Residual activity of cetrimide and chlorhexidine on Enterococcus faecalis-infected root canals

Effective final irrigation regimen is an important step in order to achieve better disinfection and ensure residual antimicrobial effects after root canal preparation. The aim of this study was to compare the residual antimicrobial activity of 0.2% cetrimide, and 0.2% and 2% chlorhexidine in root canals infected with Enterococcus faecalis. Biofilms of E. faecalis were grown on uniradicular roots for 4 weeks. After root canal preparation, root canals were irrigated with 17% ethylenediaminetetraacetic acid （EDTA） to remove the smear layer. The roots were randomly divided into three experimental groups （n=26） according to the final irrigating solution： group I, 5 mL 0.2% cetrimide; group II, 5 mL 0.2% chlorhexidine; and group III, 5 mL 2% chlorhexidine. Samples were collected for 50 days to denote the presence of bacterial growth. The proportion of ungrown specimens over 50 days was evaluated using the nonparametric Kaplan-meier survival analysis. Differences among groups were tested using the log-rank test and the level of statistical significance was set at P〈0.05. The highest survival value was found with 2% chlorhexidine, showing statistically significant differences from the other two groups. At 50 days, E. faecalisgrowth was detected in 69.23% specimens in groups I and II, and in 34.61% specimens of group III. There were no significant differences between 0.2% cetrimide and 0.2% chlorhexidine. Final irrigation with 2% chlorhexidine showed greater residual activity than 0.2% chlorhexidine and 0.2% cetrimide in root canals infected with E. faecalis.

Detection of KRAS g12D in colorectal cancer stool by droplet digital PCR

AImTo assess KRAS g12D mutation detection by droplet digital PCR (ddPCR) in stool-derived DNA from colorectal cancer (CRC) *** this study, tumor tissue and stool samples were collected from 70 patients with stage I-IV CRC diagnosed by preoperative biopsy. KRAS mutational status was determined by pyrosequencing analysis of DNA obtained from formalin-fixed paraffin-embedded (FFPE) tumor tissues. The KRAS g12D mutation was then analyzed by ddPCR in FFPE tumors and stool-derived DNA from patients with this point mutation. Wild-type (WT) tumors, as determined by pyrosequencing, were included as controls; analysis of FFPE tissue and stool-derived DNA by ddPCR was performed for these patients as *** the total 70 patients included, KRAS mutations were detected by pyrosequencing in 32 (45.71%), whereas 38 (54.29%) had WT tumors. The frequency of KRAS mutations was higher in left-sided tumors (11 located in the right colon, 15 in the left, and 6 in the rectum). The predominant point mutation was KRAS g12D (14.29%, n = 10), which was more frequent in early-stage tumors (I-IIA, n = 7). In agreement with pyrosequencing results, the KRAS g12D mutation was detected by ddPCR in FFPE tumor-derived DNA, and only a residual number of mutated copies was found in WT controls. The KRAS g12D mutation was also detected in stool-derived DNA in 80% of all fecal samples from CRC patients with this point *** is a reliable and sensitive method to analyze KRAS g12D mutation in stool-derived DNA from CRC patients, especially at early stages. This non-invasive approach is potentially applicable to other relevant biomarkers for CRC management.

In the present work, the synthesis of micro- and nano-sized spheres of metallic bismuth by microwave-assisted solvothermal method is reported. The synthesis method was carried out at different power levels and at a unique frequency of microwave irradiation. The sphere sizes were controlled by the microwave power level and the concentration of dissolved precursor. Structural and morphological characterization was performed by SEm, HRTEm, EELS and XRD. The results demonstrated that rhombohedral zero valent Bi spheres were synthesized after microwave radiation at 600 and 1200 W. However, if the power level is decreased to 120 W, a monoclinic phase of Bi203 is obtained with a flake-like morphology. In comparison with a conventional hydrothermal process, the microwave-assisted solvothermal approach provides many advantages such as shorter reaction time, optimum manipulation of morphologies and provides a specific chemical phase and avoids the mixture of structural phases and morphologies which is essential for further applications such as drug delivery or functionalization with organic materials, thanks to its biocompatibility.

Strength Developing in Clay-Andesite Brick

The objective of this work was to develop a more and better understanding of the strength developing in clay-andesite fired brick. The purpose was to improve the quality of the clay bricks that are handmade in the southern region of Ecuador to make them suitable for more widespread use in the local construction industry. To achieve our goal, we first physically, chemically, and mechanically characterized the “clayey” and “sandy” materials used in the fabrication of handmade bricks in the region. Second, the optimal mixture (Om): the optimal proportion between the amounts of “clayey” and “sandy” material was sought. Third, clay bricks were prepared using the Om, baked at 950°C, and characterized. In addition, bricks produced by regional artisans were characterized, and the results were compared with the results obtained for the bricks prepared using the Om. Our data reveal that the optimal mixture is 50% “clayey” material and 50% “sandy” material and that with this mixture, an average improvement in brick quality of 300% can be achieved;thus, the use of the Om makes it possible to expand the use of these bricks in the local construction industry and enables an environmentally friendly production process by reducing the intensive exploitation of regional clay deposits. This improvement is achieved by virtue of the anorthite enrichment that occurs in the solid solution, which results from the evolution of andesine. The anorthite contributes to the formation of a stronger matrix among the different grains of the material. Conditions are favorable for this enrichment process to occur when “sandy” materials with high contents of andesite, which is common in Ecuadorian soils, are used.

Single sperm selection and DNA fragmentation analysis: The case of mSOmE/ImSI

Sperm of poor quality may affect syngamy after fertilization, embryo development up to the blastocyst stage and reproductive outcome. Subsequently, sperm selection based on morphological characteristics and sperm DNA quality may help to partially avoid these problems. Today, highly efficient sperm selection based on morphological characteristics can be attained using the motile sperm organelle morphology (mSOmE) examination, and the spermatozoa selected can be used for ICSI through a fertilization strategy known as intra-cytoplasmic morphologically selected sperm injection (ImSI). The aim of this investigation was to develop a simple methodology to assess sperm DNA fragmentation in single spermatozoa following mSOmE/ ImSI, to test the hypothesis that morphologically normal spermatozoa, with an absence of vacuolization, is free of DNA damage. The results indicated that mSOmE/ImSI-selected sperm, combined with the Sperm Chromatin Dispersion test (SCD;Oligo-Halosperm), can be reliably used to assess sperm DNA damage in selected single spermatozoa (75% average efficiency), thereby establishing a direct relationship between a good morphological pattern on the sperm and a good DNA quality. Furthermore, results showed spermatozoa presenting a normal morphology and no traces of vacuolization to be fully free of DNA damage. However, traces of vacuolization and more severe morphological alterations were accompanied by significant increases in the proportion of sperm containing a damaged DNA molecule. Interestingly, subtle morphological differences observed between normal and non-vacuolated and normal but vacuolated sperm exhibited significant differrences in the ability of the SCD-Oligo-Halosperm treated sperm to expand DNA fibers following protein depletion.

Osteoclasts are bone-resorbing cells that play an essential role in homeostatic bone remodeling and pathological bone *** colony stimulating factor(m-CSF)is abundant in rheumatoid arthritis(RA).However,the role of m-CSF in arthritic bone erosion is not completely ***,we show that m-CSF can promote osteoclastogenesis by triggering the proteolysis of c-FmS,a receptor for m-CSF,leading to the generation of FmS intracellular domain(FICD)*** levels of FICD fragments positively regulated osteoclastogenesis but had no effect on inflammatory ***,myeloid cell-specific FICD expression in mice resulted in significantly increased osteoclast-mediated bone resorption in an inflammatory arthritis *** FICD formed a complex with DAP5,and the FICD/DAP5 axis promoted osteoclast differentiation by activating the mNK1/2/EIF4E pathway and enhancing NFATcl protein ***,targeting the mNK1/2 pathway diminished arthritic bone *** results identified a novel role of c-FmS proteolysis in osteoclastogenesis and the pathogenesis of arthritic bone erosion.

Chlorophyll Concentration and morphological Diversity in Corn Lines at Different Vegetative Stages

The objective of this study was to assess the chlorophyll content of corn lines at different vegetative stages and the diversity between them. The study was conducted in the experimental field of the Roque Institute of Technology (ITR) located in Roque, Celaya, guanajuato, mexico. We used 32 corn lines from CImmYT and four lines from the breeding program of the ITR. The evaluation was conducted in the spring-summer cycle of 2014 at two planting dates. We used a completely random.zed block design with three replications. The experimental unit consisted of two rows of five linear meters with a distance of 0.75 meters between rows. Chlorophyll was measured at six different stages: V6, VT, R3, R4, R5 and mF. The agronomic variables recorded were: PH, CH, LAC, LBC, FF, mF, Pm, CC, CL, DC, RC, KR and gY. The results of the analysis of variance showed significant differences between lines, dates and their interaction. On average, 18 lines were found with chlorophyll concentrations above 51 SPAD units in the two environments. This indicated that these materials had a high potential for field performance. Using cluster analysis UPgmA method, we identified seven groups for chlorophyll and eight groups for agronomic characteristics for a breeding program. The groups were associated to the homogeneous characteristics of each variable.

SLC26A4 mutation testing for hearing loss associated with enlargement of the vestibular aqueduct

nPendred syndrome （PS） is characterized by autosomal recessive inheritance of goiter associated with a defect of iodide organification, hearing loss, enlargement of the vestibular aqueduct （EVA）, and mutations of the SLC26A4 gene. However, not all EVA patients have PS or SLC26A4 mutations. Two mutant alleles of SLC26A4 are detected in 1/4 of North American or European EVA populations, one mutant allele is detected in another 1/4 of patient populations, and no mutations are de-tected in the other 1/2. The presence of two mutant al-leles of SLC26A4 is associated with abnormal iodide or-ganification, increased thyroid gland volume, increased severity of hearing loss, and bilateral EVA. The pres-ence of a single mutant allele of SLC26A4 is associated with normal iodide organification, normal thyroid gland volume, less severe hearing loss and either bilateral or unilateral EVA. When other underlying correlations are accounted for, the presence of a cochlear malformation or the size of EVA does not have an effect on hearing thresholds. This is consistent with observations of an Slc26a4 mutant mouse model of EVA in which hearing loss is independent of endolymphatic hydrops or in-ner ear malformations. Segregation analyses of EVA in families suggest that the patients carrying one mutant allele of SLC26A4 have a second, undetected mutant allele of SLC26A4, and the probability of a sibling hav-ing EVA is consistent with its segregation as an autoso-mal recessive trait. Patients without any mutations are an etiologically heterogeneous group in which siblings have a lower probability of having EVA. SLC26A4 muta-tion testing can provide prognostic information to guide clinical surveillance and management, as well as the probability of EVA affecting a sibling.
