Thanks to tremendous advances in sequencing technologies and in particular to whole exome sequencing(WES),many genes have now been linked to severe sperm defects.A precise genetic diagnosis is obtained for a minority of patients and only for the most severe defects like azoospermia or macrozoospermia which is very often due to defects in the aurora kinase C(AURKC)***,we studied a subject with a severe oligozoospermia and a phenotypic diagnosis of *** analysis did not reveal any deleterious *** was then initiated which permitted to identify a homozygous loss of function variant in the zinc finger MYND-type containing 15(ZMYND15)***15 has been described to serve as a switch for haploid gene expression,and mice devoid of ZMYND15 were shown to be sterile due to nonobstructive azoospermia(NOA).In man,ZMYND15 has been associated with NOA and severe *** confirm here that the presence of a bi-allelic ZMYND15 variant induces a severe *** addition,we show that severe oligozoospermia can be associated macrozoospermia,and that a phenotypic misdiagnosis is possible,potentially delaying the genetic *** conclusion,genetic defects in ZMYND15 can induce complete NOA or severe oligozoospermia associated with a very severe *** our experience,severe oligozoospermia is often associated with severe teratozoospermia and can sometimes be misinterpreted as macrozoospermia or *** these instances,specific AURKC or dpy-19 like 2(DPY19L2)diagnosis is usually negative and we recommend the direct use of a pan-genomic techniques such as WES.

Effect of thermo-mechanical conditions during constrained friction processing on the particle refinement of AM50 Mg-alloy phases

Constrained friction Processing(CfP)is a novel solid-state processing technique suitable for lightweight materials,such Mg-and *** technique enables grain size refinement to fine or even ultrafine *** this study,the effect of CfP on the microstructural refinement of AM50 rods is investigated in terms of particle size and morphology of the eutectic and secondary phases originally present in the base material,in particular the eutecticβ-Mg_(17)Al_(12)and Al-Mn *** that purpose,as-cast and solution heat-treated base material and processed samples were *** Al_(8)Mn_(5) intermetallic phase was identified as the main secondary phase present in all samples before and after the processing.A notorious refinement of these particles was observed,starting from particles with an average equivalent length of a few micrometers to around 560 nm after the *** refinement of the secondary phase refinement is attributed to a mechanism analogous to the attrition comminution,where the combination of temperature increase and shearing of the material enables the continuous breaking of the brittle intermetallic particles into smaller *** for the eutectic phase,the results indicate the presence of the partially divorcedβ-Mg_(17)Al_(12)particles exclusively in the as-cast base material,indicating that no further phase transformations regarding the eutectic phase,such as dynamic precipitation,occurred after the *** the case of the processed as-cast material analyzed after the CfP,the thermal energy generated during the processing led to temperature values above the solvus limit of the eutectic phase,which associated with the mechanical breakage of the particles,enabled the complete dissolution of this ***,CfP was successfully demonstrated to promote an extensive microstructure refinement in multiple aspects,in terms of grain sizes of theα-Mg phase and presence and morphology of the Al-Mn and eutecticβ-Mg_(17)Al_(12).

Introduction:The state of natural resources is greatly influenced by market ***,resource trader's incentives,decisions,and willingness to comply with manage-ment an influence efforts to achieve sustainabi ***'s impacts will depend on their economic niches,which are influenced by cultural norms,skill,social relationshi ps,profitability,and the spatial scale of ***,we examined the potential of traders to influence fisheries'sustainability by evaluating their jobs,gender roles,religion,socioeconomic status,association and perceptions of management systems,and future *** studied 142 traders in 19 Kenyan coral reef fisheries landing sites distributed among four gear management ***:We found a strong role of gender,geography,and religion in the participation of these fisheries that was primarily driven by fisheries'*** associations suggest that overfished fisheries should retain traders with low education,capital,and savings-often women;whereas sustainable stocks favor the opposite characteristics,and often ***:Therefore,managing for increasing yields,profits,and sustainability could exclude women traders unless they successfully access or adopt the more traditional male economic *** coexistence is most likely to be achieved by managing for inter-mediate resource levels where net production,catch,and fish body size diversity are ***,reducing risk and increasing the capital and mobility of women traders should reduce their chances of exclusion when fisheries are sustainable.

Endometrial mesenchymal stem cells as a cell based therapy for pelvic organ prolapse

Pelvic organ prolapse(POP) occurs when the pelvic organs(bladder, bowel or uterus) herniate into the vagina, causing incontinence, voiding, bowel and sexual dysfunction, negatively impacting upon a woman's quality of life. POP affects 25% of all women and results from childbirth injury. for 19% of all women, surgical reconstructive surgery is required for treatment, often augmented with surgical mesh. The surgical treatment fails in up to 30% of cases or results in adverse effects, such as pain and mesh erosion into the bladder, bowel or vagina. Due to these complications the food and Drug Administration cautioned against the use of vaginal mesh and several major brands have been recently been withdrawn from market. In this review we will discuss new cell-based approaches being developed for the treatment of POP. Several cell types have been investigated in animal models, including a new source of mesenchymal stem/stromal cells(MSC) derived from human endometrium. The unique characteristics of endometrial MSC, methods for their isolation and purification and steps towards their development for good manufacturing practice production will be described. Animal models that could be used to examine the potential for this approach will also be discussed as will a rodent model showing promise in developing an endometrial MSC-based therapy for POP. The development of a preclinical large animal model for assessing tissue engineering constructs for treating POP will also be mentioned.

Calcium Signaling during Reproduction and Biotrophic fungal Interactions in Plants

Many recent studies have indicated that cellular communications during plant reproduction, fungal invasion, and defense involve identical or similar molecular players and mechanisms. Indeed, pollen tube invasion and sperm release shares many common features with infection of plant tissue by fungi and oomycetes, as a tip-growing intruder needs to communicate with the receptive cells to gain access into a cell and tissue. Depending on the comPatibility between cells, interactions may result in defense, inva- sion, growth support, or cell death. Plant cells stimulated by both pollen tubes and fungal hyphae secrete, for example, small cysteine-rich proteins and receptor-like kinases are activated leading to intracellular signaling events such as the production of reactive oxygen species （ROS） and the generation of calcium （Ca^2＋） transients. The ubiquitous and versatile second messenger Ca^2＋ thereafter plays a central and crucial role in modulating numerous downstream signaling processes. In stimulated cells, it elicits both fast and slow cellular responses depending on the shape, frequency, amplitude, and duration of the Ca^2＋ transients. The various Ca^2＋ signatures are transduced into cellular information via a battery of Ca^2＋-binding proteins. In this review, we focus on Ca^2＋ signaling and discuss its occurrence during plant reproduction and interactions of plant cells with biotrophic filamentous microbes. The participation of Ca^2＋ in ROS signaling pathways is also discussed.

Histone Acetyltransferase AtGCN5/HAG1 Is a Versatile Regulator of Developmental and Inducible Gene Expression in Arabidopsis

Histone acetylation/deacetylation is a dynamic process and plays an important role in gene regulation. Histone acetylation homeostasis is regulated by antagonist actions of histone acetyltransferases （HAT） and deacetylases （HDAC）. Plant genome encodes multiple HATs and HDACs. The Arabidopsis HAT gene AtGCNS/HAGlplays an essential role in many plant development processes, such as meristem function, cell differentiation, leaf and floral organogenesis, and responses to environmental conditions such as light and cold, indicating an important role of this HAT in the regulation of both long-term developmental switches and short-term inducible gene expression. AtGCN5 targets to a large number of promoters and is required for acetylation of several histone H3 lysine residues. Recruitment of AtGCN5 to target promoters is likely to be mediated by direct or indirect interaction with DNA-binding transcription factors and/or by interaction with acetylated histone lysine residues on the targets. Interplay between AtGCN5 and other HATand HDAC is demonstrated to control specific regulatory pathways. Analysis of the role of AtGCN5 in light-inducible gene expression suggests a function of AtGCN5 in preparing chromatin commitment for priming inducible gene activation in plants.

Synthesis and purification of long copper nanowires. Application to high performance flexible transparent electrodes with and without PEDOT：PSS

We demonstrate the hydrothermal synthesis of long copper nanowires based on a simple protocol. We show that the purification of the nanowires is very important and can be achieved easily by wet treatment with glacial acetic acid. fabrication of random networks of purified copper nanowires leads to flexible transparent electrodes with excellent optoelectronic performances （e.g., 55 Ω/sq. at 94% transparency）. The process is carried out at room temperature and no post-treatment is necessary. Hybrid materials with the conductive polymer PEDOT：PSS show similar properties （e.g., 46 Ω/sq, at 93% transparency）, with improved mechanical properties. Both electrodes were integrated in capacitive touch sensors.

The recalcitrance of pathogens to traditional antibiotics has made treating and eradicating bacterial infections more *** this regard,developing new antimicrobial agents to combat antibiotic-resistant strains has become a top *** peptides(AMPs),a ubiquitous class of naturally occurring compounds with broadspectrum antipathogenic activity,hold significant promise as an effective solution to the current antimicrobial resistance(AMR)*** AMPs have been identified and evaluated for their therapeutic application,with many already in the drug development *** distinct properties,such as high target specificity,potency,and ability to bypass microbial resistance mechanisms,make AMPs a promising alternative to traditional ***,several challenges,such as high toxicity,lability to proteolytic degradation,low stability,poor pharmacokinetics,and high production costs,continue to hamper their clinical ***,recent research has focused on optimizing the properties of AMPs to improve their *** understanding the physicochemical properties of AMPs that correspond to their activity,such as amphipathicity,hydrophobicity,structural conformation,amino acid distribution,and composition,researchers can design AMPs with desired and improved *** this review,we highlight some of the key strategies used to optimize the performance of AMPs,including rational design and de novo *** also discuss the growing role of predictive computational tools,utilizing artificial intelligence and machine learning,in the design and synthesis of highly efficacious lead drug candidates.
