Base excision repair accessory factors in senescence avoidance and resistance to treatments

cancer cells,in which the RAS and PI3K pathways are activated,produce high levels of reactive oxygen species(ROS),which cause oxidative DNA damage and ultimately cellular *** process has been documented in tissue culture,mouse models,and human pre-cancerous *** this context,cellular senescence functions as a tumour suppressor *** rare cancer cells,however,manage to adapt to avoid senescence and continue to *** well-documented mode of adaptation involves increased production of antioxidants often associated with inactivation of the KEAP1 tumour suppressor gene and the resulting upregulation of the NRf2 transcription *** this review,we detail an alternative mode of adaptation to oxidative DNA damage induced by ROS:the increased activity of the base excision repair(BER)pathway,achieved through the enhanced expression of BER enzymes and DNA repair accessory *** proteins,exemplified here by the cUT domain proteins cUX1,cUX2,and SATB1,stimulate the activity of BER *** ensued accelerated repair of oxidative DNA damage enables cancer cells to avoid senescence despite high ROS *** a by-product of this adaptation,these cancer cells exhibit increased resistance to genotoxic treatments including ionizing radiation,temozolomide,and ***,considering the intrinsic error rate associated with DNA repair and translesion synthesis,the elevated number of oxidative DNA lesions caused by high ROS leads to the accumulation of mutations in the cancer cell population,thereby contributing to tumour heterogeneity and eventually to the acquisition of resistance,a major obstacle to clinical treatment.

Novel elastomeric spiropyran‑doped poly(dimethylsiloxane)optical waveguide for UV sensing

Novel poly(dimethylsiloxane)(PDMS)doped with two diferent spiropyran derivatives(SP)were investigated as potential candidates for the preparation of elastomeric waveguides with UV-dependent optical ***,free-standing flms were prepared and evaluated with respect to their photochromic response to UV ***,reversibility as well as photofatigue and refractive index of the SP-doped PDMS samples were ***,SP-doped PDMS waveguides were fabricated and tested as UV sensors by monitoring changes in the transmitted optical power of a visible laser(633 nm).UV sensing was successfully demonstrated by doping PDMS using one spiropyran derivative whose propagation loss was measured as 1.04 dB/cm at 633 nm,and sensitivity estimated at 115%change in transmitted optical power per unit change in UV *** decay and recovery time constants were measured at 42 and 107 s,respectively,with an average UV saturation dose of 0.4 J/*** prepared waveguides exhibited a reversible and consistent response even under *** sensor parameters can be tailored by varying the waveguide length up to 21 cm,and are afected by white light and temperatures up to 70℃.This work is relevant to elastomeric optics,smart optical materials,and polymer optical waveguide sensors.

Stress tensors and quantification of fracture patterns to analyze connectivity and potential fluid flow in a mesa landform of the Northern Andes

This work applies stress tensors inversions and quantification of fracture patterns along the Mesa de Los Santos, in the Eastern cordillera of colombia, to better understand the potential fluid flow. It thus contributes to the conceptual hydrogeological model. The area was subdivided into three blocks, separated by the NW Potreros and the Los Santos faults, having minor inner faults of different orientations. This separation facilitates the analysis of the fractures measured in the field, which in general show high dip angles and a conjugate geometry in the northern block, tension fractures(Mode I) in the central block, and a random distribution in the southwestern block. WinTensor treatment of slickensides yielded a maximum horizontal stress(SHmax) of 111o, which coincides with the WNW-ESE tensor observed from the conjugate and tension joints. We then used frac Pa Q to generate interpolation maps of fracture intensity and density. The maps show the highest values in the central block and the lowest in the northern block,where the precipitation is higher, causing intensive rock weathering and homogenization of the fracture planes. Although the highest values of connectivity by line are found to the south of the mesa, we suggest the possibility of greater flow from the recharge zone(NE) along bedding planes and open NW-SE fractures.

Effects of dust controls on respirable coal mine dust composition and particle sizes:case studies on auxiliary scrubbers and canopy air curtain

control of dust in underground coal mines is critical for mitigating both safety and health *** decades,the National Institute of Occupational Safety and Health(NIOSH)has led research to evaluate the effectiveness of various dust control technologies in coal *** studies have included the evaluation of auxiliary scrubbers to reduce respirable dust downstream of active mining and the use of canopy air curtains(cAcs)to reduce respirable dust in key operator *** detailed dust characterization was not a focus of such studies,this is a growing area of *** preserved filter samples from three previous NIOSH studies,the current work aims to explore the effect of two different scrubbers(one wet and one dry)and a roof bolter cAc on respirable dust composition and particle size *** this,the preserved filter samples were analyzed by thermogravimetric analysis and/or scanning electron microscopy with energy dispersive *** indicate that dust composition was not appreciably affected by either scrubber or the ***,the wet scrubber and cAc appeared to decrease the overall particle size *** an effect of the dry scrubber was not consistently observed,but this is probably related to the particular sampling location downstream of the scrubber which allowed for significant mixing of the scrubber exhaust and other return *** from the insights gained with respect to the three specific dust control case studies revisited here,this work demonstrates the value of preserved dust samples for follow-up investigation more broadly.

Mathematical modeling and simulations of stress mitigation by coating polycrystalline particles in lithium-ion batteries

A chemo-mechanical model is developed to investigate the effects on the stress development of the coating of polycrystalline Ni-rich LiNixMnyco_(z)O_(2)(x≥0.8)(NMc)particles with poly(3,4-ethylenedioxythiophene)(PEDOT).The simulation results show that the coating of primary NMc particles significantly reduces the stress generation by efficiently accommodating the volume change associated with the lithium diffusion,and the coating layer plays roles both as a cushion against the volume change and a channel for the lithium transport,promoting the lithium distribution across the secondary particles more ***,the lower stiffness,higher ionic conductivity,and larger thickness of the coating layer improve the stress *** paper provides a mathematical framework for calculating the chemo-mechanical responses of anisotropic electrode materials and fundamental insights into how the coating of NMc active particles mitigates stress levels.

Adsorption of pharmaceutical pollutants on Zncl_(2)-activated biochar from corn cob:Efficiency,selectivity and mechanism

The occurrence of pharmaceuticals in water bodies and drinking water poses risks for the envi-ronment and human health,thus it is necessary to study methodologies that allow the efficient removal of these *** this work,corn cob-derived biochar was obtained by Zncl_(2)-activation,and subsequent carbonization at 700℃.The effect of contact time,temperature,pH,and initial concentration on the adsorption capacity of acetaminophen(AcE)and amoxicillin(AMX)was determined through batch *** addition,the kinetics,isotherms,and ther-modynamics parameters were *** activated biochar exhibited a maximum adsorp-tion capacity of 332.08 mg/g for AcE and 175.86 mg/g for *** adsorption kinetics and adsorption isotherm of AcE corresponded to the pseudo-second order and Langmuir model,***,pseudo-first-order kinetics and the freundlich isotherm model were well-fitted to AMX *** AcE and AMX co-adsorption had a synergistic effect on AMX but an antagonistic effect on AcE removal,achieving a maximum adsorption capacity of 193.51 and 184.58 mg/g,*** the other hand,fixed-bed column experiments showed that the adsorption capacity depends on the influent concentration,and the breakthrough curve fits the Thomas and Yoon-Nelson *** mechanism adsorption studies showed that surface inter-actions(hydrogen bonding formation and n-𝜋interactions)are the main driving forces for the adsorption process,and pore filling is the rate-limiting *** this way,the prepared biochar exhibits a high potential for the adsorption of pharmaceutical compounds from water.

comparisons of oxbow lake fish assemblages in relation to bigheaded carp establishment in the lower White River, Arkansas

Background Since the late 1990s, bigheaded carps(largely silver carp [Hypophthalmichthys molitrix] but also bighead carp [H. nobilis]) have established throughout the lower Mississippi River basin. Using previously studied oxbow lakes in the lower White River basin, Arkansas, we compared current(2017, “post-carp” establishment) fish assemblages to historical(2002, “pre-carp” establishment) fish assemblages. fish assemblages were comprehensively assessed using multiple gears, including boat electrofishing, mini-fyke nets, and experimental small-mesh gill *** T-tests suggested that fish assemblage indices of richness, diversity, evenness, and dominance were often greater(P carp period as reflected by boat electrofishing and experimental gill nets. However, all indices were generally similar(P > 0.05) between the pre-carp and post-carp period with fish assemblages depicted using mini-fyke nets. Nonmetric multidimensional scaling analyses indicated that fish assemblages differed structurally between pre-carp and post-carp periods. Assemblage differences were linked to both small and large abundance changes for more than 20 species. Abundances of gizzard shad(Dorosoma cepedianum), emerald shiner(Notropis atherinoides), pugnose minnow(Opsopoeodus emiliae), crappies(Pomoxis spp.), bluegill(Lepomis macrochirus), orangespotted sunfish(L. humilis), and yellow bass(Morone mississippiensis) declined between the pre-carp and postcarp periods. conversely, abundances of weed shiner(N. texanus), pallid shiner(Hybopsis amnis), longear sunfish(L. megalotis), buffalofishes(Ictiobus spp.), and gars(Lepisosteus spp.) generally increased during the same *** Although not possible to conclude assemblage shifts were entirely related to bigheaded carps due to the absence of an appropriate reference system where carps did not establish, the wide establishment of these carps is one of the most pervasive changes to have occurred in the lower White

Radar Absorbing Materials(RAM)are a class of composites that can attenuate incident electromagnetic waves to avoid radar *** carbon allotropes that have the potential to be used as RAM are either carbon nano-tubes(cNTs),graphene,carbon black(cB)and ultimately,sustainable porous carbon(SPc).Here,black wattle bark waste(following tannin extraction)was used as a sustainable source to produce SPc made from biomass *** was characterized and used as aﬁller for a silicone rubber matrix to produce aﬂexible *** elec-tromagnetic performance of this composite was compared with composites made with commercial cB and cNT through reﬂection loss(RL),where-10 dB is equivalent to 90%of *** composites were evaluated in single-layer,double-layer,and as radar absorbing structures(RAS)with the aim of improving their effective absorption bandwidth(EAB)performances and a reduction in *** cNT composite presented a RL of-26.85 dB at 10.89 GHz and an EAB of 2.6 GHz with a 1.9 mm thickness,while the double-layer structures using cNT and SPc provided a RL of-19.74 dB at 10.75 GHz and an EAB of 2.51 ***,the double-layer structures are~42%cheaper than the composite using only cNT since less material is ***,the largest EAB was achieved with a RAS using SPc,reaching~2.8 GHz and a RL of-49.09 dB at 10.4 ***,SPc made of black wattle bark waste can be a competitive,alternative material for use as RAM and RAS since it is cheaper,sustainable,and suitable for daily life uses such as absorbers for anechoic chambers,sensors,and elec-tromagnetic interference shields for electronics,wallets,vehicles,and others.
