A Brief Review on Hydroxyapatite Nanoparticles Interactions with Biological Constituents

With the pursuit of new cancer therapies and more effective treatment to diseases in the last decades, nanotechnology has been an important ally for healthcare professionals and patients in critical clinical conditions. Nanomaterials offer an alternative way to deliver toxic chemotherapeutic drugs to specific biological tissues, specific cells or specific microbial beings, resulting in avoidance of strong side effects or resilience to effective drugs. Among these materials, stands out the hydroxyapatite nanoparticles, a ceramic class of calcium phosphates that present chemical and structural similarities with the mineral phase of the human skeleton’s bone matrix, resulting in important biological features, such as biocompatibility, osteoconductive, osteoinduction and osteoaffinity, which led to a lot of scientific researches to apply these nanoparticles for bone diseases diagnosis and therapeutics. Due to the hydroxyapatite biological activities and due to the possibility to promote chemical and physical modifications in these nanoparticles, they can interact with biological cells or microorganisms in different ways, resulting in multiple potentialities to be explored such as apoptosis induction to cancerous cells, osteogenesis promotion, cellular proliferation, angiogenesis and tissue recovery, in addition to promote cell adhesion and cell uptake. Furthermore, chemical and physical modifications, such as surface functionalization, dopant inclusions and radiolabeling process, allow scientists to track the particle activities in biological environments. In the last decades of scientific productions, the literature brings together important data on how hydroxyapatite nanoparticles interact with biological tissues and such data are crucial for the development of more effective therapeutic and diagnostic agents. AIM To evaluate the sex-specific effects of a hydroalcoholic extract from Eugenia punicifolia(HEEP) leaves on gastric ulcer *** In this rat study involving males, intact(cycling) females, and ovariectomized females, gastric ulcers were induced using acetic acid. A vehicle, lansoprazole, or HEEP was administered for 14 d after ulcer induction. Body weight was monitored throughout the treatment period. At the end of treatment, the rats were euthanized and the following in vivo and in vitro investigations were performed: macroscopic examination of the lesion area and organ weights, biochemical analysis, zymography, and evaluation of protein expression levels. Additionally, the concentration-dependent effect of HEEP was evaluated in terms of subacute toxicity and *** Compared to the vehicle, HEEP demonstrated a great healing capacity by substantially reducing the ulcerative lesion area in males(52.44%), intact females(85.22%), and ovariectomized females(65.47%), confirming that HEEP accelerates the healing of acetic acidinduced gastric lesions and suggesting that this effect is modulated by female sex hormones. The antiulcer effect of HEEP was mediated by prostaglandin E2 only in male rats. Overall, the beneficial effect of HEEP was the highest in intact females. Notably, HEEP promoted the expression of vascular endothelial growth factor(intact vs ovariectomized females) and decreased the expression of Caspase-8 and Bcl-2(intact female vs male or ovariectomized female). Additionally, HEEP enhanced fibroblast proliferation and migration into a wounded area in vitro, confirming its healing effect. Association between Infrared Thermography, Blood Count and Creatine Kinase in the Evaluation of the Welfare of Vaquejada Horses

The Quarter-Horses regularly participate in non-traditional sports that simulate activities on cattle ranches. In the vaquejada races, the horses run at about 30 km/h, reaching maximum heart rates of around 200 bpm, indicating the great physical effort during competitions and with that could impact welfare. To test the hypothesis that vaquejada horses can be evaluated for the quality of welfare through a combination of non-invasive and invasive methods, an experiment was developed that aimed to assess the quality of welfare through thermography and blood biomarkers before and after vaquejada races. Ten vaquejada horses, which were in regular competition, were submitted to the vaquejadas racing test were used. Thermography was performed in 14 regions of interest (head, neck and thorax) in the following phases: pre-race and +1, +4 and +24 hours of the races. Blood samples were collected in pre-race, immediately after and +1, +4 and +24 hours of the races. Results were submitted to ANOVA and Tukey’s test, with p set at 5%. Thermography showed that local temperatures before the race and +24 hours after the races were similar (p > 0.05), as well as between +1 and +4 hours after the races (p > 0.05), which showed higher surface temperatures than the first group (pre-race and +24 hours) (p 0.05) after the exercises. In conclusion, the vaquejada horses evaluated by infrared thermography and blood analyses maintained the quality of well-being, with the physiological elevation of several parameters after the races and recovery in less than 24 hours after the races. Thermographic images were also able to help in the analysis of the regions of interest that have contact with harnesses, demonstrating that the use of adequate and correct equipment does not compromise animal welfare.

Effort and Recovery in Nellore Oxen during Vaquejada Assessed with Ocular and Tail Infrared Thermography Superficial Temperature

Infrared thermography (IRT) has emerged for evaluation of animal welfare. To test the hypothesis that cattle subjected to vaquejada increased temperature in the eye (CLO) and tail due to physical effort, a research was developed that aimed to measure maximum surface temperature in the CLO and base of tail using IRT. Eighty Nellore cattle were used, which were subjected to physical effort in three periods (morning, afternoon and night). IRT was performed at CLO and base of tail, in rest pen/corral (control) and during the vaquejada (pre- and post-run). Tails’ analysis was divided in 3 points (E1, E2 and E3). ANOVA and Tukey’s test (p < 0.001) were used for analysis. Temperatures were higher in morning and afternoon and different from those at night on control (p < 0.001). During vaquejada with one run, CLO was higher in pre-run, followed by a slight reduction in post-run (p < 0.001). Analyzing temperatures variations at tail, higher temperatures were observed in the morning, followed by the evening and night (p < 0.001), but without differences within the pre- and post-run periods. Temperatures at tail’s points were higher in morning period in cattle with two runs (p < 0.001). Finally, it was concluded that there were no increases in CLO or at tail’s points after vaquejada races. The IRT method was efficient in determining surface temperatures in CLO and tail points in cattle under the same conditions and may be a good noninvasive method for clinical and welfare assessments.

In recent decades, the intensity of training and equestrian competitions has significantly increased, thus the assessment of the well-being of the equine athlete has become essential in all equestrian modalities. The aim of this study was to ascertain whether equine athletes submitted to a vaquejada simulation test (VqST), comprised of three races, presented changes in blood biomarkers related to stress and health status. Fourteen healthy Quarter Horses, used as pull horses in this equestrian modality, were evaluated. Ten animals were submitted to the VqST and the remaining four were used as a control group. Blood samples were collected pre-test (during fast), immediately after, and at 1, 4 and 24 hours of recovery. The assessed blood biomarkers included cortisol, interleukin (IL)-6, IL-1β, iron, urea, creatinine, and gamma-glutamyl transferase (GGT) concentrations and results were analyzed using One Way ANOVA (time) with the SigmaStat 13.0 software. No differences between sample times were detected in both groups (p > 0.05) and no differences were found between groups (p > 0.05). The results suggest that all horses were well conditioned for the level of effort imposed by the three vaquejada races. The adaptation to physical exercise may enable the regulation of the acute response to stress in the tissues involved in the exercises, with no differences being observed in stress and health biomarkers, such as IL-6, IL-1β and cortisol. In conclusion, well-conditioned vaquejada horses exhibit a balanced regulation of biological processes, which contributes an increased athletic longevity and better quality of athletic life.

Adenosine A1 receptor activation inhibits histamine release in gastric enterochromaffin-like cells

Adenosine acts as a gastroprotective factor decreasing inflammation and reducing gastric acid secretion. Quantitative RT-PCR was used to determine the expression of the adenosine receptor genes (A1AR, A2AAR, A2BAR and A3AR) and that of the gastrin receptor B gene (CCKBR) in isolated, short-term cultured enterochromaffin-like (ECL) cells. Both the A1AR and the CCKBR genes were expressed at a level high-er than the other genes. Also, the effect of 2-chloroadenosine, a stable agonist of A1 and A2A receptors, was explored on ECL cells, with a resulting inhibition of both basal and gastrin-stimulated histamine release. Also, dipropylcyclopentylxanthine (DPCPX), a selective A1 antagonist, prevented the inhibitory effects of 2-chloro-adenosine, suggesting the effect of 2-chloroadenosine is mediated by A1 receptors. It is concluded that isolated, short-term cultured ECL cells are a suitable model for studies relating gene expression and function, and that the gastroprotective actions of adenosine are at least partly mediated through A1 receptors.

Degenerative musculoskeletal disorders are one of the top causes of pain and disability in the adult *** available alternatives to mitigate symptoms include conservative treatments such as the administration of pharmacological agents and an educative approach towards lifestyle *** use of certain analgesics,such as opiates and corticosteroids,delivers short term results but do not address the etiological source of pain and ***,prolonged use of such medications may cause additional ***,the demand for musculoskeletal tissue regeneration has led to an alternative approach referred to as“orthobiologics”.This alternative is based on cellular and molecular components capable of inducing and promoting tissue *** marrow(BM)aspirate(BMA)and concentrate are well-known orthobiologics used to treat musculoskeletal *** derived from the BM have been discussed in the literature;however,the lack of standardization regarding collection and processing protocols presents a challenge for generalization of study outcomes and determination of *** BM-derived orthobiologics have not yet been classified,to our knowledge,this manuscript proposes the ACH classification system,which speaks to BMA(A),BMA and concentrate(C)and hybrid(H),which combines A and *** classification proposes and describes 8 parameters that are relevant for the quality of biological *** more parameters used would imply greater characterization and complexity of the evaluation of the biological product *** ACH classification envisages a necessary contribution to the comprehension of both clinical procedures and research outcomes,ultimately ushering in a standardization of best practice.
