Osteoarthritis(OA)is a major clinical challenge,and effective disease-modifying drugs for OA are still lacking due to the complicated pathology and scattered treatment *** early treatments are urgently needed to prevent OA *** excessive amount of transforming growth factorβ(TGFβ)is one of the major causes of synovial fibrosis and subchondral bone sclerosis,and such pathogenic changes in early OA precede cartilage *** we report a novel strategy of intra-articular sustained-release of pirfenidone(PFD),a clinically-approved TGFβinhibitor,to achieve disease-modifying effects on early OA *** found that PFD effectively restored the mineralization in the presence of excessive amount of TGFβ1(as those levels found in patients’synovial fluid).A monthly injection strategy was then designed of using poly lactic-co-glycolic acid(PLGA)microparticles and hyaluronic acid(HA)solution to enable a sustained release of PFD(the“PLGA-PFD+HA”strategy).This strategy effectively regulated OA progression in destabilization of the medial meniscus(DMM)-induced OA mice model,including preventing subchondral bone loss in early OA and subchondral bone sclerosis in late OA,and reduced synovitis and pain with cartilage preservation *** finding suggests the promising clinical application of PFD as a novel disease-modifying OA drug.

Training gastroenterology fellows to perform gastric polypectomy using a novel ex vivo model

AIM:To evaluate the effect of hands-on training of gastroenterology fellows in gastric polypectomy using an ex vivo ***:Eight gastroenterology fellows at Mackay Memorial Hospital,Taipei were evaluated in gastricpolypectomy techniques using a pig stomach with artificial polyps created by a rubber band ligation *** performance of four second year(year-2)fellows who had undergone one year of clinical training was compared with that of four f irst year(year-1)fellows both before and after a 4-h workshop using the ex vivo *** workshop allowed for hands-on train-ing in the removal of multiple artif icial polyps and the placement of hemoclips at the excision *** included observation of technical skills,procedure time,and the fellows' conf idence ***:One week after the workshop,the year-1 fellows were re-evaluated and had significantly im-proved mean performance scores(from 17.9 ± 1.8 to 22.5 ± 0.7),conf idence scale(from 4.5 ± 1.0 to 7.8 ± 0.5)and procedure time(from 615.0 ± 57.4 s to 357.5 ± 85.0 s)compared with their baseline *** 4 h of training using the ex vivo simulator,the skills of the year-1 fellows were statistically similar to those of the year-2 ***:Use of this ex vivo simulator significantly improved the endoscopic gastric polypectomy skills of gastroenterology fellows who had not had previous clinical training in gastric polypectomy.

Enhancement of motor functional recovery in thoracic spinal cord injury: voluntary wheel running versus forced treadmill exercise

Spinal cord injury necessitates effective rehabilitation strategies, with exercise therapies showing promise in promoting recovery. This study investigated the impact of rehabilitation exercise on functional recovery and morphological changes following thoracic contusive spinal cord injury. After a 7-day recovery period after spinal cord injury, mice were assigned to either a trained group(10 weeks of voluntary running wheel or forced treadmill exercise) or an untrained group. Bi-weekly assessments revealed that the exercise-trained group, particularly the voluntary wheel exercise subgroup, displayed significantly improved locomotor recovery, more plasticity of dopaminergic and serotonin modulation compared with the untrained group. Additionally, exercise interventions led to gait pattern restoration and enhanced transcranial magnetic motor-evoked potentials. Despite consistent injury areas across groups, exercise training promoted terminal innervation of descending axons. In summary, voluntary wheel exercise shows promise for enhancing outcomes after thoracic contusive spinal cord injury, emphasizing the role of exercise modality in promoting recovery and morphological changes in spinal cord injuries. Our findings will influence future strategies for rehabilitation exercises, restoring functional movement after spinal cord injury.

Double-lined M dwarf eclipsing binaries from Catalina Sky Survey and LAMOST

Eclipsing binaries provide a unique opportunity to determine fundamental stellar properties. In the era of wide-field cameras and all-sky imaging surveys, thousands of eclipsing binaries have been reported through light curve classification, yet their basic properties remain unexplored due to the ex- tensive efforts needed to follow them up spectroscopically. In this paper we investigate three M2-M3 type double-lined eclipsing binaries discovered by cross-matching eclipsing binaries from the Catalina Sky Survey with spectroscopically classified M dwarfs from the Large Sky Area Multi-Object Fiber Spectroscopic Telescope survey data release one and two. Because these three M dwarf binaries are faint, we further acquire radial velocity measurements using GMOS on the Gemini North telescope with R ~4000, enabling us to determine the mass and radius of individual stellar components. By jointly fitting the light and radial velocity curves of these systems, we derive the mass and radius of the primary and secondary components of these three systems, in the range between 0.28-0.42 MQ and 0.29-0.67 R, respectively. Future observations with a high resolution spectrograph will help us pin down the uncertainties in their stellar parameters, and render these systems benchmarks to study M dwarfs, providing inputs to improving stellar models in the low mass regime, or establishing an empirical mass-radius relation for M dwarf stars.

Parametric studies on relationships between flutter derivatives of slender bridge(Ⅱ)

Based on curve fitting of coefficients of three component forces of the Messina Straits Bridge, and the previously proposed semi-analytical expressions of flutter derivatives of flexible structure, the change of flutter derivatives of slender bridge cross-section with respect to its aerodynamic center, rotational speed and angle variation is studied using a parametric method. The calculated results are compared with the measured ones, and expressions of flutter derivatives of the Messina Straits Bridge are derived. The intrinsic relationships existing in flutter derivatives are validated again. It is shown that the influence of the rotational speed on flutter derivatives is not negligible. Therefore, it provides an additional semi-analytical approach for analyzing flutter derivatives of the bridge with streamlined cross-section to get its aerodynamic information.

Modeling of turbulent,isothermal and cryogenic cavitation under attached conditions

Cavitation is often triggered when the fluid pres- sure is lower than the vapor pressure at a local thermo- dynamic state. The present article reviews recent progress made toward developing modeling and computational strat- egies for cavitation predictions under both isothermal and cryogenic conditions, with an emphasis on the attached cav- ity. The review considers alternative cavitation models along Reynolds-averaged Navier-Stokes and very lager eddy simu- lation turbulence approaches to ensure that the computational tools can handle flows of engineering interests. Observing the substantial uncertainties associated with both modeling and experimental information, surrogate modeling strategies are reviewed to assess the implications and relative impor- tance of the various modeling and materials parameters. The exchange between static and dynamic pressures under the influence of the viscous effects can have a noticeable impact on the effective shape of a solid object, which can impact the cavitation structure. The thermal effect with respect to evaporation and condensation dynamics is examined to shed light on the fluid physics associated with cryogenic cav- itation. The surrogate modeling techniques are highlighted in the context of modeling sensitivity assessment. Keywords

SECOND ORDER APPROXIMATION SOLUTION OF NONLINEAR LARGE DEFLECTION PROBLEMS OF YONGJIANG RAILWAY BRIDGE IN NINGBO

The solution and computational aspects on nonlinear deflection of Yongjiang Railway Bridge in Ningbo were investigated. An approximate iteration algorithm on nonlinear governing equation was presented, and the obtained results show that, if altitude difference and span of the riverbanks are taken as 5 meters and 100 meters, respectively, the maximum gradient in the middle of the bridge exceeds 5%, much larger than maximum allowance gradient in railway design code. Therefore, a new solution scheme for decreasing gradient of the bridge is put forward, that is, the altitude difference between two riverbanks can be decreased to about 1/10 of the initial magnitude by building roadbeds with 0.5% gradient and 1 kilometer length at two riverbanks. As a direct result, the deflection gradient of the railway bridge is much reduced and the value is between 0.5% similar to 0.6%.

Rtecently a lot of works have been investigating to find the tenuous groups,i.e.,groups with few social interactions and weak relationships among members,for reviewer selection and psycho-educational group ***,the metrics(e.g.,k-triangle,k-line,and k-tenuity)used to measure the tenuity,require a suitable k value to be specified which is difficult for users without background ***,in this paper we formulate the most tenuous group(MTG)query in terms of the group distance and average group distance of a group measuring the tenuity to eliminate the influence of parameter k on the tenuity of the *** address the MTG problem,we first propose an exact algorithm,namely MTGVDIS,which takes priority to selecting those vertices whose vertex distance is large,to generate the result group,and also utilizes effective filtering and pruning *** MTGVDIS is not fast enough,we design an efficient exact algorithm,called MTG-VDGE,which exploits the degree metric to sort the vertexes and proposes a new combination order,namely degree and reverse based branch and bound(DRBB).MTG-VDGE gives priority to those vertices with small *** a large p,we further develop an approximation algorithm,namely MTG-VDLT,which discards candidate attendees with high degree to reduce the number of vertices to be *** experimental results on real datasets manifest that the proposed algorithms outperform existing approaches on both efficiency and group tenuity.

Development of seismic fragility curves for low-rise masonry infilled reinforced concrete buildings by a coefficient-based method

This study presents a seismic fragility analysis and ultimate spectral displacement assessment of regular low-rise masonry infilled （MI） reinforced concrete （RC） buildings using a coefficient-based method. The coefficient-based method does not require a complicated finite element analysis; instead, it is a simplifed procedure for assessing the spectral acceleration and displacement of buildings subjected to earthquakes. A regression analysis was first performed to obtain the best-fitting equations for the inter-story drift ratio （IDR） and period shift factor of low-rise MI RC buildings in response to the peak ground acceleration of earthquakes using published results obtained from shaking table tests. Both spectral acceleration- and spectral displacement-based fragility curves under various damage states （in terms of IDR） were then constructed using the coefficient-based method. Finally, the spectral displacements of low-rise MI RC buildings at the ultimate （or near- collapse） state obtained from this paper and the literature were compared. The simulation results indicate that the fragility curves obtained from this study and other previous work correspond well. Furthermore, most of the spectral displacements of low-rise MI RC buildings at the ultimate state from the literature fall within the bounded spectral displacements predicted by the coefficient-based method.
