Novel UBE2B-assoc.ated polymorphisms in an azoospermic./ oligozoospermic.population

Aim： To assess whether abnormalities exist in the UBE2B gene in a population of infertile human males, and to establish biologic.plausibility of any disc.vered mutations. Methods： We c.rried out polymerase c.ain reac.ion （Pc.） amplific.tion and sequenc. analysis of the 5＇-untranslated region and six exons of the UBE2B gene, inc.uding flanking intronic.regions, in a group of fertile and infertile men. Following the identific.tion of a putative promoter region that c.ntained single or dual triplet deletions within a 10-c.G repeat island, we evaluated the binding affinity of these identified polymorphisms as c.mpared to the wild-type sequenc. to transc.iption fac.or SP1 using a DNA-protein gel shift assay. Results： There was a novel exonic.single nuc.eotide polymorphism （SNP） noted in exon 4 in 5% of infertile men. In silic. 3D modeling of the altered protein showed an innoc.ous isoleuc.ne for valine substitution. There were no mutations noted within any of the other exons. Three novel intronic.SNPs were identified within the fertile group, and seven novel intronic.SNPs identified in the infertile group. The DNA-protein gel shift assay noted that both single c.G deletion and double c.G deletion bands had approximately twic. the binding affinity c.mpared to the wild-type for SP1. The negative c.ntrol c.nfirmed no non-spec.fic.protein binding. c.nc.usion- By themselves, a single or double c.G deletion is unlikely to pose biologic.signific.nc.. However, suc. deletions in this suspec.ed promoter region are assoc.ated with inc.eased binding affinity for SP1, and might represent one of several fac.ors required for alteration of UBE2B gene expression.

BAc.GROUND There is c.nc.rn regarding potential long-term c.rdiotoxic.ty with systemic.distribution of metals in total joint arthroplasty(TJA)*** To determine the assoc.ation of c.mmonly used implant metals with ec.oc.rdiographic.measures in TJA *** The study c.mprised 110 TJA patients who had a rec.nt history of high c.romium,c.balt or titanium *** underwent two-dimensional,three-dimensional,Doppler and spec.le-strain transthorac.c.ec.oc.rdiography and a blood draw to measure metal *** and sex-adjusted linear and logistic.regression models were used to examine the assoc.ation of metal c.nc.ntrations(exposure)with ec.oc.rdiographic.measures(outc.me).RESULTS Higher c.balt c.nc.ntrations were assoc.ated with inc.eased left ventric.lar end-diastolic.volume(estimate 5.09;95%c.:0.02-10.17)as well as left atrial and right ventric.lar dilation,partic.larly in men but no c.anges in c.rdiac.*** titanium c.nc.ntrations were assoc.ated with a reduc.ion in left ventric.e global longitudinal strain(estimate 0.38;95%c.:0.70 to 0.06)and c.rdiac.index(estimate 0.08;95%c.,-0.15 to-0.01).c.Nc.USION Elevated c.balt and titanium c.nc.ntrations may be assoc.ated with struc.ural and func.ional c.rdiac.c.anges in some *** studies are warranted to better understand the systemic.effec.s of metals in TJA patients.

Obesity has a negative effec. on male reproduc.ive func.ion. It is assoc.ated with low testosterone levels and alteration in gonadotropin sec.etion. Male obesity has been linked to reduc.d male fertility. Data regarding the relation of obesity to sperm parameters are c.nflic.ing in terms of the nature and magnitude of the effec.. New areas of interest are emerging that c.n help explain the variation in study results, suc. as genetic.polymorphism and sleep apnea. Sleep disorders have been linked to altered testosterone produc.ion and hypogonadism in men. It was also c.rrelated to erec.ile dysfunc.ion. The relation of sleep disorders to male fertility and sperm parameters remains to be investigated. Men with hypogonadism and infertility should be sc.eened for sleep apnea. Treatment of obesity and sleep apnea improves testosterone levels and erec.ile func.ion.

Vitamin D levels have been linked to various health outc.mes inc.uding reproduc.ive disorders. The purpose of this study was to explore the assoc.ation between serum vitamin D level （25-hydroxy-vitamin D, or 250HD） and semen and hormonal parameters. This is a c.oss-sec.ional study that inc.uded 170 healthy men rec.uited for the study of spermatogenesis from the general population. Men c.mpleted general and reproduc.ive health questionnaires, and donated blood and semen samples. The main measures were hormonal （total and free testosterone, sex hormone-binding globulin, estradiol, follic.e-stimulating hormone and luteinizing hormone） and semen parameters, adjusted （n= 147） for age, body mass index （BMI）, season, alc.hol intake and smoking, in relation to c.tegories of vitamin D levels, determined apriori. The mean age of the study population was 29.0±8.5 years and mean BMI was 24.3±3.2 kg m-2. The mean 250HD was 34.1± 15.06 ng m1-1. BMI showed a negative assoc.ation with 250HD. Sperm c.nc.ntration, sperm progressive motility, sperm morphology, and total progressively motile sperm c.unt were lower in men with ‘250HD ≥ 50 ng ml-1＇ when c.mpared to men with‘20 ng ml-1 ≤ 250H D〈 50 ng ml-1,. Total sperm c.unt and total progressive motile sperm c.unt were lower in men with ‘250HD〈20 ng ml-1＇ when c.mpared to men with‘20 ng ml-1≤250HD〈50 ng ml-1＇. The adjusted means of various hormonal parameters did not show statistic.l differenc. in the different c.tegories of 250HD. In c.nc.usion, serum vitamin D levels at high and low levels c.n be negatively assoc.ated with semen parameters.

Rheumatic.fever is an auto-immune disease c.used by exposure to Streptoc.c.us pyogenes. Over the last 50 years, reports of rheumatic.fever within the United States have diminished. The dec.ease was attributed to the advent of penic.llin in the treatment of streptoc.c.us infec.ions. We propose that c.rrent diagnostic.and treatment methodologies may adversely inc.ease the morbidity rate of rheumatic.fever within the United States. Public.tion rates and interest in rheumatic.fever has diminished over the last 30 years. Bec.use of this dec.ine, many physic.ans are only vaguely aware of the disorder. Additionally, the fear of antibiotic.resistanc. has influenc.d thec.nterofDisease c.ntrol to suggest a signific.nt dec.ease in the use of antibiotic. by physic.ans. Although extremely valid for the future health and well-being of the population, suc. polic.es must be examined for eac. individual c.se c.refully. The Americ.n Heart Assoc.ation presc.ibes long-term antibiotic.prophylaxis as the only c.rrent treatment;however literature reviews indic.te that suc. therapy is rarely used. Therefore individuals diagnosed with rheumatic.fever are not being treated. Additionally, bec.use many physic.ans are not routinely testing for streptoc.c.us or early signs of endoc.rditis, it is likely that c.ses of rheumatic.fever will inc.ease in the future, and many individuals may not be diagnosed until sever damage or morbidity oc.urs. Physic.an educ.tion and c.ear revised guidelines are nec.ssary to ensure adequate treatment of individuals with rheumatic.fever. Mis-understandings of the disease and how it should be treated by first responders (i.e. primary c.re providers and pediatric.ans) are disc.ssed.

Detec.ing and Adjusting Temporal Inhomogeneity in c.inese Mean Surfac. Air Temperature Data

Adopting the Easterling-peterson (EP) tec.niques and c.nsidering the reality of c.inese meteorologic.l observations, this paper designed several tests and tested for inhomogeneities in all c.inese historic.l surfac. air temperature series from 1951 to 2001. The result shows that the time series have been widely impac.ed by inhomogeneities resulting from the reloc.tion of stations and c.anges in loc.l environment suc. as urbanization or some other fac.ors. Among these fac.ors, station reloc.tions c.used the largest magnitude of abrupt c.anges in the time series, and other fac.ors also resulted in inhomogeneities to some extent. Ac.ording to the amplitude of c.ange of the differenc. series and the monthly distribution features of surfac. air temperatures, disc.ntinuities identified by applying both the E-P tec.nique and supported by c.ina's station history rec.rds, or by c.mparison with other approac.es, have been adjusted. Based on the above proc.ssing, the most signific.nt temporal inhomogeneities were eliminated, and c.ina's most homogeneous surfac. air temperature series has thus been c.eated. Results show that the inhomogeneity testing c.ptured well the most important c.ange of the stations, and the adjusted dataset is more reliable than ever. This suggests that the adjusted temperature dataset has great value of dec.easing the unc.rtaities in the study of observed c.imate c.ange in c.ina.

Group Orbits of GPS Satellite c.nfigurations for c.nstellation Management

Air Forc. Spac. c.mmand is interested in improving the ac.urac. of GPS rec.iver positioning, navigation, and timing. To this end, it is useful to identify a set of optimal satellite c.nstellations where eac. c.rresponds to a c.nfiguration spec.fying the number of satellites in eac. orbital plane. These c.nstellations c.uld then be maintained in a library for future use as satellites fail and are launc.ed. We utilize symmetry in the geometry of the GPS satellite orbits to partition the c.nfigurations into a muc. smaller set of equivalenc. c.asses where eac. c.ass has the same overall rec.iver ac.urac. performanc.. We apply a c.assic.l algebraic.c.mbinatorial result, Polya＇s Theorem, to c.unt and c.tegorize the c.asses. Inc.rporating our results into a GPS c.nstellation optimization c.mputer tool will reduc. run time by about an order of magnitude. We apply other algebraic.and c.mbinatorial tec.niques in original ways to c.unt the c.ass sizes and the c.asses that c.ntain a given number of satellites. Finally, we break the equivalenc. c.asses into a still smaller set of new ＂struc.ure＂ c.asses that are useful in applying the GPS c.mputer tool.

Mining diversity of the natural biorefinery housed within Tipula abdominalis larvae for use in an industrial biorefinery for produc.ion of lignoc.llulosic.ethanol

Although they are the largest taxonomic.group of animals, relatively few insec.s have been examined for symbiotic.relationships with mic.o-organisms. However, this is rapidly c.anging bec.use of the potential for examination of the natural insec.mic.obe-lignoc.llulose interac.ions to provide insights for biofuel tec.nology. Mic.o-organisms assoc.ated with lignoc.llulose-c.nsuming insec.s often fac.litate the digestion of the rec.lc.trant plant diet; therefore these mic.obial c.mmunities may be mined for novel lignoc.llulose-degrading mic.obes, or for robust and inexpensive bioc.talysts nec.ssary for ec.nomic.lly feasible biofuel produc.ion from lignoc.llulose. These insec.-mic.obe interac.ions are influenc.d by the ec.system and spec.fic.lignoc.llulose diet, and appre- c.ating the whole ec.system-insec.-mic.obiota-lignoc.llulose as a natural biorefinery provides a ric. and diverse framework from whic. to design novel industrial proc.sses. One suc. natural biorefinery, the Tipula abdominalis larvae in riparian ec.systems, is reviewed herein with applic.tions for bioc.emic.l proc.sses and overc.ming c.allenges involved in c.nversion of lignoc.llulosic.biomass to fuel ethanol. From the dense and diverse T. abdominalis larval hindgut mic.obial c.mmunity, a c.llulolytic.bac.erial isolate, 27c.4, demonstrated enzymatic.ac.ivity toward many model plant polymers and also produc.d a bac.erial antibiotic. 27c.4 was c.-c.ltured with yeast in fermentation of pine to ethanol, whic. allowed for a 20% reduc.ion of c.mmerc.al enzyme. In this study, a mic.o-organism from a lignoc.llulose-c.nsuming insec. was suc.essfully applied for improvement of biomass-to-biofuel tec.nology.
