Investigation of HcAR2 antagonists as a potential strategy to modulate bovine leukocytes

Background Dairy cows experiencing ketosis after calving suffer greater disease incidence and are at greater risk of leaving the herd. In vitro administration of beta-hydroxybutyric acid(BHBA;the primary blood ketone) has inhibitory effects on the function of bovine leukocytes. BHBA is a ligand of HcAR2 and the activation of these receptors promotes an anti-inflammatory response which may be related with immunosuppression observed in transition dairy cattle. The objective of this study was to identify and test antagonists for HcAR2 in bovine immune cells cultured with *** We observed expression of HcAR2 at the protein level within lymphocytes, monocytes, and granulocytes. The proportion of cells expressing HcAR2 tended to be greater in mid-lactation compared to early lactation cows;the increase was a result of increased proportion of T and B cells expressing HcAR2. Stimulation of HcAR2 with niacin or BHBA promoted ca^(2+) mobilization in neutrophils and mononuclear cells. Mononuclear cells treated with BHBA had diminished intracellular ca^(2+) responses when HcAR2 was knocked down by si RNA silencing, indicating ca^(2+) mobilization was mediated by HcAR2 signaling. Two candidate antagonists for HcAR2, synthesized from niacin(NA-1 and NA-5), were tested;monocytes and neutrophils pre-treated with NA-1 and NA-5 had reduced ca^(2+) mobilization after incubation with BHBA. Furthermore, NA-5 but not NA-1 prevented BHBA-associated reductions in cyclic *** We demonstrated that HcAR2 is present on bovine leukocytes and has greater expression later in lactation. We confirmed that BHBA and niacin derived HcAR2 antagonists alter bovine leukocyte activity. Our results demonstrate that both BHBA and niacin affect bovine leukocyte ca^(2+) mobilization in a HcAR2-dependent manner.

Isolating higher yielding and more stable rice genotypes in stress environments: fine-tuning a selection method using production and resilience score indices

In Asia, the rice crop sustains millions of people. However, growing demand for this crop needs to be met while simultaneously reducing its water consumption to cope with the effects of climate change. Lowland cropping systems are the most common and productive but have particularly high water requirements. High-yielding rice genotypes adapted to drier environments(such as rainfed or aerobic rice ecosystems) are needed to increase the water use efficiency of cropping. Identifying these genotypes requires fast and more accurate selection methods. It is hypothesized that applying a new quantitative selection method(the score index selection method), can usefully compare rice yield responses over different years and stress intensities to select genotypes more rapidly and efficiently. Applying the score index to previously published rice yield data for 39 genotypes grown in no-stress and two stress environments, identified three genotypes(ARB 8, IR55419-04 and ARB 7) with higher and stable yield under moderate to severe stress conditions. These genotypes are postulated to be better adapted to stress environment such as upland and aerobic environments. Importantly, the score index selection method offers improved precision than the conventional breeding selection method in identifying genotypes that are well-suited to a range of stress levels within the target environment.

Synchronized off-harmonic probe laser with highly variable pulse duration for laser-plasma interaction experiments

This paper presents the development and experimental utilization of a synchronized off-harmonic laser system designed as a probe for ultra-intense laser±plasma interaction experiments. The system exhibits a novel seed-generation design,allowing for a variable pulse duration spanning over more than three orders of magnitude, from 3.45 picoseconds to 10 nanoseconds. This makes it suitable for various plasma diagnostics and visualization techniques. In a side-view configuration, the laser was employed for interferometry and streaked shadowgraphy of a laser-induced plasma while successfully suppressing the self-emission background of the laser±plasma interaction, resulting in a signal-to-self-emission ratio of 110 for this setup. These properties enable the probe to yield valuable insights into the plasma dynamics and interactions at the PHELIX facility and to be deployed at various laser facilities due to its easy-to-implement design.

In situ digital testing method for quality assessment of soft soil improvement with polyurethane

This study purposes an in situ testing method on quality assessment of soil *** drilling data includes the spatial distribution and in situ strength of untreated and treated soil along three different drillholes measured by on-site drilling monitoring *** factual drilling data can characterize the degree of soil improvement by penetration injection with permeable *** from on-site drilling monitoring shows that the linear zones represent constant drilling speeds shown in the plot of drill bit advancement *** drilling time,which indicates the spatial distributions of soil *** soil profile at the study site is composed of four layers,which includes fill,untreated silty clay,treated silty clay,and mucky *** results of soil profile are verified by the parallel site *** constant drilling speeds profile the coring-resistant strength of drilled *** comparing with the untreated silty clay,the constant drilling speeds of the treated silty clay have been decreased by 13.0-62.8%.Two drilling-speed-based indices of 61.2%and 65.6%are proposed to assess the decreased average drilling speed and the increased in situ strength of treated silty *** tests,*** compressive strength(UcS)test,have been performed with core sample to investigate and characterize in situ strength by comparing that with drilling *** show that the average predicted strengths of treated silty clay are 2.4-6.9 times higher than the average measured strength of untreated silty *** UcS-based indices of 374.5%and 344.2%verified the quality assessment(QA)results by this new in situ *** method provides a cost-effective tool for quality assessment of soil improvement by utilizing the digital drilling data.

BAcKGROUND There is concern regarding potential long-term cardiotoxicity with systemic distribution of metals in total joint arthroplasty(TjA)*** To determine the association of commonly used implant metals with echocardiographic measures in TjA *** The study comprised 110 TjA patients who had a recent history of high chromium,cobalt or titanium *** underwent two-dimensional,three-dimensional,Doppler and speckle-strain transthoracic echocardiography and a blood draw to measure metal *** and sex-adjusted linear and logistic regression models were used to examine the association of metal concentrations(exposure)with echocardiographic measures(outcome).RESULTS Higher cobalt concentrations were associated with increased left ventricular end-diastolic volume(estimate 5.09;95%cI:0.02-10.17)as well as left atrial and right ventricular dilation,particularly in men but no changes in cardiac *** titanium concentrations were associated with a reduction in left ventricle global longitudinal strain(estimate 0.38;95%cI:0.70 to 0.06)and cardiac index(estimate 0.08;95%cI,-0.15 to-0.01).cONcLUSION Elevated cobalt and titanium concentrations may be associated with structural and functional cardiac changes in some *** studies are warranted to better understand the systemic effects of metals in TjA patients.

HI in High Gas-phase Metallicity Dwarf Galaxy WISEA j230615.06+143927.9

We present resolved Giant Metrewave Radio Telescope H I observations of the high gas-phase metallicity dwarf galaxy WISEA j230615.06+143927.9(z = 0.005)(hereafter j2306) and investigate whether it could be a Tidal Dwarf Galaxy(TDG) candidate. TDGs are observed to have higher metallicities than normal dwarfs. j2306 has an unusual combination of a blue g-r color of 0.23 mag, irregular optical morphology and high-metallicity(12 +log(O/H) = 8.68 ± 0.14), making it an interesting galaxy to study in more detail. We find j2306 to be an H I rich galaxy with a large extended, unperturbed rotating H I disk. Using our H I data we estimated its dynamical mass and found the galaxy to be dark matter(DM) dominated within its H I radius. The quantity of DM, inferred from its dynamical mass, appears to rule out j2306 as an evolved TDG. A wide area environment search reveals j2306 to be isolated from any larger galaxies which could have been the source of its high gas metallicity. Additionally, the H I morphology and kinematics of the galaxy show no indication of a recent merger to explain the *** detailed optical spectroscopic observations of j2306 might provide an answer to how a seemingly ordinary irregular dwarf galaxy achieved such a high level of metal enrichment.

Polio eradication surveillance in Sri Lanka,2019-2023

Objective:To evaluate the polio laboratory surveillance carried out from january,2019 to May,2023 by the Polio Regional Reference Laboratory,Sri ***:This retrospective study analyzed all stool samples received under the acute flaccid paralysis(AFP)and immunodeficient vaccine-derived poliovirus(VDPV)surveillance at Polio Regional Reference Laboratory,Sri Lanka from january,2019 to May,*** results of the testing methodologies were extracted from the laboratory data system,i.e.,poliovirus virus isolation,intra-typic differentiation/VDPV real time reverse transcriptase polymerase chain reaction(ITD/VDPV rRTPcR)and sequencing,along with the data on timing of reporting results,stool adequacy and *** was analyzed using descriptive ***:A total of 2141 stool samples from 1644 cases were received for AFP surveillance from Sri Lanka(93.61%),Maldives(1.52%),and immunodeficient VDPV(4.86%)*** polioviruses(19/1644,1.15%)and non-polio enteroviruses(73/1644,4.44%)were isolated,while Sabin-like 3 virus was detected in majority(12/19,63.15%)among the poliovirus *** polioviruses or circulating VDPVs were not detected among the *** all years of the study,the non-polio AFP detection rate was>1/100000 in children aged less than 15 years,whereas stool adequacy rate was>80%.All results were reported within 14 days of receipt,ensuring timely reporting as per global ***:The Polio Regional Reference Laboratory,Sri Lanka plays a vital role in maintaining the polio-free status in the country through its robust laboratory surveillance,while adhering to the surveillance ***-detection of wild polioviruses and circulating VDPV during the study period reinforces the polio-free status in the country.

A Sharding Scheme Based on Graph Partitioning Algorithm for Public Blockchain

Blockchain technology,with its attributes of decentralization,immutability,and traceability,has emerged as a powerful catalyst for enhancing traditional industries in terms of optimizing business ***,transaction performance and scalability has become the main challenges hindering the widespread adoption of *** to its inability to meet the demands of high-frequency trading,blockchain cannot be adopted in many *** improve the transaction capacity,researchers have proposed some on-chain scaling technologies,including lightning networks,directed acyclic graph technology,state channels,and shardingmechanisms,inwhich sharding emerges as a potential scaling ***,excessive cross-shard transactions and uneven shard workloads prevent the sharding mechanism from achieving the expected *** paper proposes a graphbased sharding scheme for public blockchain to efficiently balance the transaction *** crossshard transactions and evening-out workloads among shards,the scheme reduces transaction confirmation latency and enhances the transaction capacity of the ***,the scheme can achieve a high-frequency transaction as well as a better blockchain *** results show that the scheme effectively reduces the cross-shard transaction ratio to a range of 35%-56%and significantly decreases the transaction confirmation latency to 6 s in a blockchain with no more than 25 shards.

cO oxidation under lean and stoichiometric conditions over ceriazirconia with very low metal contents(cu,co,Ag and Pt)

In this work,several ceria-zirconia based catalysts with very lo w（and equimolar） metal contents were prepared,characterised and tested for the cO oxidatio n reaction（under lean and stoichiometric conditions）,trying to emulate those conditions found in a diesel oxidation catalyst（DOc） system from a diesel engine and those encountered under gasoline exhaust（λ=1）.The metals chosen are cu,co,Ag and Pt（as a reliable benchmark）.The results reveal enormous differences among reducibility and catalytic activity despite quite similar structural and textural properties of the catalysts,showing differences among dispersion（Ag-catalyst seems to present a low level of dispersion）.This catalyst seems to be characterised,as well,by a strong electronic interaction between ce and Ag centres which is suggested to yield an improved reducibility under H2-TPR ***,the order in catalytic activity（cu>Ag>co≈Pt>>support） seems not to follow the order found in reducibility and the cu-catalyst seems to be the most active independently on the reaction conditions,yielding nearly overlapped cO oxidation catalytic *** restingly,a stro ng correlation between the catalytic activity under the two conditions tested and the OSc values of the ce0.8Zr0.2O2-supported metal catalysts is ***,OSc parameter measured at 150℃ can be used as a relevant descriptor to evaluate the cO oxidation activity at low and medium conversions for the investigated catalysts,much better than the H2-TPR measurements.

Land use change and occupation have led to modifications in the environment causing loss of biodiversity and ecosystem services throughout the *** environments with high economic relevance,such as the ferruginous campo rupestre(rupestrian grassland known as canga in Brazil),are even more susceptible to severe impacts due to their extreme habitat conditions and low *** determination of reference ecosystems based on the intrinsic characteristics of the ecosystem is essential for conservation as well as to the implementation of ecological *** proposed the reference ecosystem of the three main types of habitats of the ferruginous campo rupestre based on their floristic *** described the floristic composition of each habitat and evaluated the physicochemical properties of the soils and the relationship between plants and *** three habitats showed high diversity of plant species and many endemic species,such as chamaecrista choriophylla,cuphea pseudovaccinium,Lychnophora pinaster,and Vellozia *** distribution of vegetation was strongly related with the edaphic characteristics,with a set of species more adapted to high concentration of base saturation,fine sand,organic carbon,and iron,while another set of species succeeded in more acidic soils with higher S and silt *** provide support for the contention that the ferruginous campo rupestre is a mosaic of different habitats shaped by intrinsic local *** to recognize the floristic composition of each particular habitat can lead to inappropriate restoration,increased habitat homogenization and increased loss of biodiversity and ecosystem *** study also advances the knowledge base for building the reference ecosystem for the different types of ferruginous campo rupestre habitats,as well as a key database for highlighting those species contribute most to community assembly in this diverse and threatened tropical mountain ecosystem.
