This paper reports the development and validation of an assay for ifenprodil based on liquid chromatography–tandem mass spectrometry(LC–MS/MS)and its application to a pharmacokinetic study involving single and multiple intravenous infusions to healthy Chinese *** sample preparation of plasma by liquid–liquid extraction with ethyl acetate,the analyte and internal standard,urapidil,were separated by reversed phase chromatography in a run time of 4 min and detected by positive ion electrospray ionization followed by multiple reaction monitoring of the precursor-to-product ion transitions at m/z 326.2-308.1 for ifenprodil and m/z 388.4-205.3 for *** assay was linear in the concentration range 0.2–50.0 ng/mL with recovery 476.4%.In the pharmacokinetic study of single intravenous infusions of 5,10 and 15 mg ifenprodil,peak plasma concentrations and areas under the plasma concentration–time curve were both linearly related to *** the pharmacokinetic study of multiple once daily intravenous infusions of 10 mg ifenprodil for 7 days,pharmacokinetic parameters were similar to those after the single dose showing that ifenprodil does not accumulate on repeated administration.

Molecular Epidemiology of Klebsiella pneumoniae from Clinical Bovine Mastitis in Northern Area of China,2018–2019

Due to the different imaging principle of infrared image and visible image, the information characteristics of the two images are different. Fusion technology is the organic fusion of images from two different sensors, with complementary information. In this paper, a novel algorithm is proposed for the fusion of infrared and visible images. The algorithm are firstly performs HIS transform on the infrared image and the visible image to obtain two sets of intensity（I）, hue（H） and saturation（S） components. Then, after the wavelet transform of the I-components, a simple fusion algorithm based on spatial domain is adopted for low frequency sub-band, and the high-frequency sub-bands adopt the fusion algorithm based on window-based average gradient. Finally, the fused I-component was wavelet reconstructed and transformed by the inverse HIS transform with S-component and H-component of visible image to obtain a fused image. Experiments show that the novel fusion algorithm of infrared and visible image proposed in this paper can preserve the background information and texture information of the visible image better when the object is highlighted in the infrared image, the fused image is clearer, and the fusion effect is better.

Klebsiella pneumoniae(***,KpⅠ)is a predominate inducement of bovine mastitis,which is associated with high mortality and milk yield ***,data is lacking on the molecular characteristics of bovine ***,limiting the risk assessment of its transmission through the food ***,we investigated the prevalence of *** in 6301 clinical mastitis(CM)milk samples from dairy cattle in northern area of *** total,183 *** isolates were recovered,with detection rates of 3.0% and 2.8% in 2018 and 2019,*** human clinical ***,all CM *** isolates belonged to one of three phylogroups:KpⅠ(n=143),*** ***(KpⅡ-B)(n=37),and Klebsiella variicola(KpⅢ)(n=3).We detected the extendedspectrum β-lactamase-encoding genes bla_(SHV-2a),blac_(CTX-M-14),and bla_(CTX-M-15),as well as clpC,lpfA,lacI,lacZ,lacY,and the fecABDEIR operon in the KpⅠ isolates,which may contribute to their pathogenicity and host adaptability in *** high prevalence of KpⅠ in dairy farms may be problematic,as it showed relatively higher rates of antibiotic resistance and virulence gene carriage than the KpⅡ-B and KpⅢ ***,we observed distinct differences in population structure between CM-and human infection-associated KpⅠ isolates,with the genes associated with invasive infection in humans rarely being observed in bovine isolates,indicating that few CM-associated *** isolates pose a threat to human ***,bovine KpⅡ-B isolates shared a high level of nucleotide sequence identity with isolates from human infections and frequently carried the nitrogen-fixation gene nif,suggesting an association between KpⅡ-B isolates from cattle and humans,and plant-derived bacteria.

A sensitive and specific liquid-chromatography tandem mass spectrometry(LC-MS/MS)assay has been developed and validated for the simultaneous quantification of ivabradine and its active metabolite N-desmethylivabradine in human plasma and *** assay employed a single liquid–liquid extraction of the analytes from plasma and urine samples,and diazepam was used as internal standard(IS).The chromatographic separation was achieved on a Diamonsil C18 column(150 mm4.6 mm,5 mm,Dikma)using a mixture of methanol and aqueous 5 mM ammonium acetate buffer containing 0.2%formic acid(80:20,v/v)as mobile *** assay for ivabradine and N-desmethylivabradine in plasma showed good linearity(r≥0.99)over the ranges 0.1013–101.3 ng/mL and 0.085–25.5 ng/mL,*** assay for ivabradine and N-desmethylivabradine in urine showed good linearity(r≥0.99)over the ranges 10.13–6078 ng/mL and 8.5–850 ng/mL,*** intra-and inter-day accuracy and precision values were found to be within the assay variability limits(RSD<15%)in accordance with FDA *** methods were successfully used for evaluating the pharmacokinetic properties of ivabradine and N-desmethylivabradine in human plasma and urine in Chinese healthy volunteers.

Brazilin is an important constituent of Caesalpinia sappan L., and has several bioactivities. In this study, a rapid and sensitive analytical method based on high-performance liquid chromatography–tandem mass spectrometry （HPLC–MS/MS） has been developed for the determination of brazilin in rat plasma, urine, feces and tissues （brain, heart, liver, lung and kidney and spleen）. Biological samples were processed with ethyl acetate containing 5% formic acid extraction, and salicylic acid （SA） was chosen as the internal standard （IS）. The separation of brazilin was achieved on an Inspire C 18 column （4.6 mm×150 mm, 5 μm） with a mobile phase consisting of methanol / 5 mM ammonium acetate （80:20, v/v）. The MS/MS detection was carried out by monitoring the fragmentation of m/z 285.1→163.0 for brazilin and m/z 137.1→93.1 for SA on a triple quadrupole mass spectrometer. The total run time was only 5.0 min. The analyte showed good linearity over a wide concentration range （R2 ＞ 0.995） and its lower limit of quantification was 2 ng/mL. The accuracy and precision ranged from 97.1 to 103.3% and 1.7 to 9.1%, respectively. Recoveries （78.9–93.8%） and matrix effects （81.0-97.8%） were satisfactory in all the biological matrices examined. Stability studies （86.4–99.8%） showed that brazilin was stable during the assay procedure and long-term storage. The assay was successfully applied to plasma pharmacokinetics, tissue distribution and excretion study of rats. The pharmacokinetic parameters, such as half-life, mean residence time, maximum concentration were determined. These preclinical data of brazilin would be useful for the clinical reference.

采用不同的方法及改性剂处理秸秆纤维,并对聚丁二酸丁二醇酯(PBS)进行共混改性,利用热压工艺得到了秸秆纤维/PBS复合材料。研究了水煮和微波处理工艺对秸秆纤维提取量的影响;研究了苯甲酸等改性剂的种类、添加量对秸秆纤维及秸秆纤维/PBS复合材料性能的影响。采用扫描电子显微镜观察了秸秆纤维处理前后的表面形貌。研究结果表明:水煮工艺为煮沸30 min 2次,微波处理工艺为微波连续处理15 min时为最佳工艺;对比2种工艺,微波连续处理15 min得到的秸秆纤维比水煮处理后的更为疏松,比表面积更大,同时得到的秸秆纤维/PBS复合材料综合性能良好,并且在硬脂酸质量分数约为3.5%时复合材料的力学性能最优。

江西宜春是我国华南地区重要的稀有金属矿产地之一,分布有全国最大的锂和钽矿生产基地,而雅山复式岩体是华南地区著名的稀有金属花岗岩。本文通过对前人岩石地球化学和岩石矿物学数据的梳理,发现锂及其他稀有元素在黄玉锂云母花岗岩中高度富集,具有以下规律:黑磷云母-白云母花岗岩→锂云母钠长花岗岩→黄玉锂云母花岗岩,岩浆分异程度逐步增加且逐渐从熔体过渡到熔体-流体共存体系,全岩SiO_(2)含量、Nb/Ta和Zr/Hf比值、A/CNK和δEu值发生规律性变化;Ta、Nb等稀有矿物在矿床中呈浸染状均匀分布而不是流体成因的脉状或网脉状,且被主要造岩矿物所包裹,并见有雪球结构;各阶段的云母具有岩浆结晶作用形成的特征,如呈片状产出,边界平直,有锯齿状、裂片等。综合分析,我们认为:(1)在经过壳幔岩浆组合的过程中使得华南变质基底部分熔融形成初始岩浆,初始岩浆经历了原地高度结晶分异演化过程;(2)富含稀有金属的岩浆热液经过高度结晶分异过程,使得锂、铌、钽等稀有金属元素愈来愈富集于残余熔体中,最终形成稀有金属花岗岩;(3)伟晶岩壳构成岩浆房与外界的隔绝屏障,使F、B、Li、P、H_(2)O等易挥发组分无法逃逸。由于上覆伟晶岩壳形成的封闭体系和下伏幔源岩浆提供的充足热量,使得岩浆房在热驱动机制下长期保持原地分异状态,最终形成雅山锂铌钽矿床。
