Gastric tissue samples were studied using mid-IR fiber-optic attenuated total reflectance (ATR) spectroscopy. FTIR spectra of 90 tissue samples from 48 patients, including 32 normal and 58 malignant tissue samples, were chosen as examples. Malignancy was usually characterized by the absence of cH and c=O bands, a weak amide II band near 1545 cm-1, a shift of the amide I band to lower wave number, a decrease in the ～1450 cm-1 peak to less than the～1400 cm-1 peak. Subtraction spectra indicate that the amide I and amide II bands of normal and malignant tissues have larger differences in peak positions and relative intensities. The statistical analysis results confirm this conclusion. The results indicate that FTIR fiber optic techniques provide important information about cancerous tissue of the stomach, which can be used to differentiate the malignant tissue from the normal tissue. Based on the above results we successfully realize the detection of the tumor tissues of digestive tract in vivo and in situ. And the results of detection cancer near operating room and in vivo and in situ in the operating room are consistent with the conclusions for the samples stored in liquid N2, which is the basis for the clinical application.

Purpose:To investigate the express ion of c-kit in human salivary gland adenoid cystic carcinoma and their relationships with clinicopathological parameters and prognostic value and then to determine mutational status of exons 9, 11, 13 and 17 o f c-k it gene which provide theoretical foundation for predicting prognosis and targeted therapy. Methods:The expression of c-kit protein was detected measured in 54 human salivary gland adenoid cystic carcinoma by immunohistochemistry and its relationship to prognosis was analyzed statistically. The mutation of c-kit in exons 9, 11, 13 and 17 was determined by SScP. Results:The expression rate of c-kit protein in human salivary gland adenoid cystic carcinoma was 85.2% （ 46/54）. The human salivary gland adenoid cystic carcinoma with higher expression rate of c-kit was prone to recur locally. The survival time after operation of the patients with c-kit higher expression were significantly lower than that of the patients with c-kit lower expression. The mutation in exon 9, 11, 13 and exon 17 were founded in 19, 8, 7, 9 of 54 cases with human salivary gland adenoid cystic carcinoma and DNA sequencing also detect the mutation. conclusions:The expression of c-kit could serve as a prognostic factor in human salivary gland adenoid cystic carcinoma. The mutation in exon 9, 11, 13 and exon 17 was found in human salivary gland adenoid cystic carcinoma. Therefore, these exons may function as targets for treatment modalities based on the suppression of c-kit tyrosine kinase activity.

Expression and chromosomal Mapping of Mouse Gpx2 Gene Encoding the Gastrointestinal Form of Glutathione Peroxidase, GPX-GI

GPX-GI is a cytosolic tetrameric Se-dependent glutathione peroxidase, similar in properties to GPX-1. Unlike the almost ubiquitous GPX-1, GPX-GI is mainly expressed in the epithelium of gastrointestinal tract. GPX-GI contributes to at least fifty percent of GPX activity in rodent small intestmal epithelium. The total GPX activity consists of at least 70% of selenium-dependent GPX activity in this *** analyzing a panel of mouse mterspecies DNA from the Jackson Laboratory's backcross resource,we mapped Gpx2 gene to mouse chromosome 12 between D12Mit4 and D12Mit5, near the ccs1 locus which contains a colon cancer susceptibility gene. A pseudogene, Gpx2-ps is mapped to mouse chromosome *** of Gpx2 gene expression in three pairs of c57BL/6Ha and IcR/Ha mice which are respectively resistant and sensitive to dimethylhydrazine-induced colon cancer, we found a higher Gpx2 mRNA level in c57BL/6Ha colon than IcR/Ha colon. Interestingly, a lower level of GPX activity is found in the resistant strain of mice. Because GPX-1 has three times higher specific activity than GPX GI, our data suggest that the decreased GPX activity may result from a higher level of Gpx2 gene expression in those cells co-express GPx1 gene

Recent studies have revealed that the γ-chain of theIL-2 receptor is shared by the receptors for IL-4, IL7, IL-9, IL-13, and IL-15, and it is therefore also referred toas the common γ-chain (γc). Mutations of γc result inX-linked severe combined immunodeficiency syndrome inhumans, indicating that rye is essential for normal development and function of the immune system. We demonstratethat human hematopoietic cells express two γc transcriptsdiffering in their carboxyl terminal coding region. Onetranscript is the previously reported sequence (γc-long),whereas the newly identified sequence exhibits a deletion of72 nucleotides close to the 3'-end of the open reading frame(γc-short). This alteration predicts a loss of 24 amino acidsincluding a conserved tyrosine residue which is shared byseveral members of the cytokine receptor family. Thepresence of these two distinct forms of rye transcripts wasdemonstrated by sequencing of reversely transcribed andpolymerase chain reaction (RT-PcR) amplified mRNA, restriction digestion of the RT-PcR products, RNAse protection, and Northern blotting from human cell lines andhuman peripheral blood lymphocytes. Furthermore, thetwo variants were present in peripheral blood lymphocytesfrom both female and male donors, which rules out allelicvariants since rye is a single copy gene located on the Xchromosome. A truncation mutant at a site near the observed changes in γc-short has been reported by othersto alter biochemical events activated by cytokines. Thiscombined with the loss of a potential SH2 "docking" sitein γc-short suggests that γc-long and γc-short may link todifferent signaling pathways and may play an importantrole in determining the cellular response to IL-2, IL-4, IL-7, IL-9, IL-13, IL-15.

Objective: The aim of this study was to investigate the clinicopathologic and prognostic factors of the partial subtypes of lung adenocarcinoma, including pure bronchioloalveolar carcinoma (BAc), adenocarcinoma (Ac) with BAc component and Ac without BAc component. Methods: One hundred and six adenocarcinoma specimens which were followed up completely for 3 years, were obtained from 106 patients (45 men and 61 women) who underwent surgical resection for pathologically confirmed pulmonary adenocarcinoma in the cancer Hospital of Tianjin Medical University, from June 2004 to December 2005. According to the recent 2004 World Health Organization (WHO) pathological classification criteria of lung cancer, lung adenocarcinomas were divided into three subgroups: pure BAc, Ac with BAc component and Ac without BAc component. The clinical data were retrospectively analyzed based on statistical methods. All data were analyzed using SPSS statistics software and Kaplan-Meier survival curves were constructed, meanwhile, we conducted a Log-rank test. Results: The statistical analysis showed that no significant association was found among the three groups in gender and age; however, smoke index, tumor size, N stage, TNM stage, postoperative recurrence and metastasis had a statistically significant correlation among three groups (P ctively, which had a statistically significant difference. And the 3-year survival rate was significantly higher in the patients with pure BAc than in the patients with other types of lung adenocarcinomas (P contrast to the other two groups (pure BAc and Ac with BAc component), we found the evidence that the 3-year prognosis of lung adenocarcinoma without BAc component was worse than the two formers. conclusion: The three groups (pure BAc, Ac with BAc component and Ac without BAc component) have their own distinct clinicopathologic features respectively and c
