Importance of the stem cell microenvironment for ophthalmological cell-based therapy

Cell therapy is a promising treatment for diseases that are caused by cell degeneration or death. The cells for clinical transplantation are usually obtained by culturing healthy allogeneic or exogenous tissue invitro. However, for diseases of the eye, obtaining the adequate number of cells for clinical transplantation is difficult due to the small size of tissue donors and the frequent needs of long-term amplification of cells in vitro, which results in low cell viability after transplantation. In addition, the transplanted cells often develop fibrosis or degrade and have very low survival. Embryonic stem cells(ESCs) and induced pluripotent stem cells(i PS) are also promising candidates for cell therapy. Unfortunately, the differentiation of ESCs can bring immune rejection, tumorigenicity and undesired differentiated cells, limiting its clinical application. Although i PS cells can avoid the risk of immune rejection caused by ES cell differentiation post-transplantation, the low conversion rate, the risk of tumor formation and the potentially unpredictable biological changes that could occur through genetic manipulation hinder its clinical application. Thus, the desired clinical effect of cell therapy is impaired by these factors. Recent research findings recognize that the reason for low survival of the implanted cells not only depends on the seeded cells, but also on the cell microenvironment, which determines the cell survival, proliferation and even reverse differentiation. When used for cell therapy, the transplanted cells need a specific three-dimensional structure to anchor and specific extra cellular matrix components in addition to relevant cytokine signaling to transfer the required information to support their growth. These structures present in the matrix in which the stem cells reside are known as the stem cell microenvironment. Evaluation of Slow Light Periodic Signals Considering the Distortion in EDF

Based on the fundamental harmonic phase delay, a new definition of fundamental harmonic fractional delay （FHFD） is proposed to evaluate slow light with the consideration of signal distortion, to eliminate the dependence on the choice of the reference point. By solving the rate equation of erbium-doped fiber （EDF）, it is shown that the slow light always accompanies the signal distortion when the periodic signal propagates in EDF, and FHFD depends on the signal distortion, as well as the average input power, the modulation depth and the length of EDF. The results of simulations and experiments indicate that the definition of FHFD is reasonable and effective to evaluate the slow light of periodic signals.

Low concentration of sodium hyaluronate temporarily elevates the tear film lipid layer thickness in dry eye patients with lipid deficiency

AIM： To investigate the effects of different concentrations of artificial tears on lipid layer thickness （LLT） and blink rate （BR） in dry eye patients. METHODS： This study included 106 eyes of 58 patients with dry eye. The lipid deficiency type was defined as the LLT baseline 〈75 nm. The LLT and BR were measured at baseline and 1, 5 and 15min after the instillation of 0.1% or 0.3% sodium hyaluronate （SH） eye drops by using the LipiView ocular surface interferometer. RESULTS： In the lipid deficiency group, the LLT increased from baseline at 1rain post instillation. The LLT after the instillation of 0.1% SH was significantly higher than that after the instillation of 0.3% SH （P〈0.001）. The LLT returned to baseline at 15min post instillation of 0.1% SH and at 5min post instillation of 0.3% SH. In the non-lipid deficiency group, the LLT decreased from baseline at lmin and returned to baseline at 5rain for both treatments. The BRs were not significantly different at different time points for both treatments. CONCLUSION： SH eye drops induce a short-term increase in LLT of patients with lipid deficiency. A low concentration of artificial tears have a stronger effect than a high concentration of artificial tears on the increase in LLT. in comparison, SH eye drops induce a transient and slight decrease in LLT of patients without lipid deficiency. A low concentration of artificial tears might be better for patients with lipid deficiency.

Lipid layer thickness and tear meniscus height measurements for the differential diagnosis of evaporative dry eye subtypes

AIM： To explore a new diagnostic index for differentiating the evaporative dry eye（EDE） subtypes by analysis of their respective clinical characteristics. METHODS： A cross-sectional study of 139 patients（139 eyes） with EDE who were enrolled and classified as obstructive meibomian gland dysfunction（MGD）（n=81） and non-obstructive MGD（n=58） EDE. All patients completed a Standard Patient Evaluation of Eye Dryness（SPEED） questionnaire and were evaluated for average lipid layer thickness（LLT）, tear meniscus height measurements（TMH）, tear break-up time（TBUT）, ocular surface staining score, Schirmer I test（SIT）, lid margin abnormalities, and meibomian gland function and morphology. RESULTS： Age, average LLT, TMH, scores of lid margin abnormalities, meibum quality, meibomian gland loss（MGL）（all P≤0.001）, and TBUT（P=0.03） were all significantly different between obstructive MGD EDE patients and nonobstructive MGD EDE patients. Average LLT in obstructive MGD EDE was correlated with meibomian expressibility（r=-0.541, P≤0.001）, lid margin abnormalities were marginally not significant（r=0.197, P=0.077）, and TMH was correlated with MGL（total MGL： r=0.552, P≤0.001; upper MGL： r=0.438, P≤0.001; lower MGL： r=0.407, P≤0.001）. Average LLT in non-obstructive MGD EDE, was correlated with meibomian expressibility and Oxford staining（r=-0.396, P=0.002; r=-0.461, P≤0.001）. The efficiency of combining average LLT and TMH was optimal, with a sensitivity of 80.2% and a specificity of 74.1%. Obstructive MGD EDE patients had an average LLT≥69 nm and TMH≥0.25 mm, while non-obstructive MGD EDE patients had an average LLT〈69 nm and TMH〈0.25 ***： Obstructive MGD EDE and nonobstructive MGD EDE have significantly different clinical characteristics. Combining average LLT and TMH measurements enhanced their reliability for differentiating these two subtypes and provided guidance for offering more precise treatments for EDE subtypes.

Aim:The antihyperglycemic effects of the total ginsenosides in Chinese ginseng(TGCG),extracted from leaves and the stem,were evaluated in diabetic C57BL/6Job/ob ***:Animals received daily intraperitoneal injections of TGCG(100 and 200 mg/kg)or oral administration(150 and 300 mg/kg)for 12 *** glucose levels and body weight were measured after fasting the animals for4 *** glucose use was also measured using an intraperitoneal glucosetolerance ***:In the injection group,a high dose of TGCG(200 mg/kg)significantly lowered the fasting blood glucose levels in ob/ob mice on d 12(153±16 mg/dL vs 203±9.8 mg/dL,P<0.01,compared to vehicle-treated group).In theoral group,blood glucose decreased notably with a dose of TGCG(300 mg/kg)ond 12(169.1±12.6 mg/dL vs 211.6±13.8 mg/dL,P<0.05,compared to the vehicle-treated group).Glucose tolerance was also improved markedly in ob/ob ***,a significant reduction in bodyweight(P<0.05)was observed after12 d ofTGCG(300 mg/kg)treatment in mice from the oral ***:Theresults indicated that in a diabetic ob/ob mouse model TGCG was endowed withsignificant anti-hyperglycemic and anti-obesity ***,the totalginsenosides extracted from Chinese ginseng leaves and the stem may have somepotential for treating diabetes.

Gas quenching and vacuum quenching process are widely applied to accelerate solvent volatilization to induce nucleation of perovskites in blade-coating *** this work,we found these two pre-crystallization processes lead to different order of crystallization dynamics within the perovskite thin film,resulting in the differences of additive *** then tailor-designed an additive molecule named 1,3-bis(4-methoxyphenyl)thiourea to obtain films with fewer defects and holes at the buried interface,and prepared perovskite solar cells with a certified efficiency of 23.75%.Furthermore,this work also demonstrates an efficiency of 20.18%for the large-area perovskite solar module(PSM)with an aperture area of 60.84 cm^(2).The PSM possesses remarkable continuous operation stability for maximum power point tracking of T_(90)>1000 h in ambient air.

As one of the most commonly used medicinal plants,ginseng has been an attractive model herb for study.A wide range of analytical methods has been used to characterize its ***,less effort has been devoted to the rare ginseng saponins,especially their isomers and sugar *** this study,we used segmental monitoring and diagnostic ion filtering to characterize ginseng saponins by rapid liquid chromatography with time-of-flight mass spectrometry(LC-TOF-MS).By using selected diagnostic ions,specific groups of ginseng saponins were readily extracted from the complicated matrix.20(R) and 20(S) stereo-saponins were differentiated using the peak abundance ratio of[M-H20+H]+ to[M-2H20+H]+.The fragmentation behavior of ginsenosides was first reported in negative ion mode by MS/MS with high-energy collision-induced dissociation, producing rules to determine sugar numbers,positions and *** the rules,we identified and compared the nontarget ginseng saponin profiling of raw and steamed American ginseng roots and *** characterized 70 saponins in the *** strategy can be extended to screen and characterize other rare ginseng saponins and their metabolites

Chemical Constituents from Leaves of Oplopanax horridus

Objective To study the chemical constituents from the leaves of Oplopanax horridus. Methods The chemical constituents were isolated and purified by column chromatography on silica gel and Sephadex LH-20 gel columns, 1H-NMR and 13C-NMR were applied for the identification of chemical structure. Results Ten compounds were isolated and identified as dammara-20,24-dien-3β-ol acetate（1）, phytol（2）, 16 Z,19Z-pentacosadienoic acid（3）, β-sitosterol（4）,（3S,8S）-falcarindiol（5）, maltol（6）, acankoreagenin（7）, daucosterol（8）, stigmasterol-3-O-β-D-glucopyranoside（9）, and acankoreoside A（10）. Conclusion Compounds 1-3, 6, and 10 are isolated from this plant for the first time. Compounds 1-3 and 6 are isolated from the plants in genus Oplopanax Miq. for the first time. Moreover, Compounds 1, 3, and 6 are isolated from the plants in the family of Araliaceae for the first time.

Overview of in-situ oxygen production technologies for lunar resources

The rich resources and unique environment of the Moon make it an ideal location for human expansion and the utilization of extraterrestrial ***,crucial for supporting human life on the Moon,can be extracted from lunar regolith,which is highly rich in oxygen and contains polymetallic *** oxygen and metal extraction can be achieved using existing metallurgical ***,the ample reserves of water ice on the Moon offer another means for oxygen *** paper offers a detailed overview of the leading technologies for achieving oxygen production on the Moon,drawing from an analysis of lunar resources and environmental *** delves into the principles,processes,advantages,and drawbacks of water-ice electrolysis,two-step oxygen production from lunar regolith,and one-step oxygen production from lunar *** two-step methods involve hydrogen reduction,carbothermal reduction,and hydrometallurgy,while the one-step methods encompass fluorination/chlorination,high-temperature decomposition,molten salt electrolysis,and molten regolith electrolysis(MOE).Following a thorough comparison of raw materials,equipment,technology,and economic viability,MOE is identified as the most promising approach for future in-situ oxygen production on the *** the corrosion characteristics of molten lunar regolith at high temperatures,along with the Moon's low-gravity environment,the development of inexpensive and stable inert anodes and electrolysis devices that can easily collect oxygen is critical for promoting MOE technology on the *** review significantly contributes to our understanding of in-situ oxygen production technologies on the Moon and supports upcoming lunar exploration initiatives.
