This study investigates poly(vinyl alcohol) (PVA) membranes as controlled release micro-matrices, which can be useful in therapeutic applications for optimizing the administration of drugs. currently, the use of hydrogels is limited by protein size. This study investigates the delivery of PspA, a large protein of approximately 38 kD. Pneumococcal surface protein A (PspA) has been shown to provide protective immunity against pneumococcal infection and is considered as a pneumococcal vaccine. The protein release experiments demonstrated that from an initial pH 7.4, approximately 60% of PspA diffuse into a neutral environment with an initial burst and a declining rate reaching equilibrium. The results indicate that the protein was successfully incorporated and released from the membrane over time. The hydrogel and protein interaction is temporary, and the membrane system is ideal for protein drug delivery. The data confirm that the protein did not aggregate and was active after release. The protein release is promising and a step forward to develop microneedles to facilitate high molecular weight protein delivery as well as vaccine delivery.

Genome-wide association studies(GWASs) have been instrumental in understanding complex phenotypic traits. However, they have rarely been used to understand lineage-specific pathways and functions that contribute to the trait. In this study, by integrating lineage-specific enhancers from mesenchymal and myeloid compartments with bone mineral density loci, we were able to segregate osteoblast-and osteoclast(Oc)-specific functions. Specifically, in Ocs, a PU.1-dependent transcription factor(TF)network was revealed. Deletion of PU.1 in Ocs in mice resulted in severe osteopetrosis. Functional genomic analysis indicated PU.1 and MITF orchestrated a TF network essential for Oc differentiation. Several of these TFs were regulated by cooperative binding of PU.1 with BRD4 to form superenhancers. Further, PU.1 is essential for conformational changes in the superenhancer region of Nfatc1. In summary, our study demonstrates that combining GWASs with genome-wide binding studies and model organisms could decipher lineage-specific pathways contributing to complex disease states.

Interactive Effects of Elevated cO_2 and Growth Temperature on the Tolerance of Photosynthesis to Acute Heat Stress in c_3 and c_4 Species

Determining effects of elevated cO2 on the tolerance of photosynthesis to acute heat-stress （heat wave） is necessary for predicting plant responses to global warming, as photosynthesis is thermolabile and acute heat-stress and atmospheric cO2 will increase in the future. Few studies have examined this, and past results are variable, which may be due to methodological variation. To address this, we grew two c3 and two c4 species at current or elevated cO2 and three different growth temperatures （GT）. We assessed photosynthetic thermotolerance in both unacclimated （basal tolerance） and preheat-stressed （preHS = acclimated） plants. In c3 species, basal thermotolerance of net photosynthesis （Pn） was increased in high cO2, but in c4 species, Pn thermotlerance was decreased by high cO2 （except Zea mays at low GT）; cO2 effects in preHS plants were mostly small or absent, though high cO2 was detrimental in one c3 and one c4 species at warmer GT. Though high cO2 generally decreased stomatal conductance, decreases in P, during heat stress were mostly due to non-stomatal effects. Photosystem Ⅱ （PSⅡ） efficiency was often decreased by high cO2 during heat stress, especially at high GT; cO2 effects on post-PSⅡ electron transport were variable. Thus, high cO2 often affected photosynthetic theromotolerance, and the effects varied with photosynthetic pathway, growth temperature, and acclimation state. Most importantly, in heat-stressed plants at normal or warmer growth temperatures, high cO2 may often decrease, or not benefit as expected, tolerance of photosynthesis to acute heat stress. Therefore, interactive effects of elevated cO2 and warmer growth temperatures on acute heat tolerance may contribute to future changes in plant productivity, distribution, and diversity.

The urban resilience concept was introduced in 2016 as a key concept in the Habitat iii New Urban Agenda for the next 20 *** wonder how this urban resilience concept was elaborated and who influenced it the most?The preparatory events structured several stakeholders’*** relations between stakeholders allowed the flow of ideas in the consultation and production *** influential stakeholders strongly oriented definition of urban resilience concepts by taking power in the networking process of the consecutive *** paper analyzes the network of stakeholders/concepts,during the building process between 2012 and 2016(5,539 discourses from 290 stakeholders,in 357 events).The application of textual mining and machine learning topic modelling algorithm exposed the structure of the principal topics for building the concept of urban resilience,and presented how relations of main stakeholders with funders was crucial for the investment in policy ***,we underlined for the first time in an empirical way,different kinds of actors’power in the construction process that supported the Habitat iii resilience *** demonstrated how far some official stakeholders,but also external and private ones,oriented the construction of ideologies to validate the knowledge that supported the related actions in laboratory cities.

A Possible Multi-stranded DNA Induced by Acid Denaturation-renaturation

lambda-DNA Hind iii was denatured by Hcl, renatured by NaOH, and digested by DNase I sequentially. The produced mixture was separated with Sephadex G200 column, and the chromatograph showed a very weak peak of one component resistant to the digestion of DNase I. The results of AFM studies on this component indicated that an unusual form of DNA was present, with the height 13 times that of double-stranded DNA. By comparison with known results, it is suggested that this is a multi-stranded DNA other than simple supercoils.

Introduction: Hemorrhoidal disease is a common pathology and its surgical treatment is based, among other things, on pedicular resection after failure of medical and instrumental treatment. The aim of this study is to analyze the epidemiological, clinical, therapeutic and evolving aspects of hemorrhoidal disease at the stage of surgical treatment at the University Hospital center of Brazzaville. Patients and Methods: We conducted a retrospective and descriptive study carried out from January 2020 to December 2021, a 24 months period, in the Digestive surgery department of the University Hospital center of Brazzaville. It concerned patients who underwent a surgical procedure for hemorrhoidal disease. Results: 21 cases were collected, representing a hospital frequency of 2.3%, with a sex ratio of 4.3 in favor of men. The average age of patients was 42.2 ± 11.9 years. The symptoms were mainly proctalgia, mass sensation and rectal bleeding. We recorded five cases (19.1%) of hemorrhoidal thrombosis and 16 cases (80.9%) of hemorrhoidal prolapse including 12 cases requiring manual integration (Goligher grade iii) and four irreducible permanent cases (Goligher grade IV). The Grade iii prolapse was associated with a polyp in one patient and with posterior anal fissure in another patient. Out of the 21 patients, 14 underwent a tripedicular hemorrhoidectomy according to Milligan and Morgan. Two patients underwent mono- and bipedicular hemorrhoidectomy with resection of associated lesions and five patients underwent emergency thrombectomy. The outcome was favorable for all our patients. The average length of hospital stay was 1.5 ± 2.1 days. conclusion: In the event of failure of medical and instrumental treatment, the hemorrhoidal cure according to Milligan and Morgan is the surgical treatment of reference for hemorrhoidal disease at the University Hospital center of Brazzaville.

Herein we describe an approach to measure the volume and to characterize the volume distribution of large numbers of individual small regularly shaped seeds. The results of a preliminary investigation into the distribution of seed size of a multi-seeded sorghum mutant, as compared to the wild type from which it was developed are also reported, and are used as an example of the method’s utility.
