Blood metal concentrations and cardiac structure and function in total joint arthroplasty patients

BAcKGROUND There is concern regarding potential long-term cardiotoxicity with systemic distribution of metals in total joint arthroplasty(TJA)*** To determine the association of commonly used implant metals with echocardiographic measures in TJA *** The study comprised 110 TJA patients who had a recent history of high chromium,cobalt or titanium *** underwent two-dimensional,three-dimensional,Doppler and speckle-strain transthoracic echocardiography and a blood draw to measure metal *** and sex-adjusted linear and logistic regression models were used to examine the association of metal concentrations(exposure)with echocardiographic measures(outcome).RESULTS Higher cobalt concentrations were associated with increased left ventricular end-diastolic volume(estimate 5.09;95%cI:0.02-10.17)as well as left atrial and right ventricular dilation,particularly in men but no changes in cardiac *** titanium concentrations were associated with a reduction in left ventricle global longitudinal strain(estimate 0.38;95%cI:0.70 to 0.06)and cardiac index(estimate 0.08;95%cI,-0.15 to-0.01).cONcLUSION Elevated cobalt and titanium concentrations may be associated with structural and functional cardiac changes in some *** studies are warranted to better understand the systemic effects of metals in TJA patients.

Obesity has a negative effect on male reproductive function. It is associated with low testosterone levels and alteration in gonadotropin secretion. Male obesity has been linked to reduced male fertility. Data regarding the relation of obesity to sperm parameters are conflicting in terms of the nature and magnitude of the effect. New areas of interest are emerging that can help explain the variation in study results, such as genetic polymorphism and sleep apnea. Sleep disorders have been linked to altered testosterone production and hypogonadism in men. It was also correlated to erectile dysfunction. The relation of sleep disorders to male fertility and sperm parameters remains to be investigated. Men with hypogonadism and infertility should be screened for sleep apnea. Treatment of obesity and sleep apnea improves testosterone levels and erectile function.

Association of 25-hydroxy-vitamin D levels with semen and hormonal parameters

Vitamin D levels have been linked to various health outcomes including reproductive disorders. The purpose of this study was to explore the association between serum vitamin D level （25-hydroxy-vitamin D, or 250HD） and semen and hormonal parameters. This is a cross-sectional study that included 170 healthy men recruited for the study of spermatogenesis from the general population. Men completed general and reproductive health questionnaires, and donated blood and semen samples. The main measures were hormonal （total and free testosterone, sex hormone-binding globulin, estradiol, follicle-stimulating hormone and luteinizing hormone） and semen parameters, adjusted （n= 147） for age, body mass index （BMI）, season, alcohol intake and smoking, in relation to categories of vitamin D levels, determined apriori. The mean age of the study population was 29.0±8.5 years and mean BMI was 24.3±3.2 kg m-2. The mean 250HD was 34.1± 15.06 ng m1-1. BMI showed a negative association with 250HD. Sperm concentration, sperm progressive motility, sperm morphology, and total progressively motile sperm count were lower in men with ‘250HD ≥ 50 ng ml-1＇ when compared to men with‘20 ng ml-1 ≤ 250H D〈 50 ng ml-1,. Total sperm count and total progressive motile sperm count were lower in men with ‘250HD〈20 ng ml-1＇ when compared to men with‘20 ng ml-1≤250HD〈50 ng ml-1＇. The adjusted means of various hormonal parameters did not show statistical difference in the different categories of 250HD. In conclusion, serum vitamin D levels at high and low levels can be negatively associated with semen parameters.

Detecting and Adjusting Temporal Inhomogeneity in chinese Mean Surface Air Temperature Data

Adopting the Easterling-peterson (EP) techniques and considering the reality of chinese meteorological observations, this paper designed several tests and tested for inhomogeneities in all chinese historical surface air temperature series from 1951 to 2001. The result shows that the time series have been widely impacted by inhomogeneities resulting from the relocation of stations and changes in local environment such as urbanization or some other factors. Among these factors, station relocations caused the largest magnitude of abrupt changes in the time series, and other factors also resulted in inhomogeneities to some extent. According to the amplitude of change of the difference series and the monthly distribution features of surface air temperatures, discontinuities identified by applying both the E-P technique and supported by china's station history records, or by comparison with other approaches, have been adjusted. Based on the above processing, the most significant temporal inhomogeneities were eliminated, and china's most homogeneous surface air temperature series has thus been created. Results show that the inhomogeneity testing captured well the most important change of the stations, and the adjusted dataset is more reliable than ever. This suggests that the adjusted temperature dataset has great value of decreasing the uncertaities in the study of observed climate change in china.

Mining diversity of the natural biorefinery housed within Tipula abdominalis larvae for use in an industrial biorefinery for production of lignocellulosic ethanol

Although they are the largest taxonomic group of animals, relatively few insects have been examined for symbiotic relationships with micro-organisms. However, this is rapidly changing because of the potential for examination of the natural insectmicrobe-lignocellulose interactions to provide insights for biofuel technology. Micro-organisms associated with lignocellulose-consuming insects often facilitate the digestion of the recalcitrant plant diet; therefore these microbial communities may be mined for novel lignocellulose-degrading microbes, or for robust and inexpensive biocatalysts necessary for economically feasible biofuel production from lignocellulose. These insect-microbe interactions are influenced by the ecosystem and specific lignocellulose diet, and appre- ciating the whole ecosystem-insect-microbiota-lignocellulose as a natural biorefinery provides a rich and diverse framework from which to design novel industrial processes. One such natural biorefinery, the Tipula abdominalis larvae in riparian ecosystems, is reviewed herein with applications for biochemical processes and overcoming challenges involved in conversion of lignocellulosic biomass to fuel ethanol. From the dense and diverse T. abdominalis larval hindgut microbial community, a cellulolytic bacterial isolate, 27c64, demonstrated enzymatic activity toward many model plant polymers and also produced a bacterial antibiotic. 27c64 was co-cultured with yeast in fermentation of pine to ethanol, which allowed for a 20% reduction of commercial enzyme. In this study, a micro-organism from a lignocellulose-consuming insect was successfully applied for improvement of biomass-to-biofuel technology.

Analysis of cellulolytic and hemicellulolytic enzyme activity within the Tipula abdominalis （Diptera： Tipulidae） larval gut and characterization of crocebacterium ilecola gen. nov., sp. nov., isolated from the Tipula abdominalis larval hindgut

In forested stream ecosystems of the north and eastern United States, larvae of the aquatic crane fly Tipula abdominalis are important shredders of leaf litter detritus. T. abdominalis larvae harbor a dense and diverse microbial community in their hindgut that may aide in the degradation oflignocellulose. In this study, the activities ofcellulolytic and hemicellulolytic enzymes were demonstrated from hindgut extracts and from bacterial isolates using model sugar substrates. One of the bacterial isolates was further characterized as a member of the family Microbacteriaceae. Taxonomic position of the isolate within this family was determined by a polyphasic approach, as is commonly employed for the separation of genera within the family Microbacteriaceae. The bacterial isolate is Gramtype positive, motile, non-sporulating, and rod-shaped. The G ＋ c content of the DNA is 64.9 mol%. The cell wall contains B2y type peptidoglycan, D- and L-diaminobutyric acid as the diamino acid, and rhamnose as the predominant sugar. The predominant fatty acids are 12-methyltetradecanoic acid （ai-c15：0） and 14-methylhexadecanoic acid （ai-c17：0）. The isolate forms a distinct lineage within the family Microbacteriaceae, as determined by 16S rRNA sequence analysis. We propose the name crocebacterium ilecola gen. nov., sp. nov., to accommodate this bacterial isolate. The type species is T202T （ATcc BAA-1359; GenBank Accession DQ826511）.

Novel UBE2B-associated polymorphisms in an azoospermic / oligozoospermic population

Aim： To assess whether abnormalities exist in the UBE2B gene in a population of infertile human males, and to establish biologic plausibility of any discovered mutations. Methods： We carried out polymerase chain reaction （PcR） amplification and sequence analysis of the 5＇-untranslated region and six exons of the UBE2B gene, including flanking intronic regions, in a group of fertile and infertile men. Following the identification of a putative promoter region that contained single or dual triplet deletions within a 10-cGG repeat island, we evaluated the binding affinity of these identified polymorphisms as compared to the wild-type sequence to transcription factor SP1 using a DNA-protein gel shift assay. Results： There was a novel exonic single nucleotide polymorphism （SNP） noted in exon 4 in 5% of infertile men. In silico 3D modeling of the altered protein showed an innocuous isoleucine for valine substitution. There were no mutations noted within any of the other exons. Three novel intronic SNPs were identified within the fertile group, and seven novel intronic SNPs identified in the infertile group. The DNA-protein gel shift assay noted that both single cGG deletion and double cGG deletion bands had approximately twice the binding affinity compared to the wild-type for SP1. The negative control confirmed no non-specific protein binding. conclusion- By themselves, a single or double cGG deletion is unlikely to pose biologic significance. However, such deletions in this suspected promoter region are associated with increased binding affinity for SP1, and might represent one of several factors required for alteration of UBE2B gene expression.

A Totally Absorbable Multilayer PLGA Implant Device containing Doxorubicin Inhibited Tumor Growth and Metastasis without Systemic Toxicity in Murine Breast cancer and an Ideal Pharmacological Paradigm for Regional chemotherapy

We hypothesize that a cylinder implant made of multilayer Poly-lactic-co-glycolic-acid (PLGA) membrane can be a method for controlled and extended drug release. We fashioned a multilayer cylindrical implant termed STID100 that released doxorubicin for 3 weeks in an orthotopic 4T1 breast cancer model in Balb/c mice. This implant starts as a thin doxorubicin-embedded PLGA membrane, and is then rolled into a cylinder containing an air gap between the membrane layers. Its controlled sustained release delivered 2× the amount of the intravenous (IV) equivalent of doxorubicin, inhibited the primary tumor, and prevented lung metastasis. Importantly it did not cause weight loss, splenomegaly, or cardiac toxicity vs systemically administrated doxorubicin. This favorable safety profile is further substantiated by the finding of no detectable plasma doxorubicin in multiple time points during the 3-week period, and low tumor doxorubicin concentration. The implant system delivered to the specification of an ideal pharmacological paradigm might offer a better coverage of the local tumor, significantly preventing metastatic spread with less drug toxicity to many vital organs, compared to the traditional pharmacology of IV route. The profile of STID made it an attractive therapeutic alternative in metastatic tumor prevention, pain management and many other diverse clinical scenarios.
