Switchgrass Biochar Effects on Plant Biomass and Microbial Dynamics in Two Soils from Different Regions

Biochar amendments to soils may alter soil function and fertility in various ways, including through induced changes in the microbial community. We assessed microbial activity and community composition of two distinct clayey soil types, an Aridisol from Colorado （CO） in the U.S. Central Great Plains, and an Alfisol from Virginia （VA） in the southeastern USA following the application of switchgrass （Panicum virgatum） biochar. The switchgrass biochar was applied at four levels, 0%,0, 2.5%, 5%, and 10%, approximately equivalent to biochar additions of 0, 25, 50, and 100 t ha^-1, respectively, to the soils grown with wheat （Triticum aestivum） in an eight-week growth chamber experiment. We measured wheat shoot biomass and nitrogen （n） content and soil nutrient availability and n mineralization rates, and characterized the microbial fatty acid methyl ester （FAME） profiles of the soils. net n mineralization rates decreased in both soils in proportion to an increase in biochar levels, but the effect was more marked in the VA soil, where net n mineralization decreased from -2.1 to -38.4 mg kg^-1. The 10% biochar addition increased soil pH, electrical conductivity, Mehlich- and bicarbonate-extractable phosphorus （P）, and extractable potassium （K） in both soil types. The wheat shoot biomass decreased from 17.7 to 9.1 g with incremental additions of biochar in the CO soil, but no difference was noted in plants grown in the VA soil. The FAME recovery assay indicated that the switchgrass biochar addition could introduce artifacts in analysis, so the results needed to be interpreted with caution. non-corrected total FAME concentrations indicated a decline by 457o and 34% with 10% biochar addition in the CO and VA soils, respectively, though these differences became nonsignificant when the extraction efficiency correction factor was applied. Incorporation of Biochar Carbon into Stable Soil Aggregates:The Role of Clay Mineralogy and Other Soil Characteristics

A dual-electrolyte system for photoelectrochemical hydrogen generation using CuInS_2-In_2O_3-TiO_2 nanotube array thin film

The utilization of na2S/na2SO3 mixture as the electrolyte solution to stabilize sulfide anode in a photoelectrochemical cell for hydrogen evolution generally compromises the current-to-hydrogen efficiency（ηcurrent） of the system. Here, the employment of a dual-electrolyte system,that is, na2S/na2SO3 mixture and p H-neutral na2SO4 as the respective electrolyte solutions in the anode and cathode chambers of a water splitting cell is demonstrated to suppress the photocorrosion of CuInS2-In2O3-TiO2 nanotube（CISIn2O3-TnT） heterostructure, while simultaneously boosts theηcurrent. Although n-type CIS and In2O3 nanoparticles can be easily formed on TnT array via facile pulse-assisted electrodeposition method, conformal deposition of the nanoparticles homogeneously on the nanotubes wall with preservation of the TnT hollow structure is shown to be essential for achieving efficient charge generation and separation within the heterostructure. In comparison to na2S/na2SO3 solution as the sole electrolyte in both the anode and cathode chambers, introduction of dual electrolyte is shown to not only enhance the photostability of the CIS-In2O3-TnT anode, but also lead to near-unity ηcurrentas opposed to the merely 20% ηcurrentof the single-electrolyte system.

Validation of a radioimmunoassay-based fecal corticosteroid assay for Richardson＇s ground squirrels Urocitellus richardsonii and behavioural correlates of stress

We validated a radioimmunoassay-based method quantifying fecal glucocorticoid metabolites （FGMs） from captive male and female Richardson＇s ground squirrels Urocitellus richardsonii. Blood samples were drawn to explore the correlation between plasma cortisol and FGM concentrations. We also injected groups of squirrels with normal saline （CTL; control）, adre- nocorticotropic hormone （ACTH; stimulating adrenal activity）, or dexamethasone （DEX; suppressing adrenal activity）. Potential correlations between stress and behaviour were explored through quantification of fecal pellet production and the intervention necessary to elicit defecation, as well as the behaviour of subjects in the context of handling. Changes in plasma cortisol concen- tration between capture （baseline）, and following handling （stress-induced） were also quantified for free-living squirrels. While glucocorticoid concentrations recovered from feces during our captive-animal study were not well correlated with plasma eortisol concentrations, and uncorrelated with defecation or behaviour, FGM concentrations did reflect the activation of the hypothalamic- pituitary-adrenal （HPA） axis. FGM concentrations increased significantly during initial captivity, but declined to baseline level as individuals acclimated to the novel environment. Injection of subjects with ACTH increased FGMs above baseline, confirming activation of the HPA axis. Plasma cortisol concentrations increased significantly with induced stress, indicating that capture and handling activated the glucocorticoid stress response even among previously handled, free-living subjects. Our findings validate a non-invasive tool that will afford new insight into the physiological processes underlying social, reproductive and antipredator behaviour of Richardson＇s ground squirrels [Current Zoology 60 （5）： 591-601, 2014 ].

Teaching new Mothers about Infant Feeding Cues May Increase Breastfeeding Duration

The objective of this pilot study was to compare two different methods of educating prenatal women regarding breast-feeding. Comparisons were made between traditional and innovative methods to determine which was more effective in increasing breastfeeding duration. Over a 32 month period, 197 prenatal women were assigned to either a control (C, n = 139) or an experimental (E, n = 51) group. The C group received standard breastfeeding education, while the E group received standard education in addition to information about infant hunger cues. Cox Regression and Kap-lan- Meier analysis were performed. Estimated mean number of weeks for C and E groups to continue breastfeeding was 14.3 + 17.4 weeks and 18.5 + 17.1 weeks, respectively. At 26 weeks, duration of breastfeeding approached significance (chi square = 2.907, df = 1, p = 0.088), indicating probability of continuing to breastfeed was about 28% better for those in E group when compared to C group. Duration of breastfeeding may increase when prenatal women are taught to identify infant behavior such as hunger cues.

Biofilm formation is an important virulence factor of Staphylococcus epidermidis. However, little is known about the mechanisms of staphylococcal biofilm dispersal. In the present study, we investigated biofilm dispersal of the model biofilm-forming strain S. epidermidis RP62A under oligotrophic stress conditions. We found that oligotrophic stress led to rapid dispersal of pre-formed biofilms and concomitant changes in the composition of the extracellular matrix, including a decrease in poly-n-acetylglucosamine polysaccharide and an increase in proteins. Our results suggest that modifications in biofilm integrity caused by compositional changes in the biofilm matrix can induce biofilm dispersal.
