Most of the human diseases are complex diseases,which could be caused by many genetic pathways.This means that for a given phenotype(i.e.,a complex disease),there are multiple potential genes which could be genomically or epigenetically changed(i.e.,mutations,copy number variations,epigenetic modifications,and so on).Therefore,it is understandable that different individuals who share the same phenotype/diseases may have different causal genes and thus,may have different drug targets.For example,mutated genes are rarely common between the cancer patients of the same cancer type[1];furthermore,for a given drug,only 10%-30% of the patients

The K/HDEL receptor ERD2 mediates the transport of soluble endoplasmic reticulum （ER）-resident proteins containing a C-terminal K/HDEL signal from the Golgi apparatus back to the ER via COPI （COat Protein I）-coated vesicles. Sorting of ERD2 within COPI vesicles is facilitated by p24 proteins. In Arabidop- sis, p24δ5 has been shown to interact directly with ERD2 via its luminal GOLD （GOLgi Dynamics） domain and with COPI proteins via its cytoplasmic C-terminal tail at the acidic pH of the Golgi apparatus. Several members of the p24 family in mammals and yeast have been shown to be glycosylated, but whether Arabidopsis p24 proteins are glycosylated and the role of the sugar moiety in p24 function remain unclear. Here, we show that Arabidopsis p24δ5 protein is N-glycosylated in its GOLD domain. Furthermore, we demonstrate that this post-translational modification is important for its coupled transport with p241β2 at the ER-Golgi interface, for its interaction with the K/HDEL receptor ERD2, and for retrograde transport of ERD2 and K/HDEL ligands from the Golgi apparatus back to the ER.

This comment will review and discuss recent results obtained with the use of calorimetry in assessing molecular information from complex phenomena such as association in surfactant/polymer mixtures.These examples were selected to support the view that it is possible to ally the great sensitivity of modern calorimeters with carefully planned experiments and,sometimes,ancillary techniques in order to derive detailed information on molecular interactions.

Considering the environment of risks and influences inherent in the decision-making process for credit-granting operations,it has become a matter of survival for financial organizations to seek to improve how they engage in effective decision-making to ensure that their returns on invested capital will meet the expectations established at the beginning of the process.A credit-granting sorting model for financial organizations is proposed.The model aggregates the perspectives of different decision-makers to support an organization in the process of credit analysis and,consequently,to improve its operationality.The decision-making model is based on the ELECTRE TRI-B multicriteria method.It sorts credit-granting proposals into three classes,namely credit approved,technical analysis,and credit rejected.The group decision emerges from the decision rules of the organization’s executive board.This new proposed approach to dealing with credit-granting results in the systematization of credit analysis,reduction of doubt among decision-makers,avoidance of the emergence of informal groups,reduction of conflicts within a financial organization,and external interferences.

Influence of variables related to soil weathering on the geomechanical performance of tropical soils

This paper presents an experimental and analytical investigation of the influence of variables related to soil weathering on the geomechanical performance of sand-silt mixtures containing lateritic soils,i.***.intensely weathered tropical soils with the influence of interparticle bonding.The sand-silt mixtures containing different relative proportions between uniform sand and lateritic soil were produced,and geomechanical soil characterization tests were performed.Based on the results,a transition from a primarily coarse-to a fine-grained prevailing soil structure was found to cause considerable impact on the geomechanical performance of these soils,as evidenced by design variables related to soil mineralogy and size distribution characteristics.Specifically,fines contents of both individual soil particles and soil aggregations were found to correlate with experimental results,while the relative proportion between sesquioxides(aluminum,and iron oxides),and silica,i.***.sesquioxide-silica ratios(SSR^(-1)),facilitated estimates concerning changes in geomechanical performance.Finally,the application of the sandsilt mixtures containing lateritic soil on soil walls reinforced with polymeric strips was also evaluated,further emphasizing the potential advantages of adopting variables related to soil weathering on design guidelines concerning tropical soils.

Cholangiocarcinoma(CCA)is characterized by heterogeneous mutations and a refractory nature.Thus,the development of a model for effective drug screening is urgently needed.As the established therapeutic testing models for CCA are often ineffective,we fabricated an enabling three-dimensional(3D)-bioprinted CCA-on-a-chip model that to a good extent resembled the multicellular microenvironment and the anatomical microstructure of the hepato-vascular-biliary system to perform high-content antitumor drug screening.Specifically,cholangiocytes,hepatocytes,and vascular endotheliocytes were employed for 3D bioprinting of the models,allowing for a high degree of spatial and tube-like microstructural control.Interestingly,it was possible to observe CCA cells attached to the surfaces of the gelatin methacryloyl(GelMA)hydrogelembedded microchannels and overgrown in a thickening manner,generating bile duct stenosis,which was expected to be analogous to the in vivo configuration.Over 4000 differentially expressed genes were detected in the CCA cells in our 3D coculture model compared to the traditional two-dimensional(2D)monoculture.Further screening revealed that the CCA cells grown in the 3D traditional model were more sensitive to the antitumoral prodrug than those in the 2D monoculture due to drug biotransformation by the neighboring functional hepatocytes.This study provides proof-of-concept validation of our bioprinted CCA-on-a-chip as a promising drug screening model for CCA treatment and paves the way for potential personalized medicine strategies for CCA patients in the future.

A key goal of knowledge management(KM)is to foster innovation through the ceation of new knowledge.Surprisingly,there is little research on KM in the context of entrepre-neurship,a domain where innovation is considered essential.This paper extends estab-lished theories and frameworks for KM to the understudied context of KM in small and medium entrepreneurial firms,particularly young startups.Translating KM fr ameworks,such as the widely studied SECI model,into the entrepreneurial domain could eventually help startups in their quest for sustainability and growth.The SECI model is an aspir ational process with which to build new knowledge,including the explicit knowledge assets that startups commonly lack.However,an aspiration is only useful if the subject knows where he or she stands in relation to it.This study focuses on understanding whether entre-preneurial activities embed into,and can further enable,the"virtuous"knowledge-creation cycle.The secondary objective of this paper is to acknowledge Ikujiro Nonaka's contribution to western and eastern KM theories,and extend his seminal theories into other domains,such as small and medium enterprises.This work pilots the use of a content analysis technique largely used in psychology to analyze the connection between entrepreneurship and KM.With this method,the study highlights how the four phases of the SECI model apply to startup firms in a business incubator in the United States.It provides insights into the knowledge-creation process of entrepreneurs,and suggests how entrepreneurs can improve startup survival through greater awareness and use of KM in their business planning and operational activities.

Split-root system（SRS） approaches allow the differential treatment of separate and independent root systems, while sharing a common aerial part. As such, SRS is a useful tool for the discrimination of systemic（shoot origin）versus local（root/nodule origin） regulation mechanisms. This type of approach is particularly useful when studying the complex regulatory mechanisms governing the symbiosis established between legumes and Rhizobium bacteria. The current work provides an overview of the main insights gained from the application of SRS approaches to understand how nodule number（nodulation autoregulation） and nitrogen fixation are controlled both under non-stressful conditions and in response to a variety of stresses. Nodule number appears to be mainly controlled at the systemic level through a signal which is produced by nodule/root tissue, translocated to the shoot, and transmitted back to the root system, involving shoot Leu-rich repeat receptor-like kinases. In contrast, both local and systemic mechanisms have been shown to operate for the regulation of nitrogenase activity in nodules. Under drought and heavy metal stress, the regulation is mostly local,whereas the application of exogenous nitrogen seems to exert a regulation of nitrogen fixation both at the local and systemic levels.

Carbamazepine(CBZ),as one of the most frequently detected pharmaceuticals,is of great concern due to its potential impact on the ecosystem and human health.This study provides an effective approach to remove CBZ by using photocatalyst silver phosphate combined with graphene oxide(Ag_(3)PO_(4)/GO)under visible irradiation.The morphology,composition,and optical properties of Ag_(3)PO_(4)/GO were characterized employing SEM,XRD,and DRS.Graphene oxide could improve the visible-light utilization and promote electron's charge to enhance the photocatalytic performance of Ag_(3)PO_(4)/GO.With the optimal reaction condition of 5.86 mW/cm_(2)light intensity,15-25℃ temperature,5-7 pH,and 0.5 mg/L catalytic dosages,5 mg/L CBZ could be completely degraded in 30 min,and the apparent rate constant could reach 0.12 min^(-1).Additionally,the radical trapping experiments indicated·OH and O_(2)^(-)· were the main reactive oxygen species employed to eliminate CBZ.The decay pathways of CBZ had been proposed accordingly,and the main product was the low-molecular products.
