H2 strain attenuated live hepatitis A vaccines:protective efficacy in a hepatitis A outbreak

AIM:To investigate the protective efficacy of H2 strain attenuated live hepatitis A vaccines (H2-strain vaccines) in hepatitis A (HA) ***:With the permission of their parents, 5551 pre-school and grade 1-3 primary school children were inoculated with 1 dose (10(6.5) TCID(50)) of H2 strain vaccines in a nonrandomized, controlled trial conducted in Fucheng County, Hebei Province in May *** 6485 children in the same grades and compatible in gender and age were enrolled as controls. Epidemiological and serological survey was conducted to evaluate the protective efficacy of the vaccines. ELISA was used to detect serum IgM ***:HA outbreak started in early May 1998, peaked in the middle of the same month, and lasted about 80 days. Overall 302 HA cases were found, 192(63.58%) were 5-9 years old. One vaccinee and 25 control cases were found to have hepatitis A, which account for 0.28% (1/356) and 5.92% (25/422) of all vaccinees and controls in the 14 villages, respectively. The protective efficacy of vaccines was 95.27% (95% CI: 85.83%-104.72%). In subjects tested for anti-HAV IgM from 13 villages, 1(0.40%) overt and 11(4.06%) asymptomatic HAV cases were found in 271 vaccinees but 21(6.69%) of overt and asymptomatic ones were found in 314 ***:H2 strain vaccines were excellent in preventing overt hepatitis A,but not so effective in preventing asymptomatic hepatitis A virus infection.A booster dose might be needed to get permanent reliable immunity.

Interannual succession of phytoplankton community in a canyon-shaped drinking water reservoir during the initial impoundment period:Taxonomic versus functional groups

During the initial impoundment period of a canyon-shaped reservoir,the water body fluctuated violently regarding water level,hydrological condition,and thermal *** variations may alter the structure of phytoplankton community,resulting in algal blooms and seriously threatening the ecological security of the *** is of great significance to understand the continuous changes of phytoplankton in the initial impoundment period for the protection of reservoir water ***,a two-year in-situ monitoring study was conducted on water quality and phytoplankton in a representative canyonshaped reservoir named Sanhekou and the interannual changes of phytoplankton community and its response to environmental changes during the initial impoundment period were discussed at taxonomic versus functional classification *** results showed that the total nitrogen and permanganate index levels were relatively high in the first year due to rapid water storage and heavy rainfall input,and the more stable hydrological conditions in the second year promoted the increase of algae density and the transformation of community,and the proportion of cyanobacteria increased *** succession order of phytoplankton in the first year of the initial impoundment periodwas Chlorophyta-Bacillariophyta-Chlorophyta,or J/F/X1-P/MP/W1-A/X1/MP,*** the succession order in the second year was Cyanobacteria/Chlorophyta-Bacillariophyta-Chlorophyta,or L_(M)/G/P-P/A/X1-X1/J/*** temperature,relativewater column stability,mixing depth,and pHwere crucial factors affecting phytoplankton community *** study revealed the interannual succession law and driving factors of phytoplankton in the initial impoundment period and provided an important reference for the operation management and ecological protection of canyon-shaped reservoirs.

An electrically activable nanochip to intensify gas-ionic-immunotherapy

Excess intracellular H_(2)S induces destructive mitochondrial toxicity,while overload of Zn^(2+)results in cell pyroptosis and potentiates the tumor immunogenicity for ***,the precise delivery of both therapeutics remains a great ***,an electrically activable ZnS nanochip for the controlled release of H_(2)S and Zn^(2+)was developed for enhanced gas-ionic-immunotherapy(GIIT).Under an electric field,a locality with particularly high concentrations of H_(2)S and Zn^(2+)was established by the voltage-controlled degradation of the ZnS nanoparticles(NPs).Consequently,the ZnS nanochip-mediated gas-ionic therapy(GIT)resulted in mitochondrial membrane potential depolarization,energy generation inhibition,and oxidative stress imbalance in tumor ***,the cyclic guanosine monophosphate-adenosine monophosphate synthase-stimulator of interferon genes(cGAS-STING)signaling pathway was activated due to the mitochondrial ***,the released Zn^(2+)resulted in the increase of the intracellular Zn levels and cell pyroptosis,which enhanced the immunogenicity via the release of damage-associated molecular patterns(DAMPs).In vitro and in vivo studies revealed that the ZnS nanochip-based GIT effectively eliminated the tumors under an electric field and mobilized the cytotoxic T lymphocytes for *** combination withαCTLA-4 further promoted the adaptive immune response and inhibited tumor metastasis and long-term tumor *** work presented an electrically activable ZnS nanochip for combined immunotherapy,which might inspire the development of electric stimulation therapy.

Integrating well logs, 3D seismic, and earthquake data for comprehensive prediction of 3D in-situ stress orientations: A case study from the Weiyuan area in the Sichuan Basin, China

Determining the orientation of in-situ stresses is crucial for various geoscience and engineering applications. Conventional methods for estimating these stress orientations often depend on focal mechanism solutions(FMSs) derived from earthquake data and formation micro-imager(FMI) data from well ***, these techniques can be costly, depth-inaccurate, and may lack spatial coverage. To address this issue, we introduce the use of three-dimensional(3D) seismic data(active sources) as a lateral constraint to approximate the 3D stress orientation field. Recognizing that both stress and fracture patterns are closely related to seismic velocity anisotropy, we derive the orientation of azimuthal anisotropy from multi-azimuth 3D seismic data to compensate for the lack of spatial stress orientation information. We apply our proposed workflow to a case study in the Weiyuan area of the Sichuan Basin, China, a region targeted for shale gas production. By integrating diverse datasets, including 3D seismic, earthquakes, and well logs, we develop a comprehensive 3D model of in-situ stress(orientations and magnitudes). Our results demonstrate that the estimated anisotropy orientations from 3D seismic data are consistent with the direction of maximum horizontal principal stress(SHmax) obtained from FMIs. We analyzed 12earthquakes(magnitude > 3) recorded between 2016 and 2020 for their FMSs and compressional axis(Paxis) orientations. The derived SHmax direction from our 3D stress model is 110°ES(East-South), which shows excellent agreement with the FMSs(within 3.96°). This close alignment validates the reliability and precision of our integrated method for predicting 3D SHmax orientations.

Mutual regulation of microglia and astrocytes after Gas6 inhibits spinal cord injury

Invasive inflammation and excessive scar formation are the main reasons for the difficulty in repairing nervous tissue after spinal cord *** and astrocytes play key roles in the spinal cord injury micro-environment and share a close ***,the mechanisms involved remain *** this study,we found that after spinal cord injury,resting microglia(M0)were polarized into pro-inflammatory phenotypes(MG1 and MG3),while resting astrocytes were polarized into reactive and scar-forming *** expression of growth arrest-specific 6(Gas6)and its receptor Axl were significantly down-regulated in microglia and astrocytes after spinal cord *** vitro experiments showed that Gas6 had negative effects on the polarization of reactive astrocytes and pro-inflammatory microglia,and even inhibited the cross-regulation between *** further demonstrated that Gas6 can inhibit the polarization of reactive astrocytes by suppressing the activation of the Yes-associated protein signaling ***,in turn,inhibited the polarization of pro-inflammatory microglia by suppressing the activation of the nuclear factor-κB/p65 and Janus kinase/signal transducer and activator of transcription signaling *** vivo experiments showed that Gas6 inhibited the polarization of pro-inflammatory microglia and reactive astrocytes in the injured spinal cord,thereby promoting tissue repair and motor function ***,Gas6 may play a role in the treatment of spinal cord *** can inhibit the inflammatory pathway of microglia and polarization of astrocytes,attenuate the interaction between microglia and astrocytes in the inflammatory microenvironment,and thereby alleviate local inflammation and reduce scar formation in the spinal cord.

Objective To investigate the therapeutic effect of Xiangshao Granules(XSG)on post-stroke depression(PSD)and explore the underlying *** Forty-three C57BL/6J mice were divided into 3 groups:sham(n=15),PSD+vehicle(n=14),and PSD+XSG(n=14)groups according to a random number *** PSD models were constructed using chronic unpredictable mild stress(CUMS)after middle cerebral artery occlusion(MCAO).The sham group only experienced the same surgical operation,but without MACO and CUMS *** XSG group received XSG(60 mg/kg per day)by gavage for 4 *** mice in the sham and vehicle groups were given the same volume of 0.9%saline at the same *** body weight and behavior tests including open field test,sucrose preference test,tail suspension test,and elevated plus-maze test,were used to validate the PSD mouse ***-time fluorescence quantitative polymerase chain reaction(RT-qPCR),enzyme-linked immunosorbent assay(ELISA),and immunofluorescence staining were used to evaluate the anti-inflammatory effects of *** potential molecular mechanisms were explored and verified through network pharmacology analysis,Nissl staining,Western blot,ELISA,and RT-qPCR,*** The body weight and behavior tests showed that MCAO combined with CUMS successfully established the PSD *** alleviated neuronal damage,reduced the expressions of pro-apoptotic proteins Caspase-3 and B-cell lymphoma-2(BCL-2)-associated X(BAX),and increased the expression of anti-apoptotic protein BCL-2 in PSD mice(P<0.05 or P<0.01).XSG inhibited microglial activation and the expressions of pro-inflammatory cytokines including tumor necrosis factor-α,interleukin(IL)-1β,and IL-6 via the toll-like receptor 4/nuclear factor kappa-B signaling pathway in PSD mice(P<0.05 or P<0.01).Furthermore,XSG decreased the expression of indoleamine 2,3-dioxygenase1(IDO1)and increased the concentration of 5-hydroxytryptamine in PSD mice(P<0.05 or P<0.01).Conclusion XSG could reverse

All-season thermal management with zero energy consumption and emissions is more crucial to global decarbonization over traditional energy-intensive cooling/heating ***,the static single thermal management for cooling or heating fails to self-regulate the temperature in dynamic seasonal temperature ***,inspired by the dual-temperature regulation function of the fur color changes on the backs and abdomens of penguins,a smart thermal management composite hydrogel(PNA@H-PM Gel) system was subtly created though an "on-demand" dual-layer structure design *** PNA@H-PM Gel system features synchronous solar and thermal radiation modulation as well as tunable phase transition temperatures to meet the variable seasonal thermal requirements and energy-saving demands via self-adaptive radiative cooling and solar heating ***,this system demonstrates superb modulations of both the solar reflectance(ΔR=0.74) and thermal emissivity(ΔE=0.52) in response to ambient temperature changes,highlighting efficient temperature regulation with average radiative cooling and solar heating effects of 9.6℃ in summer and 6.1℃ in winter,***,compared to standard building baselines,the PNA@H-PM Gel presents a more substantial energy-saving cooling/heating potentials for energy-efficient buildings across various regions and *** novel solution,inspired by penguins in the real world,will offer a fresh approach for producing intelligent,energy-saving thermal management materials,and serve for temperature regulation under dynamic climate conditions and even throughout all seasons.

Application of Dorzagliatin in peritoneal dialysis patients with type 2 diabetes mellitus:A case report

BACKGROUND Treating diabetes in dialysis patients remains a challenge,with many hypoglycemic drugs requiring dose adjustments or avoidance in these *** SUMMARY This report describes an 83-year-old female patient with a 30-year history of type 2 diabetes(T2DM)who had struggled to control her blood sugar for more than a *** had a history of high blood pressure for 30 years,had undergone continuous ambulatory peritoneal dialysis for more than two years,was 163 cm tall,weighed 77 kg,and had a body mass index of 28.98 kg/*** intensive insulin therapy at a daily dose of 150 units,adding Dorzagliatin at a dosage of 75 mg orally twice daily led to immediate blood sugar improvement and a gradual reduction in insulin *** one month of follow-up,the fasting plasma glucose was 6-8 mmol/L,and the 2-hour postprandial glucose was 8-12 mmol/*** To our knowledge,this report is the first to use Dorzagliatin to treat type 2 diabetes peritoneal dialysis patients with challenging glucose ***,a novel glucokinase activator primarily metabolized by the liver,exhibits no pharmacokinetic differences among patients with varying degrees of chronic kidney *** has a high plasma protein binding rate and may not be cleared by peritoneal dialysis,potentially offering a new glycemic control option for Type 2 diabetic patients on peritoneal dialysis.
