Warmer temperature during asexual reproduction induce methylome, transcriptomic, and lasting phenotypic changes in Fragaria vesca ecotypes

Effects of virus infection on expression of cell cycle regulatory proteins in the unicellular marine algae Emiliania huxleyi

The authors have investigated the biochemical events by which marine algal virus infection induces cell cycle arrest. The key G 2 /M-phase regulatory proteins are analyzed by immunobloting in unicel-lular Emiliania huxleyi,suggesting that virus induced cell cycle arrest is related with virus＇s effect on cyclins and cyclin dependent kinases. E. huxleyi virus（EhV） represses cdc2/cyclinB complex activity by inhibiting the activity of cdc2 kinase in a phosphorylation-related manner,blocking host cells G 2 /M checkpoint. Dephosphorylated /inactive cdc25c combined with up-regulation of Wee1 expression at early infect period appears to be important mechanisms by which EhV represses cdc2/cyclinB complex activity that is required for entry into M phase. This study has allowed us to confirm that algal virus infection leads to selective activation or inhibition of certain cell-cycle factors,which may play a significant role in establishing a more efficient environment for viral gene expression and DNA replication.

Key stakeholders’ perspectives on the ideal partnering culture in construction projects

National Lists of Scholarly Publication channels: An Overview and Recommendations for Their construction and Maintenance

Purpose: This paper presents an overview of different kinds of lists of scholarly publication channels and of experiences related to the construction and maintenance of national lists supporting performance-based research funding systems. It also contributes with a set of recommendations for the construction and maintenance of national lists of journals and book ***/methodology/approach: The study is based on analysis of previously published studies, policy papers, and reported experiences related to the construction and use of lists of scholarly publication channels. Findings: Several countries have systems for research funding and/or evaluation, that involve the use of national lists of scholarly publication channels(mainly journals and publishers). Typically, such lists are selective(do not include all scholarly or non-scholarly channels) and differentiated(distinguish between channels of different levels and quality). At the same time, most lists are embedded in a system that encompasses multiple or all disciplines. This raises the question how such lists can be organized and maintained to ensure that all relevant disciplines and all types of research are adequately represented. Research limitation: The conclusions and recommendations of the study are based on the authors' interpretation of a complex and sometimes controversial process with many different stakeholders *** implications: The recommendations and the related background information provided in this paper enable mutual learning that may feed into improvements in the construction and maintenance of national and other lists of scholarly publication channels in any geographical context. Bacterial community succession in response to dissolved organic matter released from live jellyfish

Jellyfish blooms have increased worldwide, and the outbreaks of jellyfish population not only affect the food web structures via voracious predation but also play an important role in the dynamics of nutrients and oxygen in planktonic food webs. However, it remains unclear whether specific carbon compounds released through jellyfish metabolic processes have the potential to shape bacterial community composition. Therefore, in this study, we aimed to investigate the compositional succession of the bacterioplankton community in response to the dissolved organic matter (DOM) released by the live Scyphomedusae cyanea lamarckii and chrysaora hysoscella collected from Helgoland Roads of the North Sea. The bacterial community was significantly stimulated by the DOM released form live jellyfish and different dominant phylotypes were observed for these two Scyphomedusae species. Furthermore, the bacterial community structures in the different DOM sources, jellyfish-incubated media, Kabeltonne seawater, and artificial seawater (DOM-free) were significantly different, as revealed by automated ribosomal intergenic spacer analysis fingerprints. catalyzed reporter deposition fluorescence in situ hybridization (cARD-FISH) revealed a rapid species-specific shift in bacterial community composition. Gammaproteobacteria dominated the community instead of the Bacteroidetes community for c. lamarckii, whereas Gammaproteobacteria and Bacteroidetes dominated the community for c. hysoscella. The significant differences in the bacterial community composition and succession indicate that the components of the DOM released by jellyfish might differ with jellyfish species.

Apoptosis in the testis has two putative roles during normal spermatogenesis; limitation of the germ cell population to numbers that can be supported by the Sertoli cells, and, possibly, selective depletion of meiotic and postmeiotic abnormal germ cells. We investigated the demographic and biological correlates of the pro-apoptotic marker Fas and the anti-apoptotic marker Bcl-xL in sperm cells of fertile men. Six hundred and four men from Greenland, Poland and Ukraine were consecutively enrolled during their pregnant wife＇s antenatal visits. Semen analysis was performed as recommended by the World Health Organization. Immunofluorescence coupled to flow cytometry was utilized for detection of apoptotic markers in the sperm cell. DNA damage was assessed by flow cytometry using both the sperm chromatin structure assay （ScSA） and the terminal deoxynucleotidyl transferase dUTP nick end labelling （TUNEL） assay. The percentage of Fas-positive sperm cells was higher in men with high total sperm count （P〈O.01）, more motile sperms （P=-O.04） and fewer sperm head defects （P=-O.05）. These associations were consistent within and across study regions. Furthermore, testosterone, follicle-stimulating hormone （FSH） and sexual hormone-binding globulin （SHBG） were significantly negatively correlated with Fas within and across regions as well. The data indicated no association between the anti-apoptotic Bcl-xL marker and semen or personal characteristics. The finding of Fas-positive sperm cells associated with better semen quality in a cohort of spouses of pregnant women seems different from previous data obtained in infertile men and warrants further investigation to clarify the biological significance of sperm apoptotic markers.

A comparison of conventional and computer-assisted semen analysis.（cRISMAS software） using samples from 166 young Danish men

The aim of the present study was to compare assessments of sperm concentration and sperm motility analysed by conventional semen analysis with those obtained by computer-assisted semen analysis （cASA）（copenhagen Rigshospitalet Image House Sperm Motility Analysis System （cRISMAS） 4.6 software） using semen samples from 166 young Danish men. The cRISMAS software identifies sperm concentration and classifies spermatozoa into three motility categories. To enable comparison of the two methods, the four motility stages obtained by conventional semen analysis were, based on their velocity classifications, divided into three stages, comparable to the three cRISMAS motility categories： rapidly progressive （A）, slowly progressive （B） and non-progressive （c＋ D）. Differences between the two methods were large for all investigated parameters （P〈0.001）. cRISMAS overestimated sperm concentration and the proportion of rapidly progressive spermatozoa and, consequently, underestimated the percentages of slowly progressive and non-progressive spermatozoa, compared to the conventional method. To investigate whether results drifted according to time of semen analysis, results were pooled into quarters according to date of semen analysis. cRISMAS motility results appeared more stable over time compared to the conventional analysis; however, neither method showed any trends. Apparently, cRISMAS cASA results and results from the conventional method were not comparable with respect to sperm concentration and motility analysis. This needs to be accounted for in clinics using this software and in studies of determinants of these semen characteristics.
