Injury-induced rapid activation of MAPK signaling in dechorionated eggs and larvae of the silkworm Bombyx mori

Previous study showed that diapause in Bombyx mori eggs can be ter- minated by dechorionation and that activation in the mitogen-activated protein kinase （MAPK）/extracellular signal-regulated kinase （ERK） in dechorionated cultured eggs is involved in diapause termination. In the present study, the possible mechanism underlying activation of ERK upon dechorionation was further investigated. Results showed that me- chanical injury of diapause eggs without medium incubation also resulted in rapid increase in the phospho-ERK levels and that injury increased the phospho-ERK levels at different stages of both diapause eggs and eggs in which diapause initiation was prevented by HC1. Effects of anaerobiosis on dechorionation-stimulated phospho-ERK levels showed that the mechanical injury itself but not the dramatic increase in oxygen uptake upon injury is in- volved in a rapid activation of ERK. Chemical anaerobiosis on dechorionation-stimulated phospho-ERK levels and the in vivo effect of anaerobiosis showed that the supply of oxygen also plays a role in ERK signaling. In addition, injury induced the phosphory- lation of c-jun N-terminal kinases （JNKs） and p38 kinase, components of two parallel MAPK pathways. A kinase assay showed a dramatic increase in .INK kinase activity in egg lysates upon injury. When newly hatched first instar larvae were injured, an increase in the phospho-ERK levels similar to that in dechorionated eggs was observed. From the results, we hypothesize that the injury-induced rapid activation of MAPK signaling, which serves as a natural signal for embryonic development, is related to diapause termination in dechorionated eggs.

Liu-stain quick cytodiagnosis of ultrasound-guided fine needle aspiration in diagnosis of liver tumors

AIM： To combine ultrasound-guided fine-needle aspiration （US-FNA） and Liu （Riu） stain to make a quick study on liver tumor lesions. METHODS： Two hundred and twenty-eight aspirations from 232 patients were completely studied. The operator himself made the quick cytodiagnosis of US-FNA smear stained by Liu method within thirty minutes. The US-FNA specimen was also sent to the pathological department for cytological study and cellblock histology. The result of our Uu-stain quick cytodiagnosis in each patient was confirmed by the final cytopathological diagnosis from pathological report. RESULTS： Among 228 samples, the quick cytodiagnosis revealed 146 malignancies, 81 benign lesions and one inadequate specimen. Cytopathological diagnosis from the pathological department revealed 150 malignancies, and 78 benign lesions. Four well-differentiated hepatocellular carcinomas （HCCs） were under-diagnosed by quick cytodiagnosis as benign and 3 benign lesions were over-diagnosed as well-differentiated HCCs. Compared with cytopathological diagnosis, quick cytodiagnosis correctly diagnosed 143 malignancies and 77 benign lesions. Except for the one inadequate specimen in quick cytodiagnosis, the accuracy of quick cytodiagnosis was 96.9% （220/227）, and its sensitivity, specificity and positive and negative predictive values were 97.9%, 95.1%, 97.3% and 96.3%, respectively. CONCLUSION： Liu-stain quick cytodiagnosis is a fast, convenient, safe and effective method for hepatologists in clinic practice to diagnose liver tumor. In few cases of well-differentiated HCC, Liu-stain quick cytodiagnosis has its limitation.

Insulin resistance is associated with hepatocellular carcinoma in chronic hepatitis C infection

AIM:To elucidate the role of insulin resistance(IR) and serum adiponectin level in hepatocellular carcinoma(HCC) associated with chronic hepatitis ***:Clinical and biochemical characteristics were collected from 165 consecutive patients with newly diagnosed *** model assessment of IR(HOMA-IR) and serum adiponectin level were investigated in 188 patients with different stages of hepatitis C virus(HCV) ***:Among HCC patients,type 2 diabetics(DM) was more prevalent in HCV subjects(35.6%,n = 59) compared to hepatitis B virus(HBV;12.7%,n = 63) or non-HBV,non-HCV cases(7.1%,n = 28).In patients with chronic hepatitis C,HCC subjects had higher blood sugar(P < 0.001),insulin level(P = 0.003) and HOMAIR(P = 0.018) than those with chronic hepatitis and advanced ***,male sex and body mass index were significantly associated with serum adiponectin level,whereas HOMA-IR was *** on stepwise logistic regression analysis,age(OR:1.124,P < 0.001),serum insulin level(OR:1.585,P < 0.001),HOMA-IR(OR:0.495,P = 0.001),DM(OR:11.601,P = 0.002) and male sex(OR:3.877,P = 0.016) were independently associated with *** result was similar even if the diabetic subjects were excluded for ***:Insulin resistance measured by HOMAIR,regardless of the presence of diabetes,is significantly associated with HCC development in patients with chronic HCV infection.

In this paper, a forecasting system of electric price is proposed to predict the short-term electric prices for avoiding the risk due to the electricity price volatilityBased on the Back-propagation Neural Network（BPN） and Orthogonal Experimental Design（OED）, a New Artificial Neural Network Approach（NANNA） is constructed in the searching processThe data cluster, including Locational Marginal Price（LMP）, system load, temperature, line-flow, are first collected and embedded in the Excel DatabaseIn order to get a better solution, the OED is used to automatically regulate the parameters during the NANNA training processLinking the NANNA and Excel database, the NANNA retrieved the input data from Excel Database to perform and analyze the efficiency and accuracy of the predicting system until the forecasting system is convergentSimulation results will provide the participants to obtain the maximal profits and raise its ability of market’s competition in a price volatility environment.

CO capture from the indoor air and concentrated by cyclic adsorption-desorption processes are studied.Y-type zeolite was synthesized with a Si and A1 molar ratio of 17 referred to as *** synthesized Y-type zeolite was impregnated with amines used to capture CO for maintaining good indoor air quality(IAQ).Three kinds of amines including monoethanol amine(MEA),isopropanol amine(IPA),and tetraethylenepent amine(TEPA)were selected to study the performance of COadsorption capacity at a fixed-bed in an 80-L air chamber These amies-loaded Y17 were characterized by Scanning Electron Microscope(SEM),X-ray Diffraction(XRD),Infrared Spectroscopy(IR)and Thermogravimetric analyzer(TGA)to examine their textural properties before and after *** CO adsorption capacities of Y17-modified was substantially higher than that of original Y17,and the highest capacity can reach up to 158 mg/*** study indicated that the order of adsorption capacity was TEPA>MEA>***-order kinetic model was fitted to Y17-modified adsorption behavior very *** rate constant(k)of first-order adsorption model can be further used to design and apply in CO control *** the result of this study,amines functionalized Y17 are a great potential adsorbent fro removing indoor CO.

The moving particle semi-implicit（MPS）method has demonstrated its usefulness in practical engineering *** it has wide applicability,it is still hard to predict the pressure precisely using the MPS method.A pressure-convection particle method based on the MPS method is proposed to overcome this *** improved performance of this new method is validated with computational and measured *** approach is also applied to compute the problem of sloshing associated with floating body motion in *** pressure-convection MPS method demonstrated its capability to improve the prediction of pressure.

Microbial diversity of soil bacteria in agricultural field contaminated with heavy metals

In this study we evaluated the bacterial diversity in a soil sample from a site next to a chemical industrial factory previously contaminated with heavy metals. Analysis of 16S rDNA sequences amplified from DNA directly extracted from the soil revealed 17 different bacterial types （genera and/or species）. They included Polyangium spp., Sphingomonas spp., Variovorax spp., Hafina spp., Clostridia, Acidobacteria, the enterics and some uncultured strains. Microbes able to tolerate high concentrations of cadmium （500μmol/L and above） were also isolated from the soil. These isolates included strains of Acinetobacter （strain CD06）, Enterobacter sp. （strains CD01, CD03, CD04 and CD08）（similar strains also identified in culture-independent approach） and a strain of Stenotrophomonas sp. The results indicated that the species identified from direct analysis of 16S rDNA of the soil can be quite different from those strains obtained from enrichment cultures and the microbial activities for heavy metal resistance might be more appropriately addressed by the actual isolates.
