Metabolic changes play a crucial role in determining the status and function of macrophages,but how lipid reprogramming in macrophages contributes to tumor progression is not yet fully ***,we investigated the phenotype,contribution,and regulatory mechanisms of lipid droplet(LD)-laden macrophages(LLMs)in hepatocellular carcinoma(HCC).Enriched LLMs were found in tumor tissues and were associated with disease progression in HCC *** LLMs displayed immunosuppressive phenotypes(with extensive expression of TREM2,PD-L1,CD206,and CD163)and attenuated the antitumor activities of CD8^(+)T ***,tumor-induced reshuffling of cellular lipids and TNFα-mediated uptake of tumoral fatty acids contribute to the generation of triglycerides and LDs in *** prolong LLM survival and promote CCL20 secretion,which further recruits CCR6^(+)Tregs to HCC *** LLM formation by targeting DGAT1 and DGAT2,which catalyze the synthesis of triglycerides,significantly reduced Treg recruitment,and delayed tumor growth in a mouse hepatic tumor *** results reveal the suppressive phenotypes and mechanisms of LLM enrichment in HCC and suggest the therapeutic potential of targeting LLMs for HCC patients.

Since the living microorganisms in activated sludge continuously change, it is difficult to conduct controlled experiments and achieve reproducible results for evaluating sludge characteristics. Synthetic sludge, as a chemical surrogate to activated sludge, could be used to investigate the sludge physicochemical properties, and it is desirable to prepare synthetic sludge with similar structure and properties to real activated sludge to explore the flocculation and settlement processes in activated sludge systems. In this work, a high-strength synthetic sludge was prepared with functional polystyrene latex particles as the framework and extracellular polymeric substances （EPS） to modify its surface. The flocculation and settling characteristics of the microspheres and the prepared synthetic sludge were tested. Compared with other three functional polystyrene latex microspheres, the synthetic sludge prepared with EPS-modified polystyrene latex microspheres showed good settling characteristics and a significantly higher strength. They could be used for studying the physicochemical properties of activated sludge.

Photocatalytic removal of uranium has attracted much attention in nuclear wastewater treatment and it is highly needed to develop functional photocatalyst with excellent removal *** this work,seven kinds of carbon dots/carbon nitride(CDs/CN)composites were synthesized and Ser CDs/CN with the best photo-assisted uranium removal performance was screened *** was found that the introduction of CDs could bring in higher photocurrent density,lower interfacial charge transfer impedance and narrower band gap,resulting in a much-improved removal ***/CN had shown a removal capacity as high as 1690 mg/g and the reaction could be operated under air atmosphere which is promising in real application.

Targeting MAPK pathways by naringenin modulates microglia M1 / M2 polarization in lipopolysaccharide-stimulated cultures

Herein we report a high-gain signal-on poly(3,4-ethylenedioxythiophene):poly(styrene sulfonate)(PEDOT:PSS)organic electrochemical transistor(OECT)biosensing using an accumulation-mode PEDOT:PSS OECT modulated by a light-fueled MXene/MOFs/Ni O Schottky *** such a system,the MXene/MOFs/Ni O Schottky heterojunction exhibited superior gating effect,as it not only enabled the fast-directional charge transfer but also guaranteed the maximal accessibility of the electrolyte to topped 2D MXene with large surface *** linkage with a bi-enzyme cascade system,the quinone derivatives produced by the cascade reaction of alkaline phosphatase(ALP)and tyrosinase(TYR)could serve as effective electron acceptors for the representative Ti_(3)C_(2)/PCN-224/Ni O heterojunction,underpinning an innovative method for sensitive detection of ALP activity with a low detection limit of 0.001 U L^(-1).Remarkably,the as-developed system demonstrated a remarkable current gain as high as near 10^(4),which to our knowledge is the highest one among existing OECT biosensory *** work represents a generic protocol to develop the novel signal-on PEDOT:PSS OECT platform towards biochemical detection and beyond.

Ideal Cardiovascular Health is Inversely Associated with Subclinical Atherosclerosis: A Prospective Analysis

Objective Ideal cardiovascular health(CVH) could predict a lower risk of developing cardiovascular diseases. This study was conducted to investigate the association between ideal CVH and subclinical atherosclerosis in a population cohort of Chinese adults aged ≥ 40 years. Methods This study was designed as a cross-sectional analysis of 8,395 participants who had complete data at baseline and a prospective analysis of 4,879 participants who had complete data at 4.3 years of follow-up. Ideal CVH metrics were defined according to the American Heart Association. Subclinical atherosclerosis was evaluated by plaques in carotid arteries, carotid intima-media thickness(CIMT), brachial-ankle pulse wave velocity(baPWV), and urinary albumin-to-creatinine ratio(UACR). Results Both the prevalence and incidence of atherosclerosis measures were found to be decreased with increasing numbers of ideal CVH metrics at baseline(all P values for trend line(both P values for trend line significant association with baPWV(P for trend = 0.0505). Taking participants with 0-1 ideal metric as reference, we found that participants with 5-6 ideal metrics had significantly lower risks of developing carotid plaques(odds ratio, OR = 0.46; 95% confidence interval, CI 0.27-0.79), increased CIMT(OR = 0.60; 95% CI 0.42-0.84), and increased baPWV(OR = 0.57; 95% CI 0.34-0.97) after full adjustments. A significant interactive effect of age and CVH was detected on CIMT and baPWV progression(both P values for interaction linical atherosclerosis in middle-aged and elderly Chinese adults, whereas its dose-response effect was attenuated in individuals aged ≥ 60 years and partially weakened in male participants.

Background： Acupuncture can not only be used for well-known diseases, but also for so-called modern lifestyle-related diseases. Using innovative methods like e.g. new analyses of heart rate variability （HRV）, it is also possible to investigate diseases like burnout syndrome, ie., qi deficiency in Chinese medicine （CM）. Objective： The main object of this research protocol is to perform research on the relationship of burnout syndrome and heart rate （HR） and HRV. Methods： A total of 175 patients with burnout syndrome （qi deficiency syndrome） in five groups and 35 healthy volunteers will be investigated. Based on random numbers generated by computer and concealed in opaque envelops, the patients will be assigned to four acupuncture groups using Zusanli （ST 36） acupuncture stimulation, Guanyuan （CV4） acupuncture stimulation, both points, and both points with Streitberger device respectively, and a moxibustion group using both points mentioned above, with 35 cases in each group. Altogether four different experiments are planned. Experiment 1 includes 70 subjects and is a comparison between a burnout group and a control group （healthy volunteers）. The evaluation parameters are different scores and indices of HR and HRV. Experiment 2 includes 140 subjects and compares the efficacy of different acupuncture points. In experiment 3 （105 subjects）, acupuncture and moxibustion should be compared to healthy volunteers. Experiment 4 （70 subjects） investigates the long-term therapeutic effects of acupuncture and moxibustion on the scores of qi deficiency and HR/HRV in qi deficiency patients. In both the acupuncture and moxibustion groups, a total of 10 treatments will be performed. Conclusions： The joint research aims at the scientific evaluation of CM, mainly in the field of HRV. This parameter could be a very good indicator of the state of health and can be influenced by different acupuncture methods, as shown in the past.

Downregulation of SL-ZH13 transcription factor gene expression decreases drought tolerance of tomato

Zinc finger-homeodomain proteins(ZF-HDs) are transcription factors that regulate plant growth,development,and abiotic stress *** SL-ZH13 gene was found to be significantly upregulated under drought stress treatment in tomato(Solanum lycopersicum) leaves in our previous *** this study,to further understand the role that the SL-ZH13 gene plays in the response of tomato plants to drought stress,the virus-induced gene silencing(VIGS) method was applied to downregulate SL-ZH13 expression in tomato plants,and these plants were treated with drought stress to analyze the changes in drought *** SL-ZH13 silencing efficiency was confirmed by quantitative real-time PCR(qRT-PCR) *** SL-ZH13-silenced plants,the stems wilted faster,leaf shrinkage was more severe than in control plants under the same drought stress treatment conditions,and the mean stem bending angle of SL-ZH13-silenced plants was smaller than that of control *** analyses showed that the activity of superoxide dismutase(SOD) and peroxidase(POD) and the content of proline(Pro) in SL-ZH13-silenced plants were lower than those in control plants after 1.5 and 3 h of drought stress *** malondialdehyde(Mda) content in SL-ZH13-silenced plants was higher than that in control plants after 1.5 and 3 h of drought stress treatment,and H2O2 and O2^-· accumulated much more in the leaves of SL-ZH13-silenced plants than in the leaves of control *** results suggested that silencing the SL-ZH13 gene affected the response of tomato plants to drought stress and decreased the drought tolerance of tomato plants.

Intrahepatic cholangiocarcinoma(ICC)is the second most common liver *** remains the main therapeutic strategy for advanced ICC patients,but chemosensitivity varies ***,we applied cytometry by time-of-flight(CyTOF)to establish the immune profile of peripheral blood mononuclear cells(PBMCs)on the single-cell level at indicated time points before,during,and after *** immunofluorescence staining was applied to examine the spatial distribution of certain immune *** microarrays(TMAs)were used for prognostic evaluation.A total of 20 ICC patients treated with gemcitabine(GEM)were enrolled in our study,including eight cases with good response(R)and 12 cases with non-response(NR).Tremendous changes in PBMC composition,including an increased level of CD4/CD8 double-positive T cells(DPT),were observed after *** with higher level of CD4^(+)CD45RO^(+)CXCR3^(+)T cells before treatment had a favorable response to *** study identified a positive correlation between the percentage of T cell subpopulations and clinical response after chemotherapy,which suggests that it is practical to predict the potential response before treatment by evaluating the proportions of the cell population in PBMCs.
