Legionella dumoffii Tex-KL Mutated in an Operon Homologous to trac-traD is Defec.i.e in Epithelial cell invasion

Objec.i.e To understand the mechanism of invasion by Legionella dumoffii. Methods The L. dumoffii strain Tex-KL was mutated using the Tn903 derivative, Tn903 d iilac Z. After screening 799 transposon insertion mutants, we isolated one defec.i.e mutant. We then constructed the gene-disrupted mutant, KL16, and studied its invasion of and intracellular growth in He La and A549 cells, and in A/J mice survival experiments. The structure of trac-traD operon was analyzed by RT-PcR. Results The transposon insertion was in a gene homologous to Salmonella typhi tra c, which is required for the assembly of F pilin into the mature F pilus structure and for conjugal DNA transmission. Results from RT-PcR suggested that the trac-traD region formed an operon. We found that when the tra c gene was disrupted, invasion and intracellular growth of L. dumoffii Tex-KL were impaired in human epithelial cells. When mice were infected by intranasal inoculation with a tra c deficient mutant, their survival significantly increased when compared to mice infected with the wild-type strain. conclusion Our results indicated that the trac-traD operon is required for the invasion and intracellular growth abilities of L. dumoffii Tex-KL in epithelial cells.

New approach of medicinal herbs and sulfasalazine mixture on ulcerative colitis induced by dextran sodium sulfate

BAcKGROUND Sulfasalazine has been used as a standard-of-care in ulcerative colitis for decades,however,it results in severe adverse symptoms,such as hepatotoxicity,blood disorders,male infertility,and ***,the new treatment strategy has to enhance pharmacological efficacy and stimultaneously minimize side *** To compare the anti-inflammatory ac.i.n of sulfasalazine alone or in combination with herbal medicine for ulcerative colitis in a dextran sodium sulfate(DSS)-induced colitis mouse *** To induce ulcerative colitis,mice rec.i.ed 5%DSS in drinking water for 7 *** were divided into five groups(n=9 each)for use as normal(non-DSS),DSS controls,DSS+sulfasalazine(30 mg/kg)-treatment experimentals,DSS+sulfasalazine(60 mg/kg)-treatment experimentals,DSS+sulfasalazine(30 mg/kg)+citrus unshiu peel and Bupleuri radix mixture(30 mg/kg)(ScPB)-treatment *** The ScPB treatment showed an outstanding effec.i.eness in counterac.i.g the ulcerative colitis,as evidenced by reduc.i.n in body weight,improvement in crypt morphology,increase in antioxidant defenses,down-regulation of proinflammatory proteins and cytokines,and inhibition of proteins related to *** may represent a promising alternative therapeutic against ulcerative colitis,without inducing adverse effects.

Probing temperature-dependent interlayer coupling in a MoS2/h-BN heterostructure

Stac.i.g of atomically thin layers of two-dimensional materials has revealed extraordinary physical phenomena owing to van der Waals(vdW)interac.i.n at the ***,most of the studies focused on the transition metal dichalcogenide(TMD)/TMD heterostructure,while the interlayer coupling of the TMD/hexagonal boron nitride(h-BN)heterostructure has not been extensively explored despite its *** this study,the temperature-dependent interlayer coupling is demonstrated in a heterostructure of molybdenum disulfide(MoS2)and *** interface between MoS2 and the insulating substrate exerts a significant spectroscopic.i.pact on MoS2 through substrate-induced local strain,charged impurity,and vdW *** non-resonant conditions,temperature-dependent peak shifts in Raman and photoluminescence(PL)spectra of MoS2 reveal the evolution of interlayer *** frequencies and PL peak energies at different temperatures demonstrate how substrate-induced strain,impurity,and vdW interac.i.ns at the interface influence phonon vibration and excitonic transition of *** resonant conditions at low temperature,anomalous Raman modes appear in the MoS2/h-BN heterostructure because of the enhanced electron-phonon coupling and vdW *** anomalous Raman modes are quantitatively investigated by the deconvolution of the resonance Raman spectra and desc.i.ed by interlayer coupling at low temperature,in agreement with complementary indications from the temperature-dependent evolution of non-resonant Raman and PL spectra.

c.i.ical analysis of risk factors contributing to recurrence of pterygium after excision and graft surgery

AiM: To find the risk factors related to the reproliferation of the pterygial tissue after excision and graft ***: charts of 130 eyes of 130 patients who had pterygial excision from March 2006 to April 2011 were reviewed. Preoperative pterygium morphology, surgical methods, and adjunc.i.e treatments were statistically analyzed for their relationship with ***: During the follow-up period, recurrence was observed in 20 eyes(15.4%). None of the preoperative morphologic features were affected the rate of the recurrence. However, an age io(OR) 3.609, 95% confidence interval(ci) 0.838-15.540]and amniotic membrane graft instead of conjunc.i.al autograft(P =0.002, OR 9.093, 95% ci 2.316-35.698) were statistically significant risk factors for *** analysis revealed that intraoperative mitomycin c(MMc)(P =0.072, OR 0.298, 95% ci 0.080-1.115)decreased the rate of recurrence. cONcLUSiON: Younger age is a risk factor for reproliferation of pterygial tissue after excision and amniotic membrane transplantation(AMT) are less effec.i.e in preventing recurrence of pterygium after excision based on the comparison between conjunc.i.al autograft and AMT. intraoperative MMc application and conjunc.i.al autograft reduce recurrence.

Anti-scarring effects of butaprost on human subconjunc.i.al Tenon's fibroblasts

AiM： To investigate the toxicity of the E-prostanoid 2（EP2） receptor agonist, butaprost against human subconjunc.i.al（Tenon＇s capsule） fibroblasts, and to determine the underlying mechanism. METHODS： We isolated Tenon＇s fibroblasts from the subconjunc.i.al area of healthy subjects and evaluated the types of EP receptors expressed using quantitative realtime reverse transc.i.tion polymerase chain reac.i.n（RTPcR）. The toxicity of butaprost against the fibroblasts was evaluated using methyl thiazolyl tetrazolium and lac.i. dehydrogenase assays. The inhibition of conjunc.i.al fibroblast proliferation by butaprost was assessed by measuring α-ac.i. levels. The underlying mechanism was assessed by measuring intracellular cyc.i. adenosine monophosphate（c AMP） levels. intergroup differences were statistically analyzed using an independent t-test. Densitometry of the Western blot bands was performed using the image J software. RESULTS： Quantitative real-time RT-PcR revealed that the fibroblast EP2 receptor levels were higher than those of the other EP receptors. Butaprost did not show toxicity against Tenon＇s tissue, but inhibited conjunc.i.al fibroblast proliferation by reducing collagen synthesis. EP2 receptor ac.i.ation enhanced the c AMP cascade, which might be an important mechanism underlying this ***： Butaprost effec.i.ely reduces the subconjunc.i.al scarring response. Given the significanceof wound healing modulation in blebs, butaprost＇s inhibitory effect on subconjunc.i.al Tenon＇s fibroblasts may be beneficial in managing postoperative scarring in glaucoma surgery.

A movie trailer is a common advertising tool in the entertainment industry. Detec.i.n of a viewer’s brain responses to a movie trailer can help film producers to tailor a more appealing trailer of a movie. in this study, we acquired electroencephalographic (EEG) signals from subjects when they watched movie trailers (labeled as Movie session), and compared with their resting state session (labeled as Resting session) or when they watch nature scenes (labeled as Nature session). We used Sample Entropy (SampEn) to analyze the EEG signals between different sessions. Results showed that the complexity ratios at Fp1, Fp2 and Fz channels derived from Movie session were significantly lower than that in Resting state or when subjects watched Nature session (p in status can affect the complexity of their EEG. Further, the attrac.i.n of attention of a movie trailer can be observed from the change of EEG.

The antitumor capabilities of agonistic anti-4-1BB mAbs have made them an attrac.i.e target for tumor ***,the adverse side effects associated with agonist antibodies have hindered their c.i.ical ***,we aimed to study the immune-related adverse events of repeated doses and long-term use of agonistic anti-4-1BB *** show that chronic ac.i.ation of 4-1BB signals induced the accumulation of iFN-γ-producing PD-1^(+)cD8^(+)T cells in the secondary lymphoid organs of tumor-bearing mice by increasing the number of dividing cD8^(+)T cells,which was beneficial for suppressing tumor growth in the early phase of anti-4-1BB ***,repeated exposure to anti-4-1BB mAbs led to granuloma development in tumor-draining lymph nodes(TDLNs)of mice due to recruitment and accumulation of macrophages via the cD8^(+)T cell-iFN-γ*** was accompanied by excessive lymph node swelling,which impaired the sequential ac.i.ation of cD8^(+)T *** data provide insights into the immune-related adverse events of long-term agonist 4-1BB antibody dosing,which should be considered during the c.i.ical development of immunomodulating therapy.
