NLRP3 activation in macrophages promotes acute intestinal injury in neonatal necrotizing enterocolitis

Background Macrophages are involved in various immune inflammatory disease *** study aimed to investigate the role and mechanism of macrophages in regulating acute intestinal injury in neonatal necrotizing enterocolitis(NEC).Methods CD68,nucleotide-binding oligomerization domain,leucine-rich repeat,and pyrin domain-containing 3(NLRP3),cysteine aspartate-specific protease-1(caspase-1),and interleukin-1β(IL-1β)in paraffin sections of intestinal tissues from NEC and control patients were detected with immunohistochemistry,immunofluorescence,and western *** pet milk,hypoxia and cold stimulation were used to establish a mouse(wild type and Nlrp3^(-/-))model of *** mouse macrophage(RAW 264.7)and rat intestinal epithelial cell-6 lines were also cultured followed by various ***,intestinal epithelial cell injuries,and IL-1β release were *** Compared to the gut“healthy”patients,the intestinal lamina propria of NEC patients had high macrophage infiltration and high NLRP3,caspase-1,and IL-1β ***,in vivo,the survival rate of Nlrp3^(-/-)NEC mice was dramatically improved,the proportion of intestinal macrophages was reduced,and intestinal injury was decreased compared to those of wild-type NEC ***3,caspase-1,and IL-1β derived from macrophages or supernatant from cocultures of macrophages and intestinal epithelial cells also caused intestinal epithelial cell *** Macrophage activation may be essential for NEC ***3/caspase-1/IL-1β cellular signals derived from macrophages may be the underlying mechanism of NEC development,and all these may be therapeutic targets for developing treatments for NEC.

Based on the fact that it is challenging for the polymer flexible circuit substrates to meet the requirements of serving in high-temperature environments,this work proposed the idea of using printable ceramic fiber paper as a high-temperature flexible circuit substrate.A ceramic fiber paper with all ceramic components had been developed via electrospinning,solving the problems of low strength and severe strength drop at high temperatures of traditional ceramic fiber *** tensile strength of the prepared ceramic fiber paper is 2.63 MPa,and the reliable service temperature is 1200℃.Its bulk density is about 1.5 times that of traditional ceramic fiber *** can be printed with patterns by commercial inkjet printers like ordinary printing paper and has excellent *** feasibility of ceramic fiber paper as a flexible circuit substrate was verified by constructing a simple *** the fiber paper is significantly bent,the circuit still forms a complete path,which proves that it has a strong application potential for high-temperature flexible circuit substrate and is expected to promote the development of flexible electronic devices serving at extreme high-temperature environments.

Mg-doped,carbon-coated,and prelithiated SiO_(x) as anode materials with improved initial Coulombic efficiency for lithium-ion batteries

Silicon suboxide(SiO_(x),x≈1)is promising in serving as an anode material for lithium-ion batteries with high capacity,but it has a low initial Coulombic efficiency(ICE)due to the irreversible formation of lithium silicates during the first *** this work,we modify SiO_(x) by solid-phase Mg doping reaction using low-cost Mg powder as a reducing *** show that Mg reduces SiO_(2) in SiO_(x) to Si and forms MgSiO_(3) or Mg_(2)SiO_(4).The MgSiO_(3) or Mg_(2)SiO_(4) are mainly distributed on the surface of SiO_(x),which suppresses the irreversible lithium-ion loss and enhances the ICE of SiO_(x).However,the formation of MgSiO_(3) or Mg_(2)SiO_(4) also sacrifices the capacity of SiO_(x).Therefore,by controlling the reaction process between Mg and SiO_(x),we can tune the phase composition,proportion,and morphology of the Mg-doped SiO_(x) and manipulate the *** obtain samples with a capacity of 1226 mAh g^(–1) and an ICE of 84.12%,which show significant improvement over carbon-coated SiO_(x) without Mg *** the synergistical modification of both Mg doping and prelithiation,the capacity of SiO_(x) is further increased to 1477 mAh g^(–1) with a minimal compromise in the ICE(83.77%).

The deuterodifluoromethyl group CF_(2)D combines the structural features of CF_(3) and CD_(3) groups,finding wide applications in diverse pharmaceutical *** this work,the electrochemical monodeuterodefluorination of Ar-CF_(3) to Ar-CF_(2)D is achieved with an unreported deuterium atom transfer-coupled electron transfer pathway using D_(2)O as the only D source for the first time.A series of Ar-CF_(3) substances,including commercial pharmaceuticals,were converted to corresponding CF_(2)D products in up to 91% yield and 97% D-incorporation.A variety of functional groups,including pyridine,pyrrole,furan,thiophene,borate,thio ether,amine,amide,sulfonamide,and halide,are well tolerated in this protocol.t-BuOLi was an essential reagent to achieve chemoselective mono-defluorination.

Toughening additive manufactured Zr-based bulk metallic glass composites by martensite phase transformation

Additive manufacturing technology based on laser powder bed fusion(LPBF)offers a novel approach for fabricating bulk metallic glass(BMG)products without restriction in size and ***,the BMGs prepared by LPBF usually suffered from less plasticity and poorer fracture toughness as compared to their cast counterparts due to partial crystallization in heat-affected zones(HAZs).Since crystallization in HAZs is hard to avoid completely in LPBF BMGs,it is desirable to design a suitable alloy system,in which only ductile crystalline phase,instead of brittle intermetallics,is formed in *** unique structure could effectively increase the toughness/plasticity of the LPBF *** achieve this goal,a quaternary BMG system with a composition of Zr_(47.5)Cu_(45.5)Al_(5)Co_(2)is adopted and subjected to *** is found that nearly a single phase of B_(2)-ZrCu is precipitated in HAZs,while a fully amorphous phase is formed in molten pools(MPs).This B_(2)phase reinforced BMG composite exhibits excellent mechanical properties with enhanced plasticity and ***,it is easy to modulate the mechanical properties by altering the amount of the B_(2)phase via adjusting the laser energy ***,the best combination of strength,plasticity,and notch toughness is obtained in the BMG composite containing 27.4%B_(2)phase and 72.6%amorphous phase,which exhibits yield strength(σ_(s))of 1423 MPa,plastic strain(ε_(p))of 4.65%,and notch toughness(K_(q))of 53.9 MPa m 1/***,a notable strain-hardening is also *** improvement of plasticity/toughness and appearance of strain-hardening behavior are mainly due to the martensite phase transformation from the B_(2)phase to the Cm phase during plastic deformation(i.e.,the phase transformation-induced plasticity effect).The current work provides a guide for making advanced BMGs and BMG composites by additive manufacturing.

Activatable photoacoustic bioprobe for visual detection of aging in vivo

Aging is a natural physiological process with various challenges, related to the loss of homeostasis within the organism, which is not a disease, but a significantly strong risk factor for multiple diseases, including myocardial infarction, stroke, some age-related cancers, macular degeneration, osteoarthritis, neurodegeneration, and many others. In the body, the main manifestation of aging is cellular aging, which exists within tissues and has a local or global impact on tissue function. However, the lack of effective aging detection tools has always been an issue that cannot be ignored in the field of aging research. Therefore,it is necessary to construct a non-invasive tool for in vivo detection of aging. Here, we show that the photoacoustic probe(LGAL), which has peak excitation and emission wavelengths in the near-infrared optical window, binds in vivo and at high contrast to the hallmark of aging, and allows for the microscopic imaging of aging through the intact mice. Firstly, this tool LGAL has been successfully applied to detect senescence in cells, displaying stronger photoacoustic signals than normal cells. Then, by using the photoacoustic probe, the blood vessels and tissues inside the mice can be visualized. Young and elderly mice exhibit varying intensities of photoacoustic signals, marking the first time a probe has been used to explore the aging of blood vessels and tissues inside the mice. Finally, we monitored the changes in the degree of aging during tumor treatment under photoacoustic(PA) imaging for the first time. As the treatment time increased, the degree of aging of the tumor gradually deepened. We expect the powerful tool could be a noninvasive and powerful tool for the study of aging biology.

Carbonaceous materials have been recognized as one of the most promising anode materials for potassium-ion batteries(PIBs)due to their abundant raw materials,controllable structure,superior conductivity,and good chemical ***,the large radius of K ions and the low potassium content of intercalation compounds result in the sluggish storage kinetics and low reversible capacity of carbon *** this work,we present a unique heteroatom-doped carbon composite(denoted as NS-MC/SC)through a facile interfacial assembly route and simple heat-treatment process,where NS-MC is well grafted onto the biomass-derived spore carbon(SC).This unique structural design endows it with abundant mesoporous channels,expanded layer spacing,and highly doped N and *** these merits,the NS-MC/SC anode in PIBs exhibits a high reversible capacity of 350.4 mAh·g^(-1) at 100 mA·g^(-1) after 300 cycles,and an outstanding cycling ***,in-situ Raman spectra further verify the high reversibility of K ions insertion/***,theoretical simulations also reveal that the N,S dual-doping is an efficient approach for improving the potassium-ion storage performance of NS-MC/SC.

Small-diameter acellular porcine corneal stroma for peripheral corneal ulceration treatment

AIM:To evaluate the clinical efficacy of small-diameter acellular porcine corneal stroma(SAPS)for the treatment of peripheral corneal ulceration(PCU).METHODS:This retrospective clinical study included 18 patients(18 eyes)with PCU between April 2018 and December *** patients had PCU and underwent lamellar keratoplasty with *** indicators included preoperative and postoperative best-corrected visual acuity(BCVA)and transparency of *** infection control rate in the surgical eye-lesion area was also ***:Eighteen patients underwent lamellar keratoplasty with SAPS to treat *** of the patients experienced rejection after 6mo(18/18)and 12mo(16/16)of *** BCVA(0.47±0.30)at the 6mo followup after operation was significantly improved compared with the baseline(0.99±0.80),and the difference was statistically significant(Z=-3.415,P<0.05).The BCVA at the 12mo follow-up after operation was not statistically significant compared to the 6mo(Z=0,P=1).With time,the SAPS graft gradually became *** the 6mo(18/18)and 12mo(16/16)follow-up,none of the patients had recurrent corneal ***:SAPS is clinically effective in the treatment of PCU,improving the patient’s BCVA and reducing the incidence of rejection after keratoplasty.

Numerical investigation of the coupled aero-hydrodynamic performances of a semi-submersible floating offshore wind turbine with inclined columns

Numerical investigations of floating platforms with different outer column inclined angles under two operating conditions of regular wave and irregular wave are presented in this paper.A coupled aero-hydrodynamic computational fluid dynamics in-house solver FOWT-UALM-SJTU is applied for the ***,the validation for wave and wind generation are conducted to determine mesh distribution *** on these,the hydrodynamic motion response,aerodynamic performance and wake flow are analyzed to explore the impact of inclined *** spectral analysis on the motion response under wave action,discuss the aerodynamic attack angle and inflow wind velocity along the blade spanwise direction in detail,reveal different trends in wake development and *** results show that for the regular wave condition with the increase of inclined angles,the equilibrium position of surge motion is constantly rising,while pitch is *** maximum root mean square(rms)value occurs at angle=30°,compared with the original OC4 FOWT,the rms in power and thrust increase 0.35%,0.71%.And there are two low regions of attack angle and high regions of axial inflow velocity,corresponding to aerodynamic *** spectral analysis indicates that the natural frequency of pitch motion will increase with inclined ***,from the middle to far region of wake flow,the velocity recovery of FOWT with inclined angle will become faster,which is beneficial for downstream turbines to enhance more wind energy.
