In vitro activity and in vivo efficacy of a combination therapy of diminazene and chloroquine against murine visceral leishmaniasis

The present study evaluated the in vitro activity and in vivo efficacy of diminazene combined with chloroquine as a potential drug against leishmania donovani. Amphotericin B was used as a positive control drug. In vitro activity involved incubation of various drug concentrations with promastigotes or vero cells in culture before determination of parasite growth inhibition or cell death while in vivo evaluations involved infection of various mice groups with virulent l. donovani parasites and treatment with test drug compounds following disease establishment. Weight changes in experimental mice were also evaluated before infection and throughout the experiment. The results indicated that the diminazene-chloroquine combination was at least nine times more efficacious than individual drugs in killing promastigotes in culture. The diminazene-chloroquine combination was safer （ld50=0.03±0.04） than Amphotericin B （ld50=0.02±0.01）. Body weight in infected mice increased significantly （p=0.0007） from day 7 to day 37 following infection （p=0.026）. However, body weight remained comparable in all mice groups during treatment （p=0.16）. The diminazene-chloroquine combination significantly reduced splenic parasite numbers as compared to individual drug therapies （p=0.0001） although Amphotericin B was still more efficacious than any other treatment （p=0.0001）. Amongst the test compounds, the diminazene-chloroquine combination showed the lowest level of IgG antibody responses with results indicating significant negative correlation between antileishmanial antibody responses and protection against disease. These findings demonstrate the positive advantage and the potential use of a combined therapy of diminazene-chloroquine over the constituent drugs. Further evaluation is recommended to determine the most efficacious combination ratio of the two compounds.

This paper presents a bridging research between a modeling methodology in various fields in physics and *** the symplectic method first applied for group symmetry in particle physics,a new symplectic elasticity approach is developed for deriving exact analytical solutions to some basic problems in solid mechanics and elasticity which have long been bottlenecks in the history of *** the Hamiltonian principle with legendre’s transformation,it solves bending of thick composite beams with mechanical and electric loadings where exact solutions are hitherto unavailable. Analytical bending solutions could be obtained by eigenvalue analysis and expansion of eigenfunctions. Here,bending analysis requires the solving of an eigenvalue equation unlike in classical mechanics where eigenvalue analysis is only required in vibration and buckling ***,unlike the semi-inverse approaches in classical plate analysis employed by Timoshenko and others such as Navier’s solution,levy’s solution,Rayleigh-Ritz method,*** a trial deflection function is pre-determined, this new symplectic plate analysis is completely rational without any guess functions and yet it renders exact solutions beyond the scope of applicability of the semi-inverse *** short,the symplectic approach developed here presents solutions of analytical mechanics unaccountable by classical mechanics. ＜正＞ Ecto - nucleotidases of the cD39/E - NTpDase family are expressed in the vasculatureand immune *** ecto - enzymes hydrolyze extracellular nucleotides,ultimately to the respective nucleosides,to regulate p2 - receptor signaling . Spatial and temporal expression of cD39/NTpDasel by vascular and immune cells could regulate thrombotic and immune reactions in vivo. cD39 has the potential to modulate thrombotic reactions viz. platelet activation after ischemia reperfusion in vivo. Increases of NTpDasel biochemical activity within microparticles associated with evolving arteriolar thrombi also seems to impede further ADp - mediated platelet activation. cD39 is also a surface marker of Tregulatory cells （Treg）. co - ordinated expression of cD39 on Treg and the adenosine A2A receptor on activated effector T cells （Teff） generates an immunosuppressiveloop. Adoptive transfer of cd39 null Treg fails to inhibit allograftrejection in vivo and null mice also develop autoimmune manifestations and exhibit vascular thrombophilia. pharmacologic modalities to modulate or boost NTpDasel expression may suppress deleterious vascular or immune reactions,as seen in autoimmune disease and transplant graft rejection.

In this work,an attempt has been made for optimization of process parameters in Wire Electric Discharge Machining(WEDM)of Ti–6Al–4V while producing square and cir-cular *** input parameters,namely pulse on time,pulse off time,peak current and servo voltage,were considered to study the responses cutting speed(cS)and sur-face roughness(SR).Each input parameter was set at three *** were conducted as per central composite face(ccF)centered *** upon the exper-imental data,Gray relational analysis(GRA),a multi-objective optimization technique has been employed to find the best level of process parameters to optimize the machining *** of variance(ANOVA)has been conducted for investigating the effect of process parameters on overall machining ***,it was identified that the process parameters such as pulse on time,current and voltage have more impact on the square and circular profiles.

endotoxins(also known as lipopolysaccharides(lpS)) are undesirable by-products of recombinant proteins,purified from Escherichia *** can be considered stable under a wide range of temperature and pH,making their removal one of the most difficult tasks in downstream processes during protein *** inherent toxicity of lpS makes their removal an important step for the application of these proteins in several biological assays and for a safe parenteral *** metal affinity chromatography(IMAc) enables the affinity interactions between the metal ions(immobilized on the support through the chelating compound) and the target molecules,thus enabling high-effic.ency separation of the target molecules from other components present in a *** chromatography is applied with ca2+-iminodiacetic acid(IDA) to remove most of the lpS contaminants from the end product(more than90%).In this study,the adsorption of lpS on an IDA-ca2+ was *** adsorption Freundlich isotherm of lpS-IDA-ca2+ provides a theoretical basis for lpS *** was found that lpS is bound mainly by interactions between the phosphate group in lpS and ca2+ ligands on the *** factors such as pH(4.0 or 5.5) and ionic strength(1.0 mol/l) are essential to obtain effective removal of lpS for contaminant levels between endotoxin' concentration values less than100 EU/ml and 100 000 EU/*** new protocol represents a substantial advantage in time,effort,and production costs.

Studying nucleon-nucleon （NN） correlated pairs will teach us a great deal about the high momentum part of the nuclear wave function,the short range part of the NN interaction,and the nature of cold dense nuclear *** correlations are similar in all nuclei,differing only in *** momentum nucleons,p 〉p fermi,all have a correlated partner with approximately equal and opposite *** pair relative momenta of 300 〈prel 〈500 MeV/c,these correlated pairs are dominated by tensor *** is shown by the dominance of pn over pp pairs at pair total momentum and by the parity of pn to pp pairs at large pair total momentum.
