Polio eradication surveillance in Sri Lanka,2019-2023

Objective:To evaluate the polio laboratory surveillance carried out from January,2019 to May,2023 by the Polio Regional Reference Laboratory,Sri ***:This retrospective study analyzed all stool samples received under the acute flaccid paralysis(AFP)and immunodeficient vaccine-derived poliovirus(VDPV)surveillance at Polio Regional Reference Laboratory,Sri Lanka from January,2019 to May,*** results of the testing methodologies were extracted from the laboratory data system,i.e.,poliovirus virus isolation,intra-typic differentiation/VDPV real time reverse transcriptase polymerase chain reaction(ITD/VDPV rRTPcR)and sequencing,along with the data on timing of reporting results,stool adequacy and *** was analyzed using descriptive ***:A total of 2141 stool samples from 1644 cases were received for AFP surveillance from Sri Lanka(93.61%),Maldives(1.52%),and immunodeficient VDPV(4.86%)*** polioviruses(19/1644,1.15%)and non-polio enteroviruses(73/1644,4.44%)were isolated,while Sabin-like 3 virus was detected in majority(12/19,63.15%)among the poliovirus *** polioviruses or circulating VDPVs were not detected among the *** all years of the study,the non-polio AFP detection rate was>1/100000 in children aged less than 15 years,whereas stool adequacy rate was>80%.All results were reported within 14 days of receipt,ensuring timely reporting as per global ***:The Polio Regional Reference Laboratory,Sri Lanka plays a vital role in maintaining the polio-free status in the c.untry through its robust laboratory surveillance,while adhering to the surveillance ***-detection of wild polioviruses and circulating VDPV during the study period reinforces the polio-free status in the c.untry.

Advancing public health preparedness:Establishment of Nipah virus molecular diagnostic test at the National Reference Laboratory,Sri Lanka

Background:No studies have documented long-term trends in aboveground biomass(AGB)for mixed-dipterocarp forests(MDF),the dominant rain forest type in tropical wet equatorial *** our study,we sought to document such trends over forty years across three sites representing lowland to lower montane ***:To do this,we established fifty 100 m×25 m plots in 1978 across three sites sampled along an elevation gradient,identified as mature old-growth *** measured trees for diameter at breast height that we identified to species and *** took wood samples to calculate species wood-specific *** re-measured plots in 1998 and again in ***:We show standing AGB for all sites combined to be 517.52 Mg·ha^(-1)in 1978,but this declined by 17%over 40 years to 430.11 Mg·ha^(-1).No differences exist among sites in AGB primarily bec.use of considerable within site variation;but interactions of time with site show declines across sites were not uniform,one remained about the *** few species represented a high proportion of the AGB with the top five species comprising between 34%and 65%,depending upon site and year *** species,Mesua nagassarium,represented a disproportionately large amount of AGB and decline over time,particularly at the low elevation ***:Our results are directly relevant to estimating AGB and standing carbon sequestered in *** study is the first to demonstrate varying but overall,declining trends in amounts of AGB among forests making predictions of biomass and standing carbon in MDF difficult over wide regions.

Effects of interlayer spacing and applied pressure on the lanthanide transport in MoS2-based two-dimensional channels

Rare-earth elements(REEs)are critical to modern industry but difficult to separate due to their subtle and monotonic changes in physico-chemical ***2-based two-dimensional(2D)materials offer novel opportunities for enhancing REE separation,exhibiting a distinct volcano-shaped transport performance distribution that peaks at Sm3+.However,the specific contributions of thermodynamic and kinetic factors to ion transport within 2D confinement remain *** this study,we conducted a series of non-equilibrium all-atom molecular dynamics(MD)simulations to explore the effects of interlayer spacing and external pressure on the transport of lanthanide ions inÅ-scale acetate functionalized 2D MoS2(MoS2-cOOH)*** examined ion entry and permeation rates,water flux,dehydration,and binding *** simulation results reveal that the transport trends of lanthanide ions are jointly driven by the dehydration degree and the relative-binding strengths of ions to water and to the acetate within the 2D ***,the dehydration pattern of lanthanide ions during permeation is closely linked to kinetic ***,this study provides a detailed atomistic.understanding of the mechanisms under-lying lanthanide ion transport under *** findings point to the significant potential for tuning confinement and chemical functionalization withinÅ-scale channels for more efficient REE separation.

Hierarchical control Strategy for Load Regulation Based on Stackelberg Game Theory considering Randomness

Demand response has been recognized as a valuable functionality of power systems for mitigating power *** paper proposes a hierarchical control strategy among the distribution system operator(DSO),load aggregators(LAs),and thermostatically controlled loads(TcLs);the strategy inc.udes a scheduling layer and an executive layer to provide load *** the scheduling layer,the DSO(leader)offers compensation price(cP)strategies,and the LAs(followers)respond to cP strategies with available regulation power(ARP)*** of the DSO and LAs are modeled according to their behaviors during the load regulation *** game is adopted to capture interactions among the players and leader and to obtain the optimal strategy for each participant to achieve ***,considering inevitable random factors in practice,e.g.,renewable generation and behavior of users,two different stochastic models based on sample average approximation(SAA)and parameter modification are formulated with improved scheduling *** the executive layer,distributed TcLs are triggered based on strategies determined in the scheduling layer.A self-triggering method that does not violate user privacy is presented,where TcLs receive external signals from the LA and independently determine whether to alter their operation *** simulations are performed on the modified IEEE-24 bus system to verify effectiveness of the proposed strategy.

As a precise and versatile tool for genome manipulation,the c.ustered regularly interspaced short palindromic repeats(cRISPR)/cRISPR-associated protein 9(cas9)platform holds promise for modifying fish traits of *** the aim of reducing transgene introgression and controlling reproduction,upscaled disease resistance and reproductive intervention in catfish species have been studied to lower the potential environmental risks of the introgression of escapees as transgenic *** advantage of the cRISPR/cas9-mediated system,we succeeded in integrating the cathelicidin gene(As-cath)from an alligator(Alligator sinensis)into the target luteinizing hormone(lh)locus of channel catfish(Ictalurus punctatus)using two delivery systems assisted by double-stranded DNA(dsDNA)and single-stranded oligodeoxynucleotides(ssODNs),*** this study,high knock in(KI)efficiency(22.38%,64/286)but low ontarget events was achieved using the ssODN strategy,whereas adopting a dsDNA as the donor template led to an efficient on-target KI(10.80%,23/213).The on-target KI of As-cath was instrumental in establishing the lh knockout(LH^(–)_As-cath^(+))catfish line,which displayed heightened disease resistance and reduced fecundity compared with the wild-type(WT)sibling ***,administration of human chorionic gonadotropin(HcG)and luteinizing hormone-releasing hormone analogue(LHRHa)can restore the reproduction of the transgenic fish ***,we replaced the lh gene with an alligator cathelicidin transgene and then administered hormone therapy to move towards complete reproductive control of diseaseresistant transgenic catfish in an environmentally responsible *** strategy not only effectively improves consumer-valued traits but also guards against unwanted introgression,providing a breakthrough in aquaculture genetics to confine fish reproduction and prevent the establishment of transgenic or domestic genotypes in the natural environment.

Investigation of HcAR2 antagonists as a potential strategy to modulate bovine leukocytes

Background Dairy cows experiencing ketosis after calving suffer greater disease incidence and are at greater risk of leaving the herd. In vitro administration of beta-hydroxybutyric acid(BHBA;the primary blood ketone) has inhibitory effects on the function of bovine leukocytes. BHBA is a ligand of HcAR2 and the activation of these receptors promotes an anti-inflammatory response which may be related with immunosuppression observed in transition dairy cattle. The objective of this study was to identify and test antagonists for HcAR2 in bovine immune cells cultured with *** We observed expression of HcAR2 at the protein level within lymphocytes, monocytes, and granulocytes. The proportion of cells expressing HcAR2 tended to be greater in mid-lactation compared to early lactation cows;the increase was a result of increased proportion of T and B cells expressing HcAR2. Stimulation of HcAR2 with niacin or BHBA promoted ca^(2+) mobilization in neutrophils and mononuclear cells. Mononuclear cells treated with BHBA had diminished intracellular ca^(2+) responses when HcAR2 was knocked down by si RNA silencing, indicating ca^(2+) mobilization was mediated by HcAR2 signaling. Two candidate antagonists for HcAR2, synthesized from niacin(NA-1 and NA-5), were tested;monocytes and neutrophils pre-treated with NA-1 and NA-5 had reduced ca^(2+) mobilization after incubation with BHBA. Furthermore, NA-5 but not NA-1 prevented BHBA-associated reductions in cyclic *** We demonstrated that HcAR2 is present on bovine leukocytes and has greater expression later in lactation. We confirmed that BHBA and niacin derived HcAR2 antagonists alter bovine leukocyte activity. Our results demonstrate that both BHBA and niacin affect bovine leukocyte ca^(2+) mobilization in a HcAR2-dependent manner.

Spatio-temporal variability in microfossil and geochemical records of cenomanian-Turonian oceanic anoxic event-2: a review

The cenomanian-Turonian boundary oceanic anoxic event(OAE-2; Bonarelli event; ～94 Ma) is associated with a species turnover in the marine calcareous microfossil record, widespread marine anoxia,black shale deposition and positive δ13c excursions. This study reviews 141 cTBE sites globally, aiming to understand micropaleontological, geochemical, and sedimentological expressions of OAE-2. There is a clear palaeogeographic and palaeobathymetric heterogeneity in the development of OAE-2 marine anoxia. A majority of the documented OAE-2 sites are from deep marine environments. The calcareous nannoplankton and benthic foraminifera record a diversity decline, while planktic foraminifera shows community level shifts and no major mass extinction. The variability of total organic carbon in OAE-2 sediments across sites(10 wt.%) and the δ13c profiles(diachronous) have been attributed to different mechanisms of anoxia *** increased primary “productivity model” gains support from productivity proxies(e.g., Ba, P, cu, Ni),redox-sensitive elements(e.g., Mn, Mo, u, V, As), and eutrophic genera(e.g., benthic foraminifera Gabonita spp., calcareous nannofossils Biscutum spp. and Zeugrhabdotus erec.us). The enhanced organic carbon“preservation model” in stratified(semi) restricted basins gains support from sites with lower enrichment of redox-sensitive elements and oligotrophic flora and fauna in the OAE-2 records. Geochronology of the caribbean and the High Atlantic Large Igneous Province events(cLIP and HALIP) mark them as likely triggers of the OAE-2 related global perturbation of marine biogeochemistry. Volcanic triggers may have c.used climate warming, altered hydrological cycles, enhanced continental weathering, shifts in ocean circulation, and nutrient flows. Addressing knowledge gaps, further research is urged, utilizing innovative proxies, and exploring underrepresented depositional systems to comprehensively understand OAE-2 onset and biotic crisis.
