The present study has investigated the regulation of malignant phenotype and gene expression in a human promyelocytic leukemia cell mutant （HL-60-AR） by means of cellular engineering technique of cybridization between the fusions of the mutant cells with enucleated mouse reticulocytes. Results indicate that the cybrid cells （HL-R） incorporated with reticulocyte cytoplasts become differentiated, and the malignancy is obviously suppressed or reversed to a certain degree when compared with those of parental tumor cells. They lose the growth ability to form colony soft agar medium, become non-tumorigenic under heterotransplantation to nude mice, and are accompanied by decrease in growth rate, cellular mitotic index and DNA synthesis. No gene transcripts or homologous sequence of mRNA corresponding to c-myc oncogene can be detected in cybrid cells by Northern blot technique with cloned cmyc gene probe, suggesting that the expression of originally active c-myc has been inhibited. A cDNA expressed specifically in late stages of mouse spermatogenic cells during spermato- genesis was cloned by using overlapping RT-PCR and RACE. The cDNA contained an open reading frame (ORF) of 2625 base pairs that encoded an 874 amino acids protein. Comparison analysis of amino acid sequence showed 91% and 80% identity to a rat homologue XP-226242 and a monkey homologue BAB63.15 respectively. The expression of the mRNA was only observed in pachytene spermatocytes, round, and elongating spermatids. We named this gene as SRG-L (spermatogenesis related gene expressed in the late stages of spermatogenic cells, GenBank accession No. AY3.2586). The tissue-specific analysis showed that the SRG-L was highly expressed in spleen and testis. The results suggested that SRG-L might play special roles during spermatogenesis, particularly related to meiosis and spermiogenesis. Analysis of the amino acid sequence showed there was a coiled-coil region near the N-terminal region and rich phosphorylation sites, suggesting SRG-L might function as a transmembrane protein mediating signal transduction. This study also demonstrated that gene cloning by RT-PCR was applicable and convenient when its homologous gene was known.

Avian influenza A virus constitutes a large threat to human health. Recent outbreaks of highly pathogenic avian influenza H5N1 virus in poultry and in humans have raised concerns that an influenza pandemic will occur in the near future. Transmission from avian species to humans remains sporadic, but the mortality associated with human infection is very high (about 62%). To date, there are no effective therapeutic drugs or a prophylactic vaccines available, which means that there is still a long way to go before we can eradicate or cure avian influenza. This review focuses on the molecular pathogenesis of avian influenza H5N1 virus infection. An understanding of the viral pathogenesis may facilitate the development of novel treatments or effective eradication of this fatal disease.

In previous studies, we found that EDRF2 was an erythroid differentiation related factor, whose expression was markedly upregulated during erythroid differentiation. It suggested that this factor played a role in erythropoiesis. By using rapid amplification of cDNA ends technology, we cloned EDRF2 gene 5’-and 3.-cDNA ends successfully. Transfection and Northern blot analysis demonstrated that EDRF2 inhibited mRNA expression of a-globin gene, while did not regulate γ-globin gene expression. Gel shift assay confirmed that DNA-binding activity of erythroid specific transcription factors GATA-1, NF-E2 and AP1 was not affected by either forced overexpression or artificial down-regulation of EDRF2 gene in K562 cells. However, we detected that the negative regulator of expression of GATA-1 transcription factor was increased in EDRF2 overexpressed K562 cells. A novel element at -153.-143.bp in the inter-leukin 2 receptor α(IL-2Rα) gene has been coined as NRE-inverse repeat sequence (NIRS) due to its inversely repeated to the known negative regulatory element (NRE) further upstream of the gene. In order to explore the role of NIRS in the expression of IL-2Rα gene, luciferase reporter plasmids driven by 4 individually deleted IL-2Rα genes promoter regions were constructed. Transfection of the reporter plasmids into Jurkat cells and HeLa cells respectively, we found that both NIRS and NRE were critical for repressing the constitutive expression of IL-2Rα gene and were also necessary for promoter activity induced by PHA. EMSA results showed that double-stranded NRE- and NIRS-bind-ing proteins existed in both HeLa cells and Jurkat cells. However, single-stranded NIRS- and NRE-binding protein was only found in HeLa cells. 艰难梭菌感染(CDI)指由艰难梭菌引起的肠道感染,主要临床症状为发热、腹痛、水样便、腹泻。轻者表现为腹泻,严重者可发生伪膜性肠炎,且常伴有中毒性巨结肠、肠穿孔、感染性休克等并发症;是常见的医院内感染类型,是住院患者特别是老年患者出现并发症和死亡的重要原因。本部指南的撰写目的是对CDI患者的诊断、治疗及预防提供指导。指南内容包括CDI的诊断、治疗及预防三个方面,共20个临床问题。本指南的制定主要由指南专家组和指南工作组两个交叉合作的专家团队完成。其中指南专家组由与CDI诊断、治疗和感染控制相关的18个专业的3.名临床专家组成。本部指南采用国际公认的GRADE标准和流程制定,GRADE包括证据级别和推荐强度。GRADE系统将证据级别分为高、中、低和极低4个等级。制定推荐意见时,考虑了医学干预的利弊平衡、证据质量、价值观念与偏好,以及成本与资源耗费等因素,并将推荐意见分为强推荐和弱推荐(有条件推荐)两种。本部指南的应用对象为中国医师和患者。
