TNF-α Induces Transient Resistance to Fas-Induced Apoptosis in Eosinophilic Acute Myeloid Leukemia Cells

Tumor necrosis factor α （TNF-α） has been recognized as an activator of nuclear factor kB （NF-kB）, a factor implicated in the protection of many cell types from apoptosis. We and others have presented evidence to suggest that Fas-induced apoptosis may be an important aspect of the resolution of inflammation, and that delayed resolution of inflammation may be directly associated with NF-kB-dependent resistance to Fas. Because TNF-α activates NF-kB in many cell types including inflammatory cells such as eosinophils, we examined effects of TNF-α signaling on the Fas-mediated killing of an eosinophilic cell line AML14. While agonist anti-Fas （CHII） treatment induced apoptosis in AML14 cells, no significant cell death occurred in response to TNF-α alone. Electrophoretic mobility shift assay （EMSA） revealed that TNF-α induced NF-kB transactivation in AMLI4 cells in a time- and dose-dependent fashion, and subsequent supershift assays indicated that the translocated NF-kB was the heterodimer p65 （RelA）/p50. Pre-treatment of cells with TNF-α dramatically decreased the CHll-induced cell death in a transient fashion, accompanied by suppression of activation of caspase-8 and caspase-3 activation. Inhibition of NF-kB transactivation by inhibitors, BAY 11-7085 and parthenolide, reversed the suppression of Fas-mediated apoptosis by TNF-α. Furthermore, TNF-α up-regulated X-linked inhibitor of apoptosis protein （XIAP） transiently and XIAP levels were correlated with the temporal pattern of TNF-α protection against Fas-mediated apoptosis. This finding suggested that TNF-α may contribute to the prolonged survival of inflammatory cells by suppression of Fas-mediated apoptosis, the process involved with NF-kB transactivation, anti-apoptotic XIAP up-regulation and caspase suppression. Does a large prostate size,small lesion volume,or long needle distance from the probe to the lesion reduce magnetic resonance imaging–targeted cancer detection?

Background:We aimed to evaluate whether large prostate size,small lesion volume,or long lesion distance from the ultrasound probe tip would decrease cancer detection in transrectal magnetic resonance imaging(MRI)–targeted *** and methods:Patients who underwent MRI-targeted biopsy at our institution between May 2017 and August 2019 were enrolled in a prospective *** to 5 cores were obtained from≥2 prostate imaging reporting and data system v2 lesions.A multivariable model was created that included needle distance to the lesion,prostate specific antigen,prostate imaging reporting and data system,lesion volume,and prostate ***:A total of 377 patients with 533 lesions underwent a biopsy during the study period.A total of 233(44%)lesions were positive for prostate cancer,and 173(32%)lesions had clinically significant prostate *** mean needle distance to the lesion was 11.7mm(interquartile range,7.6–15.5mm).The likelihood of obtaining a positive core on biopsy decreased as the distance fromthe ultrasound probe increased for all prostate cancers and clinically significant prostate cancer(p=0.018 and p=0.004,respectively).Every 10mmfrom the rectum,there was an 8%–10%decrease in the rate of cancer ***,as the prostate volume increased,the odds of obtaining a positive core also decreased(p=0.039).There was no significant association between the lesion size and amount of cancer obtained on ***:Our data showed that transrectal MRI-targeted biopsy cancer detection modestly decreased the lesion from the ultrasound probe and with a large prostate volume but could not prove that lesion volume was a significant predictor of the amount of cancer detected.

Introduction:Numerical models are critical for assessing the effects of sea level rise(SLR),hurricanes,and storm surge on vegetation change in the Everglades National *** model must be capable of representing short-timescale hydrodynamics,salinity transport,and groundwater ***,there is also a strong need to adapt these numerical models to hindcast past conditions in order to examine long-term effects on the distribution of vegetation that cannot be determined using only the modern ***:Based on parameters developed for a numerical model developed for the recent 1996 to 2004 period,a hindcast model was developed to represent sea level and water management for the period of 1926 to 1932,constrained by the limited hydrology and meteorology data available from the historic *** hurricanewind and storm surge representations,required for the hindcast model,are based on information synthesized from modern storm data.A series of simulation scenarios with various hurricane representations inserted into both hindcast and recent numerical models were used to assess the utility of the storm representation in the model and compare the two ***:The comparison of the hindcast and recent models showed differences in the hydrology patterns that are consistent with known differences in water delivery systems and sea level rise.A 30×lower-resolution spatially variable wind grid for the hindcast produced similar results to the original high-resolution full wind grid representation of the recent *** effects on hydrologic patterns demonstrated with the simulations show hydrologic processes that could have a long-term effect on vegetation ***:The hindcast simulation estimated hydrologic processes for the 1926 to 1932 *** shows promise as a simulator in long-term ecological studies to test hypotheses based on theoretical or empirical-based studies at larger landscape scales.

Evolution and predictive factors of thyroid disorder due to interferon alpha in the treatment of hepatitis C

AIM： To study predictive factors of thyroid dysfunction associated with interferon-alpha （IFNa） therapy in chronic hepatitis C （CHC） and to describe its long-term evolution in a large population without previous thyroid dysfunction. METHODS： We performed a follow-up of thyroid function and detection of thyroid antibodies in 301 patients treated for CHC with IFNα from 1999 to 2004. RESULTS： Thyroid disorder developed in 30/301 （10%） patients with a mean delay of 6 ± 3.75 mo： 13 patients had hyperthyroidism, 11 had hypothyroidism, and 6 had biphasic evolution. During a mean follow-up of 41.59 ± 15.39 mo, 9 patients with hyperthyroidism, 3 with hypothyroidism, and 4 with biphasic evolution normalized thyroid function in 7.88 ± 5.46 mo. Recovery rate of dysthyroidism was not modified by treatment discontinuation, but was better for patients with negative thyroid antibodies before antiviral treatment （P = 0.02）. Women had significantly more dysthyroidism （P = 0.05）. Positive thyroid peroxidase and thyroglobulin antibodies were more frequent before antiviral treatment in patients who developed dysthyroidism （P 〈 0.0003 and P = 0.0003, respectively）. In a multivariate model, low fibrosis was found to be a predictive factor of dysthyroidism （P = 0.039）.CONCLUSION： In this monocentric population of CHC, dysthyroidism, especially hyperthyroidism, developed in 10% of patients, Low fibrosis was found to be a predictive factor of dysthyroidism, Thyroid disorder recovered in 16/30 patients （53%） and recovery was better in the non-autoimrnune form,

Phylogeny and biogeography of Maianthemum(Asparagaceae:Nolinoideae)revisited with emphasis on its divergence pattern in SW China

Maianthemum is a genus with more than 35 species from the tribe Polygonateae(Asparagaceae),widely distributed between North to Central Americas and eastern Asia with high diversity in the easternHimalayas to the Hengduan Mountains of SW *** most species from SW China form a well-supported clade,phylogenetic relationships within this clade remain *** a broad level of taxon sampling and an extensive character sampling fromeight DNA regions,this study intends to revisit the phylogeny and biogeography of the genus to better understand the divergence patterns of species from SW *** results suggested the monophyly of Maianthemum with recognition of nine strongly supported clades,but backbone relationships among these clades remained largely *** the SWChina clade,individuals from the same species are grouped into different *** results revealed that the fast radiation of the SWChina cladewas occurred in the eastern Himalayas,followed by subsequent radiation in the Hengduan Mountains in the *** disjunctions of Maianthemum in the Northern Hemisphere appear to have occurred multiple times during the late Miocene to the Pliocene,likely resulted by a combination of both vicariance and long-distance dispersal events.

Liver transplantation for hepatocellular carcinoma

Hepatocellular carcinoma (HCC) is the commonest primary malignancy of the liver. It usually occurs in the setting of chronic liver disease and has a poor prognosis if untreated. Orthotopic liver transplantation (OLT) is a suitable therapeutic option for early,unresectable HCC particularly in the setting of chronic liver disease. Following on from disappointing initial results,the seminal study by Mazzaferro et al in 1996 established OLT as a viable treatment for HCC. In this study,the "Milan criteria" were applied achieving a 4-year survival rate similar to OLT for benign disease. Since then various groups have attempted to expand these criteria whilst maintaining long term survival rates. The technique of living donor liver transplantation has evolved over the past decade,particularly in Asia,and published outcome data is comparable to that of OLT. This article will review the evidence,indications,and the future direction of liver transplantation for liver cancer.
