Aim:To examine whether isoliquiritigenin(ISL)can attenuate myocardial ischemia-reperfusion(MI/R)injury in rats by inducing metallothionein(MT)through acti-vation of janus kinase 2(JAK 2)/signal transducers and activators of transcription3.STAT 3.***:The experimental model of MI/R in rats was gener-ated by 3. min of ischemia and reperfusion for 2 *** mRNA expression of MT,COX-2,and iNOS were measured by *** protein expressions of MT,JAK/STAT,extracellular signal-regulated kinase(ERK),and Akt were determinedby Western blotting in the absence or presence of a JAK kinase inhibitor,tyrphostinAG490(1.0 mg/kg,iv,1h before ischemia).Results:Pretreatment with ISL mark-edly decreased the severity of reperfusion-induced arrhythmias and myocardialinfarct *** the ISL 20 mg/kg group,the activities of lactate dehydrogenase(LDH)and creatinine phosphokinase(CPK)were reduced by 3..4% and 51.3.when compared with the vehicle *** JAK 2/STAT 3.phosphoryla-tion was accompanied by increased synthesis of MT but not of COX-2 or iNOS inISL-treated ***490 can significantly weaken ISL-induced cardioprotectionand prevent the increase of MT expression and JAK 2/STAT 3.***:ISL protected MI/R injury through activation of JAK 2/STAT 3.sig-nal transduction pathway,which might be involved in mediating the upregulationof MT expression.

Aim:To investigate the antinociceptive effect of adenosine agonist R-phenylisopropyl-adenosine（R-PIA）given to conscious rats by intracerebro-ventricular（ICV）and intrathecal（IT）,and identify the effect of R-PIA on minimumalveolar concentration（MAC）of halothane with pretreatment of A1receptor an-tagonist 8-cyclopentyl-1,3.dipropylxanthine（DPCPX）or K+channel blocker4-aminopyridine（4-AP）.Methods:Sprague-Dawley rats were implanted with 24-gauge stainless steel guide cannula using stereotaxic apparatus and ICV method,and an IT catheter（PE-10,8.5cm）was inserted into the lumbar subarachnoidspace,while the rats were under pentobarbital *** one week ofrecovery from surgery,rats were randomly assigned to one of the followingprotocols:MAC of halothane,or tail-flick *** measurements were per-formed after R-PIA（0.8-2.0μg）microinjection into ICV and IT with or withoutpretreatment of DPCPX or ***:Microinjection of adenosine agonist R-PIA in doses of 0.8-2.0μg into ICV and IT produced a significant dose-and time-dependent antinociceptive action as reflected by increasing latency times andICV administration of adenosine agonist R-PIA（0.8μg）reducing halothane anes-thetic requirements（by 29%）.The antinociception and reducing halothane re-quirements effected by adenosine agonist R-PIA was abolished by DPCPX ***:ICV and IT administration of adenosine agonist R-PIA pro-duced an antinociceptive effect in a dose-dependent manner and decreased hal-othane MAC with painful stimulation through activation of A1receptor subtype,and the underlying mechanism involves K+channel activation.

Objective The study investigated the burden of smear-positive pulmonary TB and itsinfectivity using DALY(disability-adjusted life year) as an indicator. Methods An assumedcohort of 2 000 cases was set up based on the age-specific incidence of 794 newly registeredsmear-positive cases in Beijing in 1994. Prognostic trees and model diagrams of infectivityunder natural history and DOTS(directly observed treatment, short-course) strategy wereestablished according to the epidemiological evidence. Results The results showed that 29. 6%of DALYs would be neglected if the burden caused by the infectivity was not *** DOTS strategy may reduce 97.3. of the number of potential cases infected,92.9% of DALYs related to TB-patients themselves, and 99.9% of DALYs caused by TB’sinfectivity as well.

The aim of this study was to establish and apply a real-time quantitative reverse transcription-polymerase chain reaction （RT-PCR） for rubella virus （RV） ***,the primer and TaqMan probeconcentrations,as well as reaction temperatures were optimized to establish an efficient real-time quantitativeRT-PCR assay for RV ***,an RV-specific PCR amplicon was made as an external standard toestimate the linearity,amplification efficiency,analytical sensitivity and reproducibility of the real timequantitative ***,the assay was applied to quantify RV RNA in clinical samples for rubella *** RV-specific PCR amplicon was prepared for evaluation of the assay at 503.bp,and its original concentrationwas 2.75×10～9 copies/μ*** real time quantitative assay was shown to have good linearity （R2=0.9920）,highamplification efficiency （E=1.91）,high sensitivity （275 copies/ml）,and high reproducibility （variation coefficientrange,from 1.25% to 3.58%）.Compared with the gold standard,the specificity and sensitivity of the assayin clinical samples was 96.4% and 86.4%,***,the established quantitative RT-PCRmethod is a simple,rapid,less-labored,quantitative,highly specific and sensitive assay for RV RNA.

AIM: To study the effect of leflunomide on CCl4-induced hepatic fibrosis in rats. METHODS: Hepatic fibrosis was induced by subcutaneous injection with 50 % CCl4 in Sprague-Dawley rats. The amount of CCl4 administered was 1 mg/kg. The alanine aminotransferase (ALT), aspartate aminotransferase (AST), nitric oxide (NO) levels in plasma and hydroxyproline (Hyp) contents in liver tissue were assayed by spectrophotometry. The hyaluronic acid (HA) and procollagen III (PC III) were assessed by radioimmunoassay. The transforming growth factor-β1 (TGF-β1) in serum was determined by ELISA. The nuclear factor-kappa B (NF-κB) in liver tissue was examined by immunohistochemistry. Liver samples collected after 12 weeks of CCl4 treatment were stained with hematoxy- lin and eosin. RESULTS: Leflunomide (1, 3. and 9 mg/kg) significantly decreased indices of liver and spleen, the serum transaminase (AST, ALT) activities, HA and PC III levels, and Hyp contents in liver tissue in rats of hepatic fibrosis. Histopathological examination showed leflunomide had inhibitory effect on fibrogenesis and formation of pseudolobulus. Furthermore, leflunomide significantly inhibited NF-κB expression in liver tissue, and reduced elevated serum TGF-β1 and NO levels in rats of hepatic fibrosis. CONCLUSION: Leflunomide showed inhibitory action on hepatic fibrosis induced by CCl4 in rats.

The mip gene of Legionella pneumophila and the ctxB gene of Vibrio cholerae were amplifiedby PCR *** amplified cDNA was ligated to the pcDNA3.1(+)*** recombinant plasmidspcDNA3.1-mip and pcDNA3.1-ctxB were identified by restriction analysis and PCR,and further confirmedby sequencing ***3.3.cells were transfected with pcDNA3.1-mip and pcDNA3.1-ctxB accordingto the Lipofection *** and stable products of the co-expression of the mip gene and ctxB genewere detected by immunofluorescence and Western *** results showed that NIH3.3.cells weresuccessfully transfected,and that the transiently and stably co-expressed products can be detected in thetransfected *** detect the humoral and cellular immune response in immunized mice induced by the co-immunization of the mip and ctxB genes,female BALB/c mice were immunized intramuscularly with pcDNA3.1-mip and *** results showed that the specific antibody titer and the cytotoxic T-lymphocyteresponse for pcDNA3.1-mip immunization and co-immunization were increased compared with that ofpcDNA3.1(+) ***,the specific antibody titer and cytotoxic T-lymphocyte responsefor co-immunization were increased compared with that of pcDNA3.1-mip *** analysisusing one-way analysis of variance(ANOVA)showed that there was a significant difference between thegroups(P<0.01).The results indicated that the ctxB gene enhanced the humoral and cellular immune responseto the mip gene *** findings provide experimental evidence to support the development ofthe *** DNA vaccine.

In ultrasound elastography, the axial strain distribution within biological tissues is calculated as the numerical derivative (differentiation) of the estimated axial displacement field. Unfortunately, the numerical derivative is unstable because it can greatly amplify the noises, especially at high frequencies. This work focuses on the axial strain calculation from the estimated axial displacements using wavelet transforms (WTs), including continuous wavelet transforms (CWTs) and discrete wavelet transforms (DWTs). The feasibility of the WT-based method using the quadratic spline function is verified by computer simulations and some phantom data. Results indicate that the WT-based method can effectively reduce the noise amplification in axial strain calculation.

Aim:To study the effects of long-term,low-dose sex hormone replacement therapy(HRT)on the volume and biochemical changes of the hippocampus in postmeno-pausal women carrying apolipoprotein E(apoE)gene ε3.or ε***:Eighty-three postmenopausal women who had used a low dose of HRT for over 4 yearswere selected as the HRT group,and 99 postmenopausal women with matchedage and education were enrolled as the control *** alleles were analyzedby *** resonance imaging was performed to determine the volume ofthe brain *** magnetic resonance spectroscopy was used todetect the biochemical changes in the anterior cingulate cortex and hippocampusin apoE ε4 and ε3.*** common cognitive tests were used to make anoverall evaluation of cognitive ***:Analysis with the apoE ε4carriers showed that the volume of the hippocampus of the control group weresignificantly lower than those of the HRT *** biochemical analysis showedthat there was an increase of N-acetylaspartate(NAA)/total creatine(tCr)and adecrease of myoinositol(mI)/tCr in the hippocampus of apoE ε4 carriers in theHRT group,compared with the control *** the apoE ε3.carriers,the leastsquares means(LSMEAN)of the HRT group was higher than that of the ***:This study showed that long-term,low dose HRT might bebeneficial for reducing the risk of AD development in vulnerable ***,HRT could increase the LSMEAN of apoE ε3.carriers.
