Saikosaponin D improves nonalcoholic fatty liver disease via gut microbiota-bile acid metabolism pathway

Non-alcoholic fatty liver disease(NAFLD)is the main cause of chronic liver disease *** is widely used in the treatment of non-alcoholic fatty liver,and saikosaponin D(SSD)is one of the main active components of *** purpose of this study was to investigate the efficacy of SSD in the treatment of NAFLD and to explore the mechanism of SSD in the improvement of NAFLD based on“gut-liver axis”.Our results showed that SSD dose-dependently alleviated high fat diet-induced weight gain in mice,improved insulin sensitivity,and also reduced liver lipid accumulation and injury-related biomarkers aspartate aminotransferase(AST)and alanine aminotransferase(ALT).Further exploration found that SSD inhibited the mRNA expression levels of farnesoid X receptor(Fxr),small heterodimer partner(Shp),recombinant fibroblast growth factor 15(Fgf15)and apical sodium dependent bile acid transporter(Asbt)in the intestine,suggesting that SSD improved liver lipid metabolism by inhibiting intestinal FXR *** can significantly reduce the gut microbiota associated with bile salt hydrolase(BSH)expression,such as *** BSH expression reduced the ratio of unconjugated to conjugated bile acids,thereby inhibiting the intestinal *** data demonstrated that SSD ameliorated NAFLD potentially through the gut microbiota-bile acidintestinal FXR pathway and suggested that SSD is a promising therapeutic agent for the treatment of NAFLD.

Although the functions of metabolic enzymes and nuclear receptors in controlling physiological homeostasis have been established, their crosstalk in modulating metabolic disease has not been *** ablation of the xenobiotic-metabolizing cytochrome P450 enzyme CYP2 E1 in mice markedly induced adipose browning and increased energy expenditure to improve obesity. CYP2 E1 deficiency activated the expression of hepatic peroxisome proliferator-activated receptor alpha(PPARa) target genes,including fibroblast growth factor(FGF) 21, that upon release from the liver, enhanced adipose browning and energy expenditure to decrease obesity. Nineteen metabolites were increased in Cyp2 e1-null mice as revealed by global untargeted metabolomics, among which four compounds, lysophosphatidylcholine and three polyunsaturated fatty acids were found to be directly metabolized by CYP2 E1 and to serve as PPARa agonists, thus explaining how CYP2 E1 deficiency causes hepatic PPARa activation through increasing cellular levels of endogenous PPARa agonists. Translationally, a CYP2 E1 inhibitor was found to activate the PPARa-FGF21-beige adipose axis and decrease obesity in wild-type mice, but not in liver-specific Pparanull mice. The present results establish a metabolic crosstalk between PPARa and CYP2 E1 that supports the potential for a novel anti-obesity strategy of activating adipose tissue browning by targeting the CYP2 E1 to modulate endogenous metabolites beyond its canonical role in xenobiotic-metabolism.

BACKGROUND Despite continuous changes in treatment methods,the survival rate for advanced hepatocellular carcinoma(HCC)patients remains low,highlighting the importance of diagnostic methods for *** To explore the efficacy of texture analysis based on multi-parametric magnetic resonance(MR)imaging(MRI)in predicting microvascular invasion(MVI)in preoperative *** This study included 105 patients with pathologically confirmed HCC,categorized into MVI-positive and MVI-negative *** employed Original Data Analysis,Principal Component Analysis,Linear Discriminant Analysis(LDA),and Non-LDA(NDA)for texture analysis using multi-parametric MR images to predict preoperative *** effectiveness of texture analysis was determined using the B11 program of the MaZda4.6 software,with results expressed as the misjudgment rate(MCR).RESULTS Texture analysis using multi-parametric MRI,particularly the MI+PA+F dimensionality reduction method combined with NDA discrimination,demonstrated the most effective prediction of MVI in *** accuracy in the pulse and equilibrium phases was 83.81%.MCRs for the combination of T2-weighted imaging(T2WI),arterial phase,portal venous phase,and equilibrium phase were 22.86%,16.19%,20.95%,and 20.95%,*** area under the curve for predicting MVI positivity was 0.844,with a sensitivity of 77.19%and specificity of 91.67%.CONCLUSION Texture analysis of arterial phase images demonstrated superior predictive efficacy for MVI in HCC compared to T2WI,portal venous,and equilibrium *** study provides an objective,non-invasive method for preoperative prediction of MVI,offering a theoretical foundation for the selection of clinical therapy.

Turbulence Statistics of Thermo-Buoyancy Supercritical Fuel Flow in a Regenerative Cooling Channel

Output feedback stabilization of stochastic high-order planar nonlinear systems with stochastic inverse dynamics and output-constraint

In this paper, we solve the output feedback control problem of stochastic high-order planar nonlinear systems with output constraint and stochastic integral input-to-state stability(SiISS) inverse dynamics. By employing a key coordinate transformation, a stochastic nonlinear system with output constraint and SiISS inverse dynamics is converted into an unconstrained system. By skillfully constructing an observer and adopting SiISS small-gain conditions, we develop a new output feedback control design and analysis method, and prove that all the closed-system signals are bounded almost surely, the output constraint is not violated almost surely, and the equilibrium point of the closed-loop system is stochastically asymptotically stable.

Studies on the effects of bio-organic fertilizer on the physical and chemical characteristics of soil and the yield of cassava＊

Bio-organic fertilizer is a new type of fertilizer which have advantages of both organic manure and fertilizer. This study investigated the effects of bio-organic fertilizer on the growth and yield of cassava and the soil fertility. The study was carried out in the period of 2004-2005 and the material was cassava cultivar FUXUAN01. The bio-organic fertilizers were applied as basic fertilizers on four different levels of 450 kg/hm^2, 600 kg/hm^2, 750 kg/hm^2, 900 kg/hm^2 in this experiment. The growth of stem and leaf, the yield of earthnut and the starch content of tuber root of cassava and the unit weight, the hole percent, the content of organic matters, nitrogen, phosphorus and potassium, microbes, the activity of soil urease and invertase were analyzed during the experiment. The results showed that not only can the bio-organic fertilizer promote the growth of cassava stems and leaves, increase the chlorophyll content and the photosynthesis of leaves, improve the physiological metabolism of cassava, and strengthen physiological function of anti-senility, promote the transformation from photosynthetic organism to tuber root and increase the yield and starch content in the tuber root of cassava, but also decrease the soil unit weight, increase the hole percent of soil, promote microbe activity in the soil, increase the activity of soil urease and invertase, promote the availability of nutrients, increase the content of organic matters, available nitrogen, phosphorus and potassium, and increase the utilization rate of fertilizer. It was an effective way to apply the bio-organic fertilizer to increase the yield and starch content in the tuber root of cassava, improving the physical and chemical characters of soil and increasing the soil fertility.

Fixed-time stabilization of output-constrained stochastic high-order nonlinear systems

In this study, the fixed-time stabilization problem of stochastic high-order nonlinear systems with output constraint and high-order and low-order nonlinearities is addressed. A new coordinate transformation is employed to directly convert output-constrained stochastic systems into an equivalent unconstrained *** fully extracting the characteristics of system nonlinearities and using the stochastic fixed-time stability theorem, a new design and analysis method is constructed to guarantee that the trivial solution of the closed-loop system is stochastically fixed-time stable while fulfilling the output constraint.
