A biosynthetic gene cluster for the bioactive fungal sesterterpenoids variecolin(1)and variecolactone(2)was identified in Aspergillus aculeatus ATCC *** production of 1 and 2 was achieved in Aspergillus oryzae by expressing the sesterterpene synthase VrcA and the cytochrome P450 ***,the replacement of VrcB with homologous P450s from other fungal terpenoid pathways yielded three new variecolin analogues(5-7).Analysis of the compounds’anticancer activity in vitro and in vivo revealed that although 5 and 1 had comparable activities,5 was associated with significantly reduced toxic side effects in cancer-bearing mice,indicating its potentially broader therapeutic *** study describes the first tests of variecolin and its analogues in animals and demonstrates the utility of synthetic biology for creating molecules with improved biological activities.

EKLF is an erythroid-specific,zinc finger-containing transcription factor essential for the activation of the mamma-lian beta globin gene in erythroid cells of definitive *** have prepared a polyclonal anti-mouse EKLF antibodysuitable for Western blotting and immunoprecipitation(IP)qualities,and used it to define the expression patterns of theEKLF protein during mouse erythroid *** have also used this antibody for the chromatin-immunoprecipi-tation(ChIP)*** was found to bind in vivo at both the mouse beta-major-globin promoter and the HS2 site ofbeta-LCR in the mouse erythroleukemia cells(MEL)in a DMSO-inducible *** DMSO-induced bindings ofEKLF as well as three other proteins,namely,RNA polymerase Ⅱ,acetylated histone H3,and methylated histone H3,were not abolished but significantly lowered in CB3,a MEL-derived cell line with null-expression of p45/NF-E2,anerythroid-enriched factor needed for activation of the mammalian globin ***,binding of EKLF in vivowas also detected in the mouse alpha-like globin locus,at the adult alpha globin promoter and its far upstream regula-tory element alpha-MRE(HS26).This study provides direct evidence for EKLF-binding in vivo at the major regulatoryelements of the mouse beta-like globin gene clusters the data also have interesting implications with respect to the roleof EKLF-chromatin interaction in mammalian globin gene regulation.

Background Much is known about the cytogenetic lesions that characterize multiple myeloma （MM） patients from the USA, Europe, and East Asia. However, little has been published about the disease among Southeast Asians. The aim of this study was to determine the chromosomal abnormalities of MM patients in our Singapore population. Methods Forty-five newly-diagnosed, morphologically confirmed patients comprising 18 males and 27 females, aged 46-84 years （median 65 years） were investigated by karyotyping and fluorescence in situ hybridization （FISH）. FISH employing standard panel probes and lp36/lq21 and 6q21/15q22 probes was performed on diagnostic bone marrow samples. Results Thirty-four cases （75.6%） had karyotypic abnormalities. Including FISH, a total detection rate of 91.1% was at- tained. Numerical and complex structural aberrations were common to both hyperdiploid and non-hyperdiploid patients. Numerical gains of several recurring chromosomes were frequent among hyperdiploid patients while structural rear- rangements of several chromosomes including 8q24.1 and 14q32 characterized non-hyperdiploid patients. With FISH, immunoglobulin heavy chain （IGH） gene rearrangements, especially fibroblast growth factor receptor 3 （FGFR3）/IGH and RB1 deletion/monosomy 13 were the most common abnormalities （43.4%）. Amplification lq21 was 10 times more frequent （42.5%） than del（1p36） and del（6q21）. Conclusions We have successfully reported the comprehensive cytogenetic profiling of a cohort of newly-diagnosed myeloma patients in our population. This study indicates that the genetic and cytogenetic abnormalities, and their frequencies, in our study group are generally similar to other populations.

Background： Antibody drug conjugated （ADC） is one kind of very important method of therapy to cancer diseases. In this research, the authors introduce BLAST and some other important algorithms to create transmembrane protein databases. These databases are acquired from well-known databases such as NCBI or Swiss-Prot as template, and then collect all possible transmembrane protein by using BLAST or physical character. After collect these databases, the authors will aim at each nucleotide sequences to design the probes of oligonucleotide microarray, which can detect the high express transmembrane proteins very efficiently. Finally, the authors can accelerate the anti-cancer drug discovery by using these databases. Result： This study constructed a web service, the Transmembrane Protein Database, to researchers that are interested in or need to oligonucleotide microarray probe design for detecting potential targets of antibody drug. With user friendly web based windows containing each necessary selections, users can easily choose the parameters and get the suitable probe design suggestions. Conclusion： Transmembrane protein database is very important and powerful in detecting cancers or other human disease. By using this database, the authors offer a good strategy in transmembrane protein research as well.

Aim: Sorafenib is a multi-tyrosine kinase inhibitor and the standard therapy for advanced hepatocellular carcinoma (HCC). This retrospective study aimed to observe the anti-fibrotic effect of sorafenib in patients with advanced HCC. Methods: Seventeen patients with advanced HCC were recruited. Shear wave velocity (SWV) using acoustic radiation force impulse elastography and non-invasive serum markers for liver fibrosis, such as the aspartate aminotransferase (AST) to alanine aminotransferase ratio (AAR), the AST to platelet ratio index, the fibrosis-4 index and the Lok index, were recorded at the beginning of sorafenib treatment and 3-6 months after sorafenib treatment in 2014-2015. Results: Nine (52.9%) patients achieved disease control status and 8 had progressive disease after a mean duration of 11.1 months with sorafenib treatment. The mean SWV decreased from 2.37 m/s at the beginning to 1.90 m/s after sorafenib treatment (P < 0.01). This trend was observed in patients with and without liver cirrhosis (from 2.49 to 2.06 m/s, P = 0.06, and from 2.32 to 1.69 m/s, P < 0.05, respectively). Among the non-invasive serum markers, no statistically significant differences were observed except for the AAR in the cirrhotic group. Conclusion: Sorafenib has potential antif-ibrotic effects in patients with advanced HCC.

高电容量的电极材料对于需要高能量密度锂离子的携带式电子设备相当重要。由于需求不同,电极的制作方式与材料的选用相当繁杂。本研究是将回收硅粉和沥青粉末混合,再与炭毡复合成炭纸,经250℃稳定化与1 000℃炭化后,制备出兼具电极与集流板功能炭复合纸。经循环充放电测试后,添加不同比例硅粉(2.5 wt%, 5 wt%, 10 wt%)的负极,相较于炭复合电极放电电容量分别提升了94%、129%和41%。

本研究旨在了解“城乡”居民之绿色消费态度与绿色消费行为,并探讨不同的背景变项、独立性价值观及自利道德价值观对绿色消费态度与绿色消费行为的影响,以及绿色消费态度与绿色消费行为间的关系。对大台北都会区以及南投各乡镇两地区作调查,各发放350份问卷,有效问卷625份,结果显示城乡居民整体绿色消费态度为正向;背景变项中的性别、年龄、教育程度、职业,与绿色消费态度有显着差异;月收入与居住地则无显着差异;在绿色消费行为方面,城乡居民整体绿色消费行为均为正向的,其中唯“拒用原则”分数偏低;绿色消费态度与绿色消费行为呈现显着正相关;居民的社会独立性越低,其绿色消费的态度与行为的表现愈正向;居民的自利道德感愈低,其绿色消费的态度与行为的表现愈正向。

自1921年勿万洛夫氏***发表了蝗虫变型学说以来,昆虫学者们针对两型的差异,在形态上,蝗蝻的生活习性及成虫生殖力等问题上进行了多方面的研究,对飞蝗变型的知识已逐渐丰富起来。但关於飞蝗成虫习性的认识,过去尚不够十分明确,因而有些学者误认为散栖型飞蝗的习性是离群散棲,不群聚亦不高飞远扬,甚至以此习性作为区分两型的论据之一。
