Phosphorylation/dephosphorylation is probably the most common and important reversible post-translational modification of proteins. Analyzing the functional effects of phosphorylation is helpful for understanding the biological functions of proteins. Identification of the phosphorylation sites of phosphorylated protein is a prerequisite for research on phosphorylation. In this work, an effective and simple method of identification of protein phosphorylation sites has been developed. Phosphopeptides were selectively enriched with immobilized metal affinity chromatography (IMAC) and subsequently chemically modified by 4-sulfophenyI isothiocyanate, and then the chemically modified phosphopeptides were sequenced with post-source decay (PSD) matrix-assisted laser desorption/ionization (MALDI) time-of-flight mass spectrometry for detecting phosphorylation sites. The charge of derivatization by 4-sulfophenyl isothiocyanate introduces a negative sulfonic acid group at the N-terminus of a peptide, and enables the selective detection of only a single series of C-terminal y-type ions. This chemically assisted method greatly simplifies the extremely complex pattern of PSD fragment ions and makes the PSD spectra more easier to be interpreted. The phosphorylation sites of a synthesized model phosphopeptide and human c-myc protein have been successfully identified by this method.

Detecting Lags in Nonlinear Models Using General Mutual Information

The general mutual information （GMI） and general conditional mutual information （GCMI） are considered to measure lag dependences in nonlinear time series. Both of the measures have the property of invariance with transform. The statistics based on GMI and GCMI are estimated using the correlation integral. Under the hypothesis of independent series, the estimators have Gaussian asymptotic distributions. Simulations applied to generated nonlinear series demonstrate that the methods appear to find frequently the correct lags.

Cloning and characterization of two putative seven-transmembrane receptor genes from cotton

Using rapid amplification of cDNA ends (RACE)-PCR,two full-length cDNAs encoding putative seven-transmembrane receptors (designated Gh7TMpRI and Gh7TMpR2) were cloned from cotton *** blot and an ApaL1 restriction site polymorphism analyses revealed that Gh7TMpRI was derived from the ancestral A diploid genome,while Gh7TMpR2 was from the D subgenome. Northern blot hybridization indicated that both Gh7TMpR1 and GhTTMpR2 were expressed preferentially in the elongation phase of fiber *** of the Gh7TMpRl proteins were located within the membrane structure and displayed a punctuate pattern of *** of Gh7TMpR1 in fission yeast disrupted the polar growth and caused the formation of rounded cells. These results suggest that GhT7MpRl may play a critical role in cotton fiber development,perhaps as a signaling receptor that is involved in controlling fiber elongation.

The Euclidean Steiner minimum tree problem is a classical NP-hard combinatorial optimization *** of the intrinsic characteristic of the hard computability,this problem cannot be solved accurately by efficient algorithms up to *** to the extensive applications in real world,it is quite important to find some heuristics for *** stochastic diffusion search algorithm is a newly population-based algorithm whose operating mechanism is quite different from ordinary intelligent algorithms,so this algorithm has its own advantage in solving some optimization *** paper has carefully studied the stochastic diffusion search algorithm and designed a cellular automata stochastic diffusion search algorithm for the Euclidean Steiner minimum tree problem which has low time *** results show that the proposed algorithm can find approving results in short time even for the large scale size,while exact algorithms need to cost several hours.

Multi-spectral image fusion method based on two channels non-separable wavelets

A construction method of two channels non-separable wavelets filter bank which dilation matrix is [1, 1; 1,-1] and its application in the fusion of multi-spectral image are presented. Many 4×4 filter banks are designed. The multi-spectral image fusion algorithm based on this kind of wavelet is proposed. Using this filter bank, multi-resolution wavelet decomposition of the intensity of multi-spectral image and panchromatic image is performed, and the two low-frequency components of the intensity and the panchromatic image are merged by using a tradeoff parameter. The experiment results show that this method is good in the preservation of spectral quality and high spatial resolution information. Its performance in preserving spectral quality and high spatial information is better than the fusion method based on DWFT and IHS. When the parameter t is closed to 1, the fused image can obtain rich spectral information from the original MS image. The amount of computation reduced to only half of the fusion method based on four channels wavelet transform.

Superperiodic Feature on Silicon-Sputtered Highly Oriented Pyrolytic Graphite

Superperiodic feature was observed by scanning tunneling microscopy (STM) on the surface of highly oriented pyrolytic graphite (HOPG) on which silicon was sputtered. The superlattice was analyzed by the moire pattern hypothesis, and the lattice constant is 7.03 nm. For the superlattice, the observed boundaries between the superlattice and the normal graphite areas were zigzag, which was in good agreement with the result predicted theoretically. In addition, the observed lattice constants varied slightly in the superperiodic feature area. This implies the role of intralayer strain in the formation of the observed superlattice on the graphite surface.

During vegetative development,higher plants continuously form new leaves in regular spatial and temporal *** with abnormal leaf developmental patterns not only provide a great insight into understanding the regulatorymechanism of plant architecture,but also enrich the ways to its modification by which crop yield could be ***,we reported the characterization of the rice leafy-head2 (lhd2) mutant that exhibits shortened plastochron,dwarfism,reduced tiller number,and failure of phase transition from vegetative to reproductive *** and histologicalstudy revealed that the rapid emergence of leaves in lhd2 was resulted from the rapid initiation ofleafprimordia whereasthe reduced tiller number was a consequence of the suppression of the tiller bud *** molecular and geneticanalysis showed that LHD2 encodes a putative RNA binding protein with 67% similarity to maize *** ofgenome-scale expression profiles between wild-type and lhd2 plants suggested that LHD2 may regulate rice shoot devel-opment through KNOXand hormone-related *** similar phenotypes caused by LHD2 mutation and the conservedexpression pattern of LHD2 indicated a conserved mechanism in controlling the temporal leaf initiation in grass.

为了解决传统K均值算法在处理大规模数据时的局限性,在近似K均值算法(AKM)基础之上,利用对聚类中心进行分类的思想,提出了快速近似K均值算法(FAKM).该算法舍去了在AKM聚类结果中只获得少数样本的聚类中心,并充分利用类内样本密集稳定的聚类中心,使得迭代过程中待聚类样本数和类别数逐步减少,达到了提高算法速度及精简聚类结果的目的.将FAKM算法运用于实际的图像检索系统中,实验结果表明,系统在检索准确率、检索时间和聚类时间方面都得到了很好的改善.
