Establishment of a system for screening and identification of novel bactericide targets in the plant pathogenic bacterium Xanthomonas oryzae pv. oryzae using Tn-seq and SPR

Xanthomonas spp. cause severe bacterial diseases. However, effective strategies for prevention and management of these diseases are scarce. Thus, it is necessary to improve the efficiency of control of diseases caused by Xanthomonas. In this study, Xanthomonas oryzae pv. oryzae(Xoo), which causes rice bacterial leaf blight, has been studied as a representative. A transposon insertion library of Xoo, comprising approximately 200,000 individual insertion mutants, was generated. Transposon sequencing data indicated that the mariner C9 transposase mapped at 35.7–36.4% of all potential insertion sites, revealing 491 essential genes required for the growth of Xoo in rich media. The results show that, compared to the functions of essential genes of other bacteria, the functions of some essential genes of Xoo are unknown, 25 genes might be dangerous for the Xanthomonas group, and 3 are specific to Xanthomonas. High-priority candidates for developing broad-spectrum, Xanthomonas-specific, and environment-friendly bactericides were identified in this study. In addition, this study revealed the possible targets of dioctyldiethylenetriamine using surface plasmon resonance(SPR) in combination with high performance liquid chromatography–mass spectrometry(HPLC–MS). The study also provided references for the research of some certain bactericides with unknown anti-bacterial mode of action. In conclusion, this study urged a better understanding of Xanthomonas,provided meaningful data for the management of bacterial leaf blight, and disclosed selected targets of a novel bactericide.

Identification of porcine fast/slow myogenic exosomes and their regulatory effects on lipid accumulation in intramuscular adipocytes

Background Pork quality is affected by the type of muscle fibers, which is closely related to meat color, tenderness and juiciness. Exosomes are tiny vesicles with a diameter of approximately 30–150 nm that are secreted by cells and taken up by recipient cells to mediate communication. Exosome-mediated muscle-fat tissue crosstalk is a newly discovered mechanism that may have an important effect on intramuscular fat deposition and with that on meat quality. Various of adipose tissue-derived exosomes have been discovered and identified, but the identification and function of muscle exosomes, especially porcine fast/slow myotube exosomes, remain unclear. Here, we first isolated and identified exosomes secreted from porcine extensor digitorum longus(EDL) and soleus(SOL), which represent fast and slow muscle, respectively, and further explored their effects on lipid accumulation in longissimus dorsi *** Porcine SOL-derived exosomes(SOL-EXO) and EDL-derived exosomes(EDL-EXO) were first identified and their average particle sizes were approximately 84 nm with double-membrane disc-shapes as observed via transmission electron microscopy and scanning electron microscopy. Moreover, the intramuscular fat content of the SOL was greater than that of the EDL at 180 days of age, because SOL intramuscular adipocytes had a stronger lipid-accumulating capacity than those of the EDL. Raman spectral analysis revealed that SOL-EXO protein content was much greater than that of EDL-EXO. Proteomic sequencing identified 72 proteins that were significantly differentially expressed between SOL-EXO and EDL-EXO, 31 of which were downregulated and 41 of which were upregulated in *** Our findings suggest that muscle-fat tissue interactions occur partly via SOL-EXO promoting adipogenic activity of intramuscular adipocytes.

Highly intelligent Unmanned Combat Aerial Vehicle(UCAV)formation is expected to bring out strengths in Beyond-Visual-Range(BVR)air *** Multi-Agent Reinforcement Learning(MARL)shows outstanding performance in cooperative decision-making,it is challenging for existing MARL algorithms to quickly converge to an optimal strategy for UCAV formation in BVR air combat where confrontation is complicated and reward is extremely sparse and *** to solve this problem,this paper proposes an Advantage Highlight Multi-Agent Proximal Policy Optimization(AHMAPPO)***,at every step,the AHMAPPO records the degree to which the best formation exceeds the average of formations in parallel environments and carries out additional advantage sampling according to ***,the sampling result is introduced into the updating process of the actor network to improve its optimization ***,the simulation results reveal that compared with some state-of-the-art MARL algorithms,the AHMAPPO can obtain a more excellent strategy utilizing fewer sample episodes in the UCAV formation BVR air combat simulation environment built in this paper,which can reflect the critical features of BVR air *** AHMAPPO can significantly increase the convergence efficiency of the strategy for UCAV formation in BVR air combat,with a maximum increase of 81.5%relative to other algorithms.

Screen sensors are the most commonly used human-machine interfaces in our everyday life,which have been extensively applied in personal electronics like *** screen sensors are attracting growing interest due to their distinct advantages of high interaction freedom,comfortability,and hand ***,the material compositions of current touchless screen sensors are rigid and fragile,hardly meeting the needs of wearable and stretchable on-skin electronics ***,these touchless screen sensors are also restricted by high power consumption,limited gesture types of recognition,and the requirement of light ***,we report a stretchable on-skin touchless screen sensor(OTSS)enabled by an ionic hydrogel-based triboelectric nanogenerator(TENG).Compared with current touchless screen sensors,the OTSS is stretchable,self-powered,and competent to recognize diverse gestures by making use of charges naturally carried on fingers without the need of sufficient light *** on-skin noncontact screen operating system is further demonstrated on the basis of the OTSS,which could unlock a cellphone interface in touchless operation mode on the human *** work for the first time introduces the on-skin touchless concept to screen sensors and offers a direction to develop new-generation screen sensors for future cellphones and personal electronics.

Research Progress of PTCH1 Gene in Lung Cancer

Background: The PTCH1 gene, also known as Patched 1, is located on the long arm of human chromosome 9 (9q22.3). It encodes the PTCH1 protein, which is a critical transmembrane receptor within the Hedgehog signaling pathway (Hh), playing a pivotal role in cellular communication and developmental processes. Recent studies have highlighted the significance of mutations in PTCH1 in the pathogenesis of lung cancer, positioning it as a crucial molecule for investigation in oncology. Purpose: This review aims to elucidate the role of the PTCH1 and the Hedgehog pathway in the initiation, progression, and potential treatment of lung cancer, thereby providing a theoretical foundation for personalized and precise therapeutic strategies. Method: To ensure a comprehensive review, this study systematically searched for literature related to the PTCH1, lung cancer, and the Hedgehog pathway across multiple databases including PubMed, Web of Science, and CNKI (China National Knowledge Infrastructure). The search strategy involved using specific keywords and advanced filtering options to include the most relevant and recent studies. Initial screening excluded irrelevant articles, followed by a detailed evaluation of the selected studies based on their scientific quality and relevance. Results: This review indicated that specific mutations in the PTCH1 gene are closely associated with the onset and progression of lung cancer. These mutations impede normal Hedgehog signaling, leading to unregulated cell proliferation and tumor growth. Targeting PTCH1, including vismodegib, have shown efficacy in clinical cases, particularly in SCCL with specific PTCH1 mutations, leading to complete remissions. Furthermore, the interaction between PTCH1 and microRNA-212 suggests potential therapeutic approaches by targeting miRNA to regulate PTCH1 expression. Applications of in-situ spectroscopic techniques towards COelectroreduction

In-situ experimental techniques have been widely applied to uncover the dynamic evolutions of both the structure of catalysts and the interfacial property of catalysis, thus serving as the most important means to gain molecular-level insights into the reaction mechanisms. In this mini review, we summarized recent progress in the applications of the interface-sensitive in-situ Raman and in-situ infrared（IR） spectroscopy towards CO2electroreduction. Specifically, we concentrated on two aspects to clarify the role of both in-situ Raman and in-situ IR in revealing reaction mechanisms of CO2electroreduction. The first one was the in-situ spectroscopy for detecting the active structures. The other one was the in-situ spectroscopy for capturing the reaction intermediates. As powerful guidance for the rational design of catalysts, the reaction mechanism was discussed in the specific examples. Finally, we try to predict the trends for the future development of in-situ spectroscopic techniques towards heterogeneous catalysis.
