Molecular detection of epithelial-mesenchymal transition markers in circulating tumor cells from pancreatic cancer patients:Potential role in clinical practice

AIM To evaluate the clinical properties of three subpopulations of circulating tumor cells(CTCs) undergoing epithelial-mesenchymal transition(EMT) in pancreatic ductal adenocarcinoma(PDAC) *** We identified CTCs for expression of the epithelial cell marker cytokeratin or epithelial cell adhesion molecule(EpCAM)(E-CTC), the mesenchymal cell markers vimentin and twist(M-CTC), or both(E/M-CTC) using the CanPatrol system. Between July 2014 and July 2016, 107 patients with PDAC were enrolled for CTC evaluation. CTC enumeration and classification were correlated with patient clinicopathological features and *** CTCs were detected in 78.5% of PDAC patients. The number of total CTCs ranged from 0 to 26 across all 107 patients, with a median value of six. CTC status correlated with lymph node metastasis, TNM stage, distant metastasis, blood lymphocyte counts, and neutrophil-to-lymphocyte ratio(NLR). Kaplan-Meier survival analysis showed that patients with ≥ 6 total CTCs had significantly decreased overall survival and progression-free survival compared with patients with < 6 total CTCs. The presence of M-CTCs was positively correlated with TNM stage(P < 0.01) and distant metastasis(P < 0.01). Additionally, lymphocyte counts and NLR in patients without CTCs were significantly different from those in patients testing positive for each CTC subpopulation(P < 0.01).CONCLUSION Classifying CTCs by EMT markers helps to identify the more aggressive CTC subpopulations and provides useful evidence for determining a suitable clinical approach.

Morphological and functional changes in the macular area in diabetic macular edema after a single intravitreal injection of aflibercept

Search for an exotic vectorlike X-quark in same-sign dilepton final states at the future 3 TeV CLIC

Vectorlike quarks(VLQs)with masses at the TeV-scale have been predicted in many new physics scenarios beyond the Standard Model(SM).Based on a simplified(X,T)doublet model including the exotic vectorlike X quark(VLQ-X)with an electric charge 5/3,we study the production of the VLQ-X decaying into tW at the future Compact Linear Collider(CLIC)with √s=3 TeV including the initial state radiation and beamstrahlung *** focus on the final signals,including same-sign dileptons(electrons or muons),at least one b-tagged jet,and large missing transverse *** performing detailed signal-to-background analyses and detector simulations,we obtain the 2σexclusion capabilities and discovery reaches,respectively,on the mass of the VLQ-Xmx as well as the relevant parameters(Br(X→_(t)W))for mx≥1500GeV and the coupling strength g^(*) for mx≥1500 GeV)for some typical luminosities at the 3 TeV CLIC.

Comparison and optimization of the method for Cry1Ac protoxin preparation in HD73 strain

Bacillus thuringiensis is one of the most widely used bioinsecticides, and cry gene is the major insecticidal *** Cry1 Ac protein shows strong toxicity against many lepidopteran species, it has been applied widely in spraying products and transgenic *** preparation of Cry protoxin is the first step in the very important processes of understanding the insecticidal mechanism, resistance screening, and biosafety *** media for crystal production and the method for Cry protoxin preparation were varied, however, it was not clear which was better for preparing a larger amount of Cry *** this paper, three media for crystal production and the method for Cry1 Ac protoxin preparation from HD73 strain were compared to find an efficacious way to prepare a large number of Cry1 Ac *** results showed that the 1/2 LB（Luria-Bertani） medium was the ideal medium for crystal production, because the total yield of Cry1 Ac protoxin in 300 m L 1/2 LB medium was（112.38±5.64） mg, the highest one among three media; the repeated crystal solubilization method was better for the preparation of the Cry protoxin comparing with the continuous crystal solubilization *** will be a reference for other Cry protoxin preparation, especially for larger number.

Bacterial fruit blotch caused by Acidovorax citrulli is a serious threat to cucurbit industry *** pathogen is seedtransmitted,so seed detection to prevent distribution of contaminated seed is crucial in disease *** this study,we adapted a quantitative real-time PCR(qPCR)assay to droplet digital PCR(ddPCR)format for *** detection by optimizing reaction *** performance of ddPCR in detecting *** pure culture,DNA,infested watermelon/melon seed and commercial seed samples were compared with multiplex PCR,qPCR,and dilution plating *** lowest concentrations detected(LCD)by ddPCR reached up to 2 fg DNA,and 102 CFU mL–1 bacterial cells,which were ten times more sensitive than those of the *** testing artificially infested watermelon and melon seed,0.1%infestation level was detectable using ddPCR and dilution plating *** 26 positive samples were identified in 201 commercial seed samples through ddPCR,which was the highest positive number among all the *** detection sensitivity achieved by ddPCR demonstrated a promising technique for improving seed-transmitted pathogen detection threshold in the future.

The Influence of Bt-Transgenic Maize Pollen on the Bacterial Diversity in the Midgut of Chinese Honeybees, Apis cerana cerana

Using culture-independent technique polymerase chain reaction-denaturing gradient gel electrophoresis （PCR-DGGE） and conventional culture techniques, ecological risk of transgenic maize pollen on gut bacteria of the Chinese honeybee, Apis cerana cerana, was assessed. Honeybees were fed with Bt-transgenic maize pollen, non-transgenic near isoline pollen, linear crylAh gene （800 ng mL^-1） and supercoiled plasmid DNA （800 ng mL^-1） under laboratory conditions. The DGGE profile showed that the number of DGGE bands varied from 10.7 to 14.7 per sample, and the Shannon＇s index ranged from 0.85 to 1.00. The similarity calculated by PAST was mostly above 92%, indicating no obvious changes among treatments or within replicates. 14 bacterial strains affiliated with Alphaproteobacteria, Gammaproteobacteria, Firmicutes, and Actinobacteria were isolated and characterized on media under aerobic and anaerobic conditions. These results demonstrated that transgenic crylAh maize pollen did not induce significant changes of the honeybee gut bacterial community composition under laboratory conditions.

Surgical treatment of cervicothoracic junction spinal tuberculosis via combined anterior and posterior approaches in children

Background Cervicothoracic junction spinal tuberculosis （CJST） in children is uncommon, especially when accompanied by a huge abscess. However, its consequences can be severe. Because of the special anatomic location of the cervicothoracic junction, surgical treatment is difficult and rarely reported. The aim of this clinical study was to assess the effectiveness of combined anterior and posterior approaches for focal debridement, decompression, allografting and anterior instrumentation in the treatment of CJST in children. Methods Ten pediatric CJST patients underwent focal debridement and cord decompression through combined anterior and posterior approaches. Then an appropriate allograft and titanium plate were applied to reconstruct the spine. The patients were asked to wear head-neck-chest braces for six months and received regular anti-tubercular drugs therapy for 12 months. Results The patients were followed-up for an average of 26 months （range, 15-32 months）. There was no recurrent tuberculous infection. The bone grafts incorporated well and the instrumentation was stable. Cervical and thoracic kyphosis was successfully corrected from 40° （range, 30-52°） before the operation to 18° （range, 12-26°） post-operation. Neurological function was improved in all patients. Conclusions Combined anterior and posterior approaches for focal debridement, decompression, bone allografting and anterior instrumentation provided an effective means of treatment in children of CJST with a huge abscess in the posterior part of the vertebral body.

Ti_(3)AlC_(2)samples are irradiated in advance by 3.5 MeV Fe-ion to the fluence of 1.0×10^(16)ion/cm^(2),and then are implanted by 500 keV He-ion with the fluence of 1.0×10^(17)ion/cm^(2)at room *** irradiated samples are investigated by grazing incidence x-ray diffraction(GIXRD)and transmission electron microscopy(TEM).GIXRD results show serious structural distortion,but without amorphization in the irradiated ***-ion irradiation and He-ion implantation create much more serious structural distortion than single Fe-ion *** results reveal that there are a large number of defect clusters in the damage region,and dense spherical He bubbles appear in the He depositional *** seems that the pre-damage does not influence the growth of He bubbles,but He-ion implantation influences the pre-created defect configurations.
