Polysaccharide of Alocasia cucullata Exerts Antitumor Effect by Regulating Bcl-2, Caspase-3 and ERK1/2 Expressions during Long-Time Administration

Objective: To study the in vitro and in vivo antitumor effects of the polysaccharide of Alocasia cucullata(PAC) and the underlying mechanism. Methods: B16F10 and 4T1 cells were cultured with PAC of 40 μg/m L, and PAC was withdrawn after 40 days of administration. The cell viability was detected by cell counting kit-8. The expression of Bcl-2 and Caspase-3 proteins were detected by Western blot and the expressions of ERK1/2 mRNA were detected by quantitative real-time polymerase chain reaction(qRT-PCR). A mouse melanoma model was established to study the effect of PAC during long-time administration. Mice were divided into 3 treatment groups: control group treated with normal saline, positive control group(LNT group) treated with lentinan at 100 mg/(kg·d), and PAC group treated with PAC at 120 mg/(kg·d). The pathological changes of tumor tissues were observed by hematoxylin-eosin staining. The cell apoptosis of tumor tissues was detected by TUNEL staining. Bcl-2 and Caspase-3 protein expressions were detected by immunohistochemistry, and the expressions of ERK1/2, JNK1 and p38 mRNA were detected by qRT-PCR. Results: In vitro, no strong inhibitory effects of PAC were found in various tumor cells after 48 or 72 h of administration. Interestingly however, after 40 days of cultivation under PAC, an inhibitory effect on B16F10 cells was found. Correspondingly, the long-time administration of PAC led to downregulation of Bcl-2 protein(P <0.05), up-regulation of Caspase-3 protein(P<0.05) and ERK1 mRNA(P<0.05) in B16F10 cells. The above results were verified by in vivo experiments. In addition, viability of B16F10 cells under long-time administration culture in vitro decreased after drug withdrawal, and similar results were also observed in 4T1 cells. Conclusions: Long-time administration of PAC can significantly inhibit viability and promote apoptosis of tumor cells, and had obvious antitumor effect in tumor-bearing mice.

STING(also known as MITA)is an adaptor protein that mediates cytoplasmic DNA-triggered signaling,and aberrant activation of STING/MITA by cytosolic self-DNA or gain-of-function mutations causes severe ***,we show that STING-mediated inflammation and autoimmunity are promoted by RNF115 and alleviated by the RNF115 inhibitor disulfiram(DSF).Knockout of RNF115 or treatment with DSF significantly inhibit systemic inflammation and autoimmune lethality and restore immune cell development in Trex1^(–/–)mice and STING^(N153S/WT) bone marrow chimeric *** addition,knockdown or pharmacological inhibition of RNF115 substantially downregulate the expression of IFN-α,IFN-γand proinflammatory cytokines in PBMCs from patients with systemic lupus erythematosus(SLE)who exhibit high concentrations of dsDNA in peripheral ***,knockout or inhibition of RNF115 impair the oligomerization and Golgi localization of STING in various types of cells transfected with cGAMP and in organs and cells from Trex1^(–/–)***,knockout of RNF115 inhibits the activation and Golgi localization of STINGN153S as well as the expression of proinflammatory cytokines in myeloid cells but not in endothelial cells or *** together,these findings highlight the RNF115-mediated cell type-specific regulation of STING and STINGN153S and provide potential targeted intervention strategies for STING-related autoimmune diseases.

Recent advances in multifunctional metal-organic frameworks for lithium metal batteries

In view of novel materials in the field of lithium metal batteries(LMBs), metal-organic frameworks(MOFs) have attracted extensive research interest owing to their controllable pore size, unsaturated metal sites and multifunctional organic groups. A variety of MOFs have been elaborately calculated and synthesized to be applied as separator coating, electrolyte modulators and solid-state electrolyte fillers in LMBs. In this mini-review, we summarize the mechanism of MOFs to limit the migration of anions, improve the Li-ion transference number and prolong the lifespan of LMBs. Suitable pore structure of MOFs can physically restrict the movement of Li^(+). Unsaturated metal sites can adsorb anions by electrostatic interaction. In addition,multifunctional organic functional groups that limit the migration of anions are discussed. Finally, the key challenges and perspectives in the development direction of MOFs-based separators and electrolytes are further elaborated.

Multi-Scale Mixed Attention Tea Shoot Instance Segmentation Model

Tea leaf picking is a crucial stage in tea production that directly influences the quality and value of the *** tea-picking machines may compromise the quality of the tea ***-quality teas are often handpicked and need more delicate operations in intelligent picking *** with traditional image processing techniques,deep learning models have stronger feature extraction capabilities,and better generalization and are more suitable for practical tea shoot ***,current research mostly focuses on shoot detection and cannot directly accomplish end-to-end shoot segmentation *** propose a tea shoot instance segmentation model based on multi-scale mixed attention(Mask2FusionNet)using a dataset from the tea garden in *** further analyzed the characteristics of the tea shoot dataset,where the proportion of small to medium-sized targets is 89.9%.Our algorithm is compared with several mainstream object segmentation algorithms,and the results demonstrate that our model achieves an accuracy of 82%in recognizing the tea shoots,showing a better performance compared to other *** ablation experiments,we found that ResNet50,PointRend strategy,and the Feature Pyramid Network(FPN)architecture can improve performance by 1.6%,1.4%,and 2.4%,*** experiments demonstrated that our proposed multi-scale and point selection strategy optimizes the feature extraction capability for overlapping small *** results indicate that the proposed Mask2FusionNet model can perform the shoot segmentation in unstructured environments,realizing the individual distinction of tea shoots,and complete extraction of the shoot edge contours with a segmentation accuracy of 82.0%.The research results can provide algorithmic support for the segmentation and intelligent harvesting of premium tea shoots at different scales.

A novel algorithm for evaluating cement azimuthal density based on perturbation theory in horizontal well

Cement density monitoring plays a vital role in evaluating the quality of cementing projects,which is of great significance to the development of oil and ***,the presence of inhomogeneous cement distribution and casing eccentricity in horizontal wells often complicates the accurate evaluation of cement azimuthal *** this regard,this paper proposes an algorithm to calculate the cement azimuthal density in horizontal wells using a multi-detector gamma-ray detection *** spatial dynamic response functions are simulated to obtain the influence of cement density on gamma-ray counts by the perturbation theory,and the contribution of cement density in six sectors to the gamma-ray recorded by different detectors is obtained by integrating the spatial dynamic response *** with the relationship between gamma-ray counts and cement density,a multi-parameter calculation equation system is established,and the regularized Newton iteration method is employed to invert casing eccentricity and cement azimuthal *** approach ensures the stability of the inversion process while simultaneously achieving an accuracy of 0.05 g/cm^(3) for the cement azimuthal *** accuracy level is ten times higher compared to density accuracy calculated using calibration ***,this algorithm enhances the accuracy of cement azimuthal density evaluation,provides valuable technical support for the monitoring of cement azimuthal density in the oil and gas industry.

Being invisible ad libitum has long captivated the popular imagination,particularly in terms of safeguarding modern high-end instruments from potential *** ago,the advent of metamaterials and transformation optics sparked considerable interest in invisibility cloaks,which have been mainly demonstrated in ground and waveguide ***,an omnidirectional flying cloak has not been achieved,primarily due to the challenges associated with dynamic synthesis of metasurface *** demonstrate an autonomous aeroamphibious invisibility cloak that incorporates a suite of perception,decision,and execution modules,capable of maintaining invisibility amidst kaleidoscopic backgrounds and neutralizing external *** physical breakthrough lies in the spatiotemporal modulation imparted on tunable metasurfaces to sculpt the scattering field in both space and frequency *** intelligently control the spatiotemporal metasurfaces,we introduce a stochastic-evolution learning that automatically aligns with the optimal solution through maximum probabilistic *** a fully self-driving experiment,we implement this concept on an unmanned drone and showcase adaptive invisibility in three canonical landscapes-sea,land,and air-with a similarity rate of up to 95%.Our work extends the family of invisibility cloaks to flying modality and inspires other research on material discoveries and homeostatic meta-devices.

Previous studies on genetic diseases predominantly focused on protein-coding variations, overlooking the vast noncoding regions in the human genome. The development of high-throughput sequencing technologies and functional genomics tools has enabled the systematic identification of functional noncoding variants. These variants can impact gene expression, regulation, and chromatin conformation, thereby contributing to disease pathogenesis. Understanding the mechanisms that underlie the impact of noncoding variants on genetic diseases is indispensable for the development of precisely targeted therapies and the implementation of personalized medicine strategies. The intricacies of noncoding regions introduce a multitude of challenges and research opportunities. In this review, we introduce a spectrum of noncoding variants involved in genetic diseases, along with research strategies and advanced technologies for their precise identification and in-depth understanding of the complexity of the noncoding genome. We will delve into the research challenges and propose potential solutions for unraveling the genetic basis of rare and complex diseases.

Analysis of metabolic characteristics of metabolic syndrome in elderly patients with gastric cancer by non-targeted metabolomics

BACKGROUND The relationship between metabolic syndrome(MetS)and gastric cancer(GC),which is a common metabolic disease,has attracted much ***,the specific metabolic characteristics of MetS in elderly patients with GC remain *** To investigate the differentially abundant metabolites and metabolic pathways between preoperative frailty and MetS in elderly patients with GC based on nontargeted metabolomics *** In this study,125 patients with nonfrail nonmeal GC were selected as the control group,and 50 patients with GC in the frail group were selected as the frail ***-five patients with GC combined with MetS alone were included in the MetS group,and 50 patients with GC combined with MetS were included in the MetS *** metabolomics techniques were used to measure plasma metabolite levels by ultrahigh-performance liquid chromatography-mass *** statistical analysis was performed by principal component analysis,orthogonal partial least squares,pattern recognition analysis,cluster analysis,and metabolic pathway *** A total of 125 different metabolites,including amino acids,glycerophospholipids,sphingolipids,fatty acids,sugars,nucleosides and nucleotides,and acidic compounds,were identified via nontargeted metabolomics *** with those in the control group,there were 41,32,and 52 different metabolites in the MetS group,the debilitated group,and the combined group,*** metabolites were significantly increased in the MetS *** the weak group,amino acids and most glycerol phospholipid metabolites decreased significantly,and fatty acids and sphingosine increased *** combined group was characterized by significantly increased levels of nucleotide metabolites and acidic *** alanine,aspartic acid,and glutamate metabolic pathways were obviously enriched in the asthenic group,and the glycerol and phospholipid metabolic pathways were obv

Compared with manual sorting of citrus fruit,vision-based sorting solutions can help achieve higher accuracy and *** this study,we present a vision system based on CNN-LSTM,which can cooperate with robotic grippers for real-time sorting and is readily applicable to various citrus processing plants.A CNN-based detector was adopted to detect the defective oranges in view and temporarily classify them into corresponding types,and an LSTM-based predictor was used to predict the position of the oranges in a future frame based on image sequential *** fusion of CNN and LSTM networks enabled the system to track defective ones during rotation and identify their true types,and their future path was also predicted which is vital for predictive control of visually guided robotic *** detection accuracy of 94.1%was obtained based on experimental results,and the error for path prediction was within 4.33 pixels 40 frames *** average time to process a frame was between 28 and 62 frames per second,which also satisfied real-time *** results proved the potential of the proposed system for automated citrus sorting with good precision and efficiency,and it can be readily extended to other fruit crops featuring high versatility.
