Antifungal Activity against <i>Aspergillus parasiticus</i>of Supernatants from Whey Permeates Fermented with Kefir Grains

Aspergillus parasiticus, a common fungal contaminant in food, produces aflatoxin B1, which is classified as human carcinogen. Kefir is an ancient fermented beverage obtained by the fermentation of different substrates with kefir grains. A very important waste produced by the dairy cheese industry is the whey permeate, which nowadays is a strong ambient contaminant. The aim of this work was to assess the effect of whey permeates fermented with kefir grains against A. parasiticus growth, aflatoxin B1 biosynthesis, and the kefir microorganisms protection against the cell damage produced by aflatoxin B1. It was observed that kefir-cell-free-supernatants (CFS) produced fungal inhibition. A fungicidal effect was observed with 65% v/v of CFS in the culture medium (final pH 4.55 and total undissociated lactic and acetic acid concentration 34.08 mM). Under these conditions, aflatoxin production was not detected. Finally, it was found that non-viable kefir microorganisms protected HepG2 cells from the damage produced by aflatoxin B1.

BACKGROUND A progressive decrease in exclusive breastfeeding(BF)is observed in Latin America and the Caribbean compared with global *** possibility of being breastfed and continuing BF for>6 months is lower in low birth weight than in healthy-weight *** To identify factors associated with BF maintenance and promotion,with particular attention to low-and middle-income countries,by studying geographic,socioeconomic,and individual or neonatal health *** A scoping review was conducted in 2018 using the conceptual model of social determinants of health published by the Commission on Equity and Health Inequalities in the United *** extracted data with common characteristics were synthesized and categorized into two main themes:(1)Sociodemographic factors and proximal determinants involved in the initiation and maintenance of BF in low-birth-weight term infants in Latin America;and(2)individual characteristics related to the self-efficacy capacity for BF maintenance and adherence in low-birth-weight term *** This study identified maternal age,educational level,maternal economic capacity,social stratum,exposure to BF substitutes,access to BF information,and quality of health services as mediators for maintaining *** Individual self-efficacy factors that enable BF adherence in at-risk populations should be analyzed for better health outcomes.

Scavenging of reactive oxygen and nitrogen species with nanomaterials

Reactive oxygen and nitrogen species （RONS） are essential for normal physiological processes and play important roles in cell signaling, immunity, and tissue homeostasis. However, excess radical species are implicated in the development and augmented pathogenesis of various diseases. Several antioxidants may restore the chemical balance, but their use is limited by disappointing results of clinical trials. Nanoparticles are an attractive therapeutic alternative because they can change the biodistribution profile of antioxidants, and possess intrinsic ability to scavenge RONS. Herein, we review the types of RONS, how they are implicated in several diseases, and the types of nanoparticles with inherent antioxidant capability, their mechanisms of action, and their biological applications.

An integrated approach,involving land use patterns and heavy metal contents of the surficial and cored sediment,was proposed to detect the characteristic spatial and temporal scale of point and non-point source human disturbances on the Oualidia lagoon in *** and description of the temporal and spatial distributions of the main contamination sources of heavy metals are analyzed using statistics and GIS *** results show that surficial sediments are highly enriched in heavy metals compared to their preindustrial background ***-use types affected lagoon sediment pollution in different ways:1) agricultural areas had the highest potential for sediment contamination by heavy metals,particularly Pb,Cu and Hg,2) ni and Cr are the main pollutants originating from urban sewage and 3) oyster farming and traditional land uses such as salt flats,pastures showed low levels of others *** enrichment factors(normalized by Al) obtained from the sediment cores indicated that the lagoon is(strongly/moderately) polluted by all metals which were attributed to agricultural activities and wastewater discharges from urbanized areas.

Traffic is a main source of air pollutants in urban areas and consequently daily peak exposures tend to occur during *** exposure to particulate matter(PM)was monitored while cycling and travelling by bus,car and metro along an assigned route in Lisbon(Portugal),focusing on PM_(2.5) and PM_(1)0(PM with aerodynamic diameterni and K were correlated,evidencing their common traffic-related *** average,the highest inhaled dose of PM_(2.5) was observed while cycling(55μg),and the lowest in car travels(17μg).Cyclists inhaled higher doses of PM_(2.5) due to both higher inhalation rates and longer journey times,with a clear enrichment in mineral *** presented results evidence the importance of considering the transport mode in exposure assessment studies.

Potential adaptation of a Q biotype whitefly population from poinsettia to field crops

The invasive Q biotype whitefly was first detected in the US on poinsettia in 2004 and is still not a pest outside of greenhouse environments in the US. To assess the potential for the establishment of the Q biotype on field crops, population cage experiments were conducted to compare the performance of a poinsettia-derived Q population named P＇06 on poinsettia and six field crops （alfalfa, tomato, melon, cotton, cowpea and cabbage）. P＇06 adults reared on poinsettia as nymphs laid eggs on all six field crops. Significantly more eggs were laid on alfalfa, tomato, melon and cotton than on cabbage, cowpea and poinsettia. These eggs hatched and the nymphs developed to adults on the six field crops. Relative to poinsettia, whitefly survival was similar on cowpea, alfalfa, tomato and cabbage, but significantly higher on cotton and melon. Moreover, P＇06 had significantly shorter development times from egg to adult on cotton, melon, cowpea, tomato and alfalfa than they did on poinsettia. However, the F1 adults raised on the six field crops had significantly shorter lifespans and laid 11- to 18-fold fewer eggs than did the F1 adults raised on poinsettia. Taken together, while P＇06 may have some potential to establish on field crops, the shorter lifespans and extremely low fectmdities of the F1 adults raised on the six field crops suggests that P＇06 is incapable of rapidly adapting to them. Poor adaptation to field crops may explain, at least partially, why the Q biotype has not established in the US field system.

Threonine 32 （Thr32） of FoxO3 is critical for TGF-β-induced apoptosis via Bim in hepatocarcinoma cells

Transforming growth factor-β （TGF-β） exerts apoptotic effects on various types of malignant ceils, including liver cancer cells. However, the precise mechanisms by which TGF-β induces apoptosis remain poorly known. In the present study, we have showed that threonine 32 （Thr32） residue of FoxO3 is critical for TGF-β to induce apoptosis via Bim in hepatocarcinoma Hep3B cells. Our data demonstrated that TGF-βinduced FoxO3 activation through specific de-phosphorylation at Thr32. TGF-β-activated FoxO3 cooperated with Smad2/ 3 to mediate Bim up-regulation and apoptosis. FoxO3 （de）phosphorylation at Thr32 was regulated by casein kinase I-ε （CKI-E）. CKI inhibition by small molecule D4476 could abrogate TGF-β-induced FoxO/Smad acti- vation, reverse Bim up-regulation, and block the sequential apoptosis. More importantly, the deregulated levels of CKI-ε and p32FoxO3 were found in human malignant liver tissues. Taken together, our findings suggest that there might be a CKI-FoxO/Smad-Bim engine in which Thr32 of FoxO3 is pivotal for TGF-β- induced apoptosis, making it a potential therapeutic target for liver cancer treatment.
