Interleukin 28B-related polymorphisms: A pathway for understanding hepatitis C virus infection?

AIM:To analyze the role of rs12979860 and rs8099917polymorphisms in hepatitis C virus(HCV)genotype 1infection of ***:A total of 145 adult patients diagnosed with genotype 1 chronic hepatitis C(CHC)who had completed a 48-wk regimen of pegylated-interferonα-2a or-2b plus ribavirin combination therapy were recruited from six large urban healthcare centers and199 healthy blood donors(controls)from a single site between January 2010 and January *** on the patients’response to treatment was *** chain reaction-restriction fragment length polymorphism genotyping of the interleukin(IL)28B gene fragment encompassing the single nucleotide polymorphisms(SNPs)rs12979860(C/T)and rs8099917(T/G)was carried out for 79 of the CHC patients and 199 of the ***-directional amplicon sequencing of the two SNPs was carried out for the remaining 66 CHC ***:SNP rs12979860 genotyping was successful in 99.5%of the controls and 97.2%of the CHC patients,whereas the SNP rs8099917 genotyping was successful in 95.5%of the controls and 100%of the CHC *** genotype and allele distributions for both rs12979860 and rs8099917 were significantly different between the control and CHC patient groups,with significantly higher genotype frequencies of CC and TT in the controls(P=0.037 and 0.046,respectively)and of TT and GG in the CHC patients(P=0.0009and 0.0001,respectively).Analysis of the CHC patients who achieved sustained virological response(SVR)to treatment(n=55)indicated that the rs12979860 C allele and CC genotype were predictors of SVR(P=0.02).No significant correlation was found between rs8099917 genotypes and treatment response,but carriers of the T allele showed significantly higher rates of SVR(P=0.02).Linkage disequilibrium analysis of the group that achieved SVR showed a significant association between rs12979860 and rs8099917(P=0.07).CoNCLUSIoN:The higher allele frequency of rs12979860 C and rs8099917 T observed in non-HCVinfec

BACKGRoUND To achieve the elimination of hepatitis B and C,there is an urgent need to develop alternative strategies to increase the access of diagnosis,particularly among key populations such as people living with human immunodeficiency virus(HIV),individuals with coagulopathies and chronic kidney disease(CKD)*** To evaluate the use of dried blood spot(DBS)in the detection of hepatitis B virus(HBV)and hepatitis C virus(HCV)*** A total of 430 individuals comprised of people living with HIV,coagulopathies and CKD provided paired serum and DBS ***,anti-HBc and anti-HCV were tested in those samples using a commercial *** and selected behavioral variables were evaluated to assess possible association with HBV and HCV *** Using DBS,HBsAg prevalence varied from 3.9%to 22.1%,anti-HBc rates varied from 25.5%to 45.6%and anti-HCV positivity ranged from 15.9%to 41.2%in key *** of HBV and HCV tests using DBS varied from 88.9%to 100%.The HBsAg assay demonstrated the best performance in CKD and coagulopathy individuals and the anti-HCV test had a sensitivity and specificity of 100%in people living with *** of HBV and HCV detection in DBS varied from 90.2%to 100%.In the CKD group,HBsAg positivity was associated with infrequent use of condoms,and anti-HBc positivity was associated with sharing nail cutters/razors/***-HCV reactivity was positively associated with a history of transplantation and length of time using hemodialysis in both *** people living with HIV,only the male gender was associated with anti-HBc positivity in serum and *** DBS with electrochemiluminescence are useful tools for the diagnosis and prevalence studies of hepatitis B and C among key populations and may increase the opportunity to foster prevention and treatment.

Femoral positioning influences ipsi-and contralateral anterior cruciate ligament rupture following its reconstruction:Systematic review and meta-analysis

AIM To systematically review the incidence of ipsilateral graft re-rupture and contralateral anterior cruciate ligament(ACL) rupture following its reconstruction, with special attention to the femoral drilling technique. METHoDS Systematic review and meta-analysis of high-level prospective studies searched in MEDLINE database following PRISMA statement. The rate of ipsilateral graft re-rupture and contralateral rupture in patients submitted to either transtibial(TT) technique(isometric) or anteromedial(AM) technique(anatomic) was compared. RESULTS Eleven studies met the criteria and were included in final analysis. Reconstructions using the AM technique had a similar chance of contralateral ACL rupture when compared to the chance of ipsilateral graft failure(oR = 1.08, P = 0.746). In reconstructions using TT technique, the chance of contralateral ACL rupture was approximately 1.5 times higher than ipsilateral graft failure(oR = 1.49, P = 0.048). Incidence of contralateral lesions were similar among the techniques TT(7.4%) and AM(7.0%)(P = 0.963), but a trend could be noticed with a lower incidence of lesion in the ipsilateral limb when using the TT technique(4.9%) compared to the AM technique(6.5%)(P = 0.081).CoNCLUSIoN ACL reconstruction by TT technique leads to lower incidence of graft re-injury than contralateral ACL lesion. There is no difference between the chance of re-injury after AM technique and the chance of contralateral ACL lesion(native ligament) with either technique.

During the operation of complex process, such as oil production or refming, abnormal situations may occur, leading to an alarm flooding. Alarm flooding is the signalling of a large number of alarms in a few minutes, in such a way that it is impossible for the operator to attend to all alarms. on these occasions, it is usual that the operator leaves the alarm summary list and gets an analysis of the plant through the screens of the DCS （digital control system）, seeking to understand the situation. The alarm summary list ceases to be a useful tool. In such cases, the operator might have the aid of a filter that would present the highest priority alarms and other information associated with them, enabling him to gain a better knowledge of the situation. This paper describes the interface of a system aimed to help the operator to have a more comprehensive knowledge of the process （a better situational awareness） during process upsets that cause alarm flooding, recovering the utility of the alarm layer to the safety of industrial processes.

Hemangioma is the most common benign tumor affecting the liver in 7% of general population. one of the rare presentations of this pathology is gigantic liver hemangioma, reported in the literature as anecdotal cases. In selected cases, liver transplantation has been a therapeutic option. We herein describe a case of massive liver hemangioma which was submitted to liver transplant, and the main indication for liver transplant in this case was a severe psychosocial impact of the disease in the patient’s life, altogether with Kasabach-Merritt syndrome and abdominal discomfort.

Biomechanical Analysis of Implant Assisted-overdentures with Variations in the Attachments Systems

The aim of this study was to assess the stress patterns on the peri-implant zone and residual alveolar ridge in different overdenture attachment system designs by the photoelasticity method. Four attachments systems were tested: o-ring, ERA, Bar-clip, and Bar-clip/o-ring association. The prostheses were loaded with 100 N in five pre-determined points and the photoelastic model was evaluated by a circular polariscope. The anterior load, o-ring, and ERA showed better stress distribution in relation to the bars systems, which presented stress levels surrounding implants. The molar load, Bar-clip/o-ring association, presented the biggest stress concentration on the peri-implant region in relation to the others. When the second molar was loaded, there was a concentration of stress in the alveolar ridge in all situations analyzed. Within the limitation of this “in vitro” study, it could be concluded that there were biomechanical differences among the attachments systems analyzed, principally between isolated and the bars systems. The o-ring showed better stress distribution and the Bar-clip/o-ring showed higher stress concentration.

Photodynamic therapy today is becoming an important role in the healing of lacerated tissues, since it has therapeutic resources capable of accelerating this process. one treatment option is the clinical phototherapy, and Photodynamic Therapy (PDT) is being widely used. This study aims to evaluate the effect of PDT on the healing of skin wounds in rats. We used a sample of 39 male rats Wistar divided into three groups, a control, a PDT-treated green and red with the last PDT. After 24 hours before the surgical incisions, PDT was used in both groups for 6 minutes and was evaluated in histological level, the inflammatory reaction and the repair process. The results showed that the granulation tissue was more developed in the irradiated group than in the control group and the amount of chronic inflammatory cells (monocytes, macrophages, lymphocytes and plasma cells) predominated with green phototherapy. The epithelialization in the wound margins and scarring with better quality occurred with red PDT (640 nm), which the higher deposition was of collagen. However, phototherapy not collimated of 640 nm (red) resulted in better anti-inflammatory effects. Given these answers with phototherapy not collimated green (525 nm) and red (640 nm), we conclude that the use of these wavelengths is possible with benefits, mainly with red LED.
