Spatial Distribution and Release Flux of carbon and Nitrogen components in Glacial Watersheds of Eastern Tibetan Plateau

The carbon cycle stands as a cornerstone among the Earth's material cycles， carrying profound implications for global climate dynamics and humanity's sustenance and advancement. Additionally， nitrogen， as a vital nutrient element， plays a pivotal role in biogeochemical processes， exerting significant influence on the ecological environment within watersheds. In this study， an analysis of the carbon （c） and nitrogen （N） components in the river water of 12 alpine basins on the eastern margin of the Tibetan Plateau （TP） reveals that the average dissolved organic carbon （DOc） concentration in the river water is 0.76±0.67 mg/L， indicating a lower level compared to the global river average organic carbon content. Specifically， the average DOc concentrations were 0.42±0.22 mg/L and 1.56±0.68 mg/L in the glacial and non-glacial basins， respectively. The average dissolved inorganic carbon （DIc） concentration in the eastern TP is 27.04±20.19 mg/L， with the lowest DIc observed at Dagu Glacier and the highest at Daxia River. Furthermore， the average dissolved total carbon （DTc） concentration in glacier river water was 18.87±12.65 mg/L， contrasting with 48.23±21.07 mg/L in non-glacial basins. The mean concentration of total nitrogen （TN） in the eastern TP averaged 0.97±0.54 mg/L， with the glacial basin registering 1.01±0.47 mg/L and the non-glacial basin at 0.89±0.70 mg/L. Evaluating the release fluxes of carbon and nitrogen across 12 river basins revealed a range of DTc emission fluxes from 0.32 to 31.02 × 109g c/a， with inorganic carbon comprising 71.0% to 99.5%. consequently， the carbon emission flux for seven glacier basins totaled 15.91 × 109g c/a， while for the five non-glacial river basins， it reached 87.37× 109g c/a. The total TN release fluxes across 12 small river basins amounted to 2.46 × 109g N/a， with 0.84 × 109g N/a released in glacial basins and 1.62 × 109g N/a in the non-glacial basin. Protein tyrosine kinases (RTKs) modulate a wide range of pathophysiological events in several non-malignant disorders, including diabetic complications. To find new targets driving the development of diabetic cardiomyopathy (DcM), we profiled an RTKs phosphorylation array in diabetic mouse hearts and identified increased phosphorylated fibroblast growth factor receptor 1 (p-FGFR1) levels in cardiomyocytes, indicating that FGFR1 may contribute to the pathogenesis of DcM. Using primary cardiomyocytes and H9c2 cell lines, we discovered that high-concentration glucose (HG) transactivates FGFR1 kinase domain through toll-like receptor 4 (TLR4) and c-Src, independent of FGF ligands. Knocking down the levels of either TLR4 or c-Src prevents HG-activated FGFR1 in cardiomyocytes. RNA-sequencing analysis indicates that the elevated FGFR1 activity induces pro-inflammatory responses via MAPKs–NFκB signaling pathway in HG-challenged cardiomyocytes, which further results in fibrosis and hypertrophy. We then generated cardiomyocyte-specific FGFR1 knockout mice and showed that a lack of FGFR1 in cardiomyocytes prevents diabetes-induced cardiac inflammation and preserves cardiac function in mice. Pharmacological inhibition of FGFR1 by a selective inhibitor, AZD4547, also prevents cardiac inflammation, fibrosis, and dysfunction in both type 1 and type 2 diabetic mice. These studies have identified FGFR1 as a new player in driving DcM and support further testing of FGFR1 inhibitors for possible cardioprotective benefits.

Passive neutron multiplicity device for^(240)Pu measurement based on FPGA

A passive neutron multiplicity measurement device,FH-NcM/S1,based on field-programmable gate arrays(FPGAs),is developed specifically for measuring the mass of plutonium-240(^(240)Pu)in mixed oxide ***-NcM/S1 adopts an inte-grated approach,combining the shift register analysis mode with the pulse-position timestamp mode using an *** optimal effective length of the^(3)He neutron detector was determined to be 30 cm,and the thickness of the graphite reflector was ascertained to be 15 cm through McNP *** fabricating the device,calibration measurements were per-formed using a^(252)cf neutron source;a detection efficiency of 43.07%and detector die-away time of 55.79μs were *** samples of plutonium oxide were measured under identical conditions using the FH-NcM/S1 in shift register analysis mode and a plutonium waste multiplicity *** obtained double rates underwent corrections for detection efficiency(ε)and double gate fraction(f_(d)),resulting in corrected double rates(D_(c)),which were used to validate the accuracy of the shift register analysis ***,the device exhibited fluctuations in the measurement results,and within a single 20 s measurement,these fluctuations remained below 10%.After 30 cycles,the relative error in the mass of^(240)Pu was less than 5%.Finally,correlation calculations confirmed the robust consistency of both measurement *** study holds specific significance for the subsequent design and development of neutron multiplicity devices.

The nucleus of Darkschewitsch(ND), mainly composed of GABAergic neurons, is widely recognized as a component of the eye-movement controlling system. However, the functional contribution of ND GABAergic neurons(NDGABA) in animal behavior is largely unknown. Here, we show that NDGABA neurons were selectively activated by different types of fear stimuli, such as predator odor and foot *** and chemogenetic manipulations revealed that NDGABA neurons mediate freezing ***, using circuit-based optogenetic and neuroanatomical tracing methods, we identified an excitatory pathway from the lateral periaqueductal gray(l PAG) to the ND that induces freezing by exciting ND inhibitory outputs to the motor-related gigantocellular reticular nucleus, ventral part(Gi V). Together,these findings indicate the NDGABA population as a novel hub for controlling defensive response by relaying fearful information from the l PAG to Gi V, a mechanism critical for understanding how the freezing behavior is encoded in the mammalian brain.

Magnesium lithospermate B enhances the potential of human-induced pluripotent stem cell-derived cardiomyocytes for myocardial repair

Background: We previously reported that activation of the cell cycle in human-induced pluripotent stem cell-derived cardiomyocytes (hiPSc-cMs) enhances their remuscularization capacity after human cardiac muscle patch transplantation in infarcted mouse hearts. Herein, we sought to identify the effect of magnesium lithospermate B (MLB) on hiPSc-cMs during myocardial repair using a myocardial infarction (MI) mouse ***: In c57BL/6 mice, MI was surgically induced by ligating the left anterior descending coronary artery. The mice were randomly divided into five groups (n = 10 per group); a MI group (treated with phosphate-buffered saline only), a hiPSc-cMs group, a MLB group, a hiPSc-cMs + MLB group, and a Sham operation group. cardiac function and MLB therapeutic efficacy were evaluated by echocardiography and histochemical staining 4 weeks after surgery. To identify the associated mechanism, nuclear factor (NF)-κB p65 and intercellular cell adhesion molecule-1 (IcAM1) signals, cell adhesion ability, generation of reactive oxygen species, and rates of apoptosis were detected in human umbilical vein endothelial cells (HUVEcs) and ***: After 4 weeks of transplantation, the number of cells that engrafted in the hiPSc-cMs + MLB group was about five times higher than those in the hiPSc-cMs group. Additionally, MLB treatment significantly reduced tohoku hospital pediatrics-1 (THP-1) cell adhesion, IcAM1 expression, NF-κB nuclear translocation, reactive oxygen species production, NF-κB p65 phosphorylation, and cell apoptosis in HUVEcs cultured under hypoxia. One-year results for myopia control of orthokeratology with different back optic zone diameters: a randomized trial using a novel multispectral-based topographer
