. The soap was made by the cold saponification process using one biosurfactant consortium formulated. This soap has a pH of 10 and showed good cleaning power and good foam stability. Similarly, the soap showed good antiseptic power and disinfection power against all pathogens tested. Handwashing is critical to preventing disease transmission. The persistence of pathogens in waste water was evaluated. The BioS produced showed good disinfection power against all pathogens tested. The valor of reduction on the hands and in the waste water was significantly more than compared to the control soaps used. This soap could be used in the prevention, fighting, and treatment of bacterial and viral infections.

First Development of a Biotechnological Ferment Based on a Consorsium of the Genus <i>Bacillus</i>for the Optimization of the Fermentation Process of Cassava Tubers

Due to its nutritional values, cassava has become an unavailable food and is one of the essential foods in the Republic of Congo. Fermentation of tubers is still traditional. Fiftyrod-shaped spore-forming bacteria were screened and carried out in batch mode for the fermentation abilities of cassava tubers in order to develop biotechnological starter. The Penetrometry Index (PI) has been used to screen bacteria and 16SrRNA as well as fibEone step multiplex PCR which were used to molecularly identify isolates. Emulsification Index, Proteolytic as well as amylolytic, and cellulolytic activities of some strains were quantitatively evaluated for prooving orgaleptic characterics. As results Bacillus subtilis (MT994787), Bacillus subtillis (MT994789), Bacillus tequilensis (MT994788), Bacillus safensis, and Bacillus subtilis have been identified. Single isolates were able to ferment tubers in 48 h and 72 hours meanwhile Bacillus consortia were able to shift fermentation of tubers from 48 hours to 24 hours. The consortium could be used as the major bacterial starters. Strains were associated with the ability to secrete biomolecules including biosurfactants, protease, amylase and cellulase.

Shigella Strain Has Developed Non-Studied Pathogenicity Mechanisms of Adaptability in the Colonization of Epithelial Cells

According to the World Health Organization, foodborne diseases are a major public health problem, particularly in developing countries including the Republic of Congo. They are responsible for several episodes of diarrhea, especially in children under five years old. There is no reliable epidemiological data on the pathogenicity of the Shigella spp. strains circulating in the whole Republic of Congo drafting this paper. The purpose of this study was to examine the Shigella spp strain pathogenicity close to an environment contaminated with faeces in the city of Brazzaville. As a result, 54 isolates have been associated with Shigella spp. The gastric acid resistance test performed on Shigella Environmental Strain (SES) and Shigella Clinical Strains (SCS) resulted in 38.8% (21/54) and 100% acid resistant, respectively. Shigella spp. Strains (SES and SCS) were ranged in a survival percentage from 11% to 93%. By monitoring Biosurfactant-Like Molecule (BLM) production, we showed that the BLM production of SES and SCS was highly dependent on bacterial culture density involving the Quorum Sensing (QS). S. flexneri, S. boydii, and S. sonnei and as well as SES and SCS were able to invade and contaminate eggs by colonizing egg yolk. The counting bacteria were ranging from two to 5 × 107 CFU/mL after contamination. Concomitantly, BLM was secreted during the post contamination of poultry eggs with 100% EI24. Further by trying to show the pathogenicity by the hemolysis test, we have shown that SES and SCS were able to show significant areas of lysis on blood agar. Finally, this work has proposed an additional model of cell invasion including biosurfactants during the pre- and post-invasion phases.

Giving More Benefits to Biosurfactants Secreted by Lactic Acid Bacteria Isolated from Plantain Wine by Using Multiplex PCR Identification

Fermented beverages have continued to give more surprises in terms of the presence of biomolecules and the diversity of microorganisms that may be contained. Republic of Congo is home to a panoply of fermented foods and beverages that are still not yet studied. This is the case of plantain wine fluently called banana wine. Within this framework, this work aims to study the role of Biosurfactant-like Biomolecules secreted during fermentation of plantain wine. Using MRS medium, 15 isolates bacteria have been found. 100% are able to secrete biosurfactant and 66.66% are extractible biosurfactants. 33% of isolates have been associated to Lactobacillus plantarum (Is2, Is9, Is12 and Is13) by using a one-step multi-plex PCR that targets genes encoding for bacteriocins. Biosurfactants secreted by L. plantarum play an important role in the preservation of banana wine. The biosurfactants extracted with chloroform and ammonium sulphate are able to inhibit the growth of pathogenic bacteria including Shigella flexneri, Salmonella spp., Pseudomonas aeruginosa and Staphy-lococcus aureus.

Prime Enzymatic Exocellular Background of Lysinibacillus louembei

Many high-reading journals reject several studies based on the basic aspects of microbiology by forgetting that this could open doors and windows of great scientific discoveries. A new spore-forming bacteria species called Lysinibacillus louembei has been previously discovered in our research unit. This new study aims to assess a real enzymatic machine produced with such bacteria. Using agarose mixed with half skimmed milk, LB medium supplemented with tween 20, Olive oil, egg yolk, cellulose, casein, pectin and starch, we showed that L. louembei has lipolytic, proteolytic (2.8 cm ± 0.1), cellulolytic, amylolytic pectinolytic activities, with percentage ranging from 30% to 80%. This species is able to secrete lipase, thermostable protease up to 60°C. Cellulase, pectinase and amylase are secreted with more stability between 30°C and 50°C with an optimum at 45°C. The effect of pH was determined after 24 h of incubation at 37°C and 50°C. The proteolytic activity is stable at pH 8 at 50°C. In addition to this first enzymatic landscape, L. louembei is able to degrade and/or tolerate gasoline and/or diesel fuel hydrocarbons in 36 hours. In this work, we also showed that L. louembei can produce biosurfactant in the presence of gasoline and SAE140 with the E24 up to 50%.

Molecular Profiling of Sugarcane Wine, a Traditional Fermented Beverage (Loungouila) from Madingou

A number of studies reported that traditional fermented beverages possessed pharmaceutical biomolecules involved in biocatalysis for good therapeutic effects on various pathology including tumor, diabetes, inflammation, and obesity. This dimension of understanding is the prerogative of the biomolecular profile found in these fermented foods and beverages. The current work aimed to study the postfermentation molecular profile of the Congolese fermented beverage (Lougwila). The determination of pH, the acidity titratable, the distillation of sugar cane, the determination of total polyphenols and flavonoids and the matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) have been used. As results, the fermenting sugar cane juice at room temperature for a period of seven days, leading to a decrease of the pH value to 4.25 ± 0.10 and increase in titratable acidity and alcohol content of at least 6.421 g/L (w/v) and 7% respectively. The polyphenol concentration of Loungouila increase from 14.9 to 20.5 mg Eq AG/g Ms after 10 days of fermentation. The flavonoid concentration varies from 1.70 to 5.22 mg Eq Cat/g Ms. among 46 isolates of Bacillus species, 55% (25/45) were able to show a very interesting clear zones in terms of cellulolytic activity with the percentage ranging from 37.3% to 83.9%, and 41.3% of amylolytic activity for the percentage ranging from 52.02% to 75%. 65.21% (30/46) of the isolates tested were found to be positive by the caseinolytic test with zones of inhibition ranging in diameter from 1.10 ± 0.09 to 3.25 ± 0.07 cm. In addition to the determination of biomolecule profile, 34.78% (16/46) of Bacillus isolates were able to produce biosurfactants with percentages ranging from 14% to 100%. Proteomic profiling of Loungouila has been investigated by using MALDI-TOF Technique. Retrospective study of the incidence, risk factors, treatment outcomes of bacterial infections at uncommon sites in cirrhotic patients

Numerical Optimization by Finite Element Method of Stainless Steel/Glass-Epoxy Composite Bolted Joint under Tension and Compression

The aim of this study was to optimize the geometry and the design of metallic/composite single bolted joints subjected to tension-compression loading. For this purpose, it was necessary to evaluate the stress state in each component of the bolted join. The multi-material assembly was based on the principle of double lap bolted joint. It was composed of a symmetrical balanced woven glass-epoxy composite material plate fastened to two stainless sheets using a stainless pre-stressed bolt. In order to optimize the design and the geometry of the assembly, ten configurations were proposed and studied: a classical simple bolted joint, two joints with an insert (a BigHeadR insert and a stair one) embedded in the composite, two “waved” solutions, three symmetrical configurations composed of a succession of metallic and composites layers, without a sleeve, with one and with two sleeves, and two non-symmetrical constituted of metallic and composites layers associated with a stair-insert (one with a sleeve and one without). A tridimensional Finite Element Method (FEM) was used to model each configuration mentioned above. The FE models taked into account the different materials, the effects of contact between the different sheets of the assembly and the pre-stress in the bolt. The stress state was analyzed in the composite part. The concept of stress concentration factor was used in order to evaluate the stress increase in the highly stressed regions and to compare the ten configurations studied. For this purpose, three stress concentration factors were defined: one for a monotonic loading in tension, another for a monotonic loading in compression, and the third for a tension-compression cyclic loading. The results of the FEM computations showed that the use of alternative metallic and composite layers associated with two sleeves gived low values of stress concentration factors, smaller than 1.4.
