Fragrant rice has a high market value,and it is a popular rice type among consumers owing to its pleasant *** methods,nitrogen(N)fertilizers,and silicon(Si)fertilizers can affect the grain yield and fragrance of fragrant ***,the core commercial rice production attributes,namely the head rice yield(HRY)and 2-acetyl-1-pyrroline(2-AP)content of fragrant rice,under various nitrogen and silicon(N-Si)fertilization levels and different plantation methods remain *** field experiment in this study was performed in the early seasons of 2018 and 2019 with two popular indica fragrant rice cultivars(Yuxiangyouzhan and Xiangyaxiangzhan).They were grown under six N-Si fertilization treatments(combinations of two levels of Si fertilizer,0 kg Si ha^(−1)(Si0)and 150 kg Si ha^(−1)(Si1),and three levels of N fertilizer,0 kg N ha^(−1)(N0),150 kg N ha^(−1)(N1),and 220 kg N ha^(−1)(N2))and three plantation methods(artificial transplanting(AT),mechanical transplanting(MT),and mechanical direct-seeding(MD)).The results showed that the N-Si fertilization treatments and all the plantation methods significantly affected the HRY and 2-AP content and related parameters of the two different fragrant rice *** with the Si0N0 treatment,the N-Si fertilization treatments resulted in higher HRY and 2-AP *** rates of brown rice,milled rice,head rice,and chalky rice of the fragrant rice also improved with the N-Si fertilization *** N-Si fertilization treatments increased the activities of N metabolism enzymes and the accumulation of N and Si in various parts of the fragrant rice,and affected their antioxidant response *** key parameters for the HRY and 2-AP content were assessed by redundancy ***,the structural equation model revealed that the Si and N accumulation levels indirectly affected the HRY by affecting the N metabolism enzyme activity,N use efficiency,and grain quality of fragrant ***,high N

Mechanisms of vacuolar phosphate efflux supporting soybean root hair growth in response to phosphate deficiency

Phosphorus is an essential macronutrient for plant growth and *** response to phosphate(Pi)deficiency,plants rapidly produce a substitutive amount of root hairs;however,the mechanisms underlying Pi supply for root hair growth remain ***,we observed that soybean(Glycine max)plants maintain a consistent level of Pi within root hairs even under external Pi *** therefore investigated the role of vacuole-stored Pi,a major Pi reservoir in plant cells,in supporting root hair growth under Pi-deficient *** findings indicated that two vacuolar Pi efflux(VPE)transporters,GmVPE1 and GmVPE2,remobilize vacuolar stored Pi to sustain cytosolic Pi content in root hair *** analysis showed that double mutants of GmVPE1 and GmVPE2 exhibited reduced root hair growth under low Pi ***,GmVPE1 and GmVPE2 were highly expressed in root hairs,with their expression levels significantly upregulated by low Pi *** analysis revealed that GmRSL2(ROOT HAIR DEFECTIVE 6-like 2),a transcription factor involved in root hair morphogenesis,directly binds to the promoter regions of GmVPE1 and GmVPE2,and promotes their expressions under low Pi ***,mutants lacking both GmRSL2 and its homolog GmRSL3 exhibited impaired root hair growth under low Pi stress,which was rescued by overexpressing either GmVPE1 or *** together,our study has identified a module comprising vacuolar Pi exporters and transcription factors responsible for remobilizing vacuolar Pi to support root hair growth in response to Pi deficiency in soybean.

Background: The absence of a safe and effective therapy for recurrent spontaneous abortion due to a maternofetal failure in immunological tolerance remains an intractable clinical obstacle for surgeons. Recently, traditional Chinese medicine has become a feasible alternative for certain diseases, including recurrent spontaneous abortion. However, because of the complex composition of the traditional Chinese medicine formula, its action mechanism remains unclear. Methods: We selected two isolated active ingredients (RAMP and baicalin) from the traditional Chinese medicine formula and used an abortion-prone CBA/J × DBA/2 model to simulate human RSA and compared the changes in fetal resorption rate, Treg cell percentage, and relevant cytokines before and after combination therapy. In addition, The mechanisms were preliminarily discussed using in vitro differentiation models. Results: In CBA/J × DBA/2 abortion-prone mice, the combination therapy resulted in a lower embryo resorption rate compared to that obtained with individual delivery of either RAMP or baicalin, thereby playing an embryo-protective role through the increase in Treg cells for the maintenance of maternal-fetal immune tolerance. In in vitro primary cell differentiation experiments, the concentration of Treg cells significantly increased from 11% to 17.9% after the combination therapy compared to that of the single administration group. Conclusion: the synergistic effects of RAMP and baicalin were responsible for Treg differentiation. The present study provides a solid basis for improving the applicability of traditional Chinese herbs in the treatment of recurrent spontaneous abortion.

Alleviated photoinhibition on nitrification in the Indian Sector of the Southern Ocean

Nitrification, a central process in the marine nitrogen cycle, produces regenerated nitrate in the euphotic zone and emits N2O, a potent greenhouse gas as a by-product. The regulatory mechanisms of nitrification in the Southern Ocean, which is a critical region for CO2sequestration and radiative benefits, remain poorly ***, we investigated the in situ and dark nitrification rates in the upper 500 m and conducted substrate kinetics experiments across the Indian Sector in the Cosmonaut and Cooperation seas in the late austral summer. Our findings indicate that light inhibition of nitrification decreases exponentially with depth, exhibiting a light threshold of 0.53% photosynthetically active radiation. A positive relationship between dark nitrification and apparent oxygen utilization suggests a dependence on substrate availability from primary ***, an increased NH4+affinity（α） increases with depth and transitions from light to dark, decreases with increasing ambient NH4+, and exhibits a latitudinal distribution, reflecting substrate utilization strategies. We also reveal that upwelling in Circumpolar Deep Water（CDW） stimulates nitrification through the introduction of potentially higher iron and deep diverse nitrifying microorganisms with higher α. We conclude that although light is the primary limiting factor for nitrification in summer, coupling between substrate availability and CDW upwelling can overcome this limitation, thereby alleviating photoinhibition by up to 45% ± 5.3%.

Potassium and calcium channels in different nerve cells act as therapeutic targets in neurological disorders

The central nervous system, information integration center of the body, is mainly composed of neurons and glial cells. The neuron is one of the most basic and important structural and functional units of the central nervous system, with sensory stimulation and excitation conduction functions. Astrocytes and microglia belong to the glial cell family, which is the main source of cytokines and represents the main defense system of the central nervous system. Nerve cells undergo neurotransmission or gliotransmission, which regulates neuronal activity via the ion channels, receptors, or transporters expressed on nerve cell membranes. Ion channels, composed of large transmembrane proteins, play crucial roles in maintaining nerve cell homeostasis. These channels are also important for control of the membrane potential and in the secretion of neurotransmitters. A variety of cellular functions and life activities, including functional regulation of the central nervous system, the generation and conduction of nerve excitation, the occurrence of receptor potential, heart pulsation, smooth muscle peristalsis, skeletal muscle contraction, and hormone secretion, are closely related to ion channels associated with passive transmembrane transport. Two types of ion channels in the central nervous system, potassium channels and calcium channels, are closely related to various neurological disorders, including Alzheimer's disease, Parkinson's disease, and epilepsy. Accordingly, various drugs that can affect these ion channels have been explored deeply to provide new directions for the treatment of these neurological disorders. In this review, we focus on the functions of potassium and calcium ion channels in different nerve cells and their involvement in neurological disorders such as Parkinson's disease, Alzheimer's disease, depression, epilepsy, autism, and rare disorders. A novel alternative for pyrogen detection based on a transgenic cell line

Pyrogen,often as a contaminant,is a key indicator affecting the safety of almost all parenteral drugs(including biologicals,chemicals,traditional Chinese medicines and medical devices).It has become a goal to completely replace the in vivo rabbit pyrogen test by using the in vitro pyrogen test based on the promoted‘reduction,replacement and refinement’principle,which has been highly considered by regulatory agencies from different *** used NF-κB,a central signalling molecule mediating inflammatory responses,as a pyrogenic marker and the monocyte line THP-1 transfected with a luciferase reporter gene regulated by NF-κB as an in vitro model to detect pyrogens by measuring the intensity of a fluorescence ***,we show that this test can quantitatively and sensitively detect endotoxin(lipopolysaccharide from different strains)and nonendotoxin(lipoteichoic acid,zymosan,peptidoglycan,lectin and glucan),has good stability in terms of NF-κB activity and cell phenotypes at 39 cell passages and can be applied to detect pyrogens in biologicals(group A&C meningococcal polysaccharide vaccine;basiliximab;rabies vaccine(Vero cells)for human use,freeze-dried;Japanese encephalitis vaccine(Vero cells),inactivated;insulin aspart injection;human albumin;recombinant human erythropoietin injection(CHO Cell)).The within-laboratory reproducibility of the test in three independent laboratories was 85%,80%and 80%and the interlaboratory reproducibility among laboratories was 83.3%,95.6%and 86.7%.The sensitivity(true positive rate)and specificity(true negative rate)of the test were 89.9%and 90.9%,*** summary,the test provides a novel alternative for pyrogen detection.
