current updates on laboratory techniques for the diagnosis of male reproductive failure

The incidence of male reproductive failure leading to infertility, whether due to delayed parenthood, environmental issues, genetic factors, drugs, etc., is increasing throughout the world. The diagnosis and prognosis of male subfertility have become a challenge. While the basic semen assessment has been performed for many years, a number of studies question the value of the traditional semen characteristics. This is partly due to inadequate methods and standardization, limited knowledge of technical requirements for quality assurance, and an incomplete understanding of what clinical information a semen assessment can provide. Laboratories currently performing semen and endocrine assessment show great variability. The World Health Organization （WHO） manual for the evaluation of semen has been the core of andrology and fertility evaluation that has helped in further development of this field over many years. These include the physical appearance of the ejaculate, assessments of sperm count, motility, vitality, morphology, and functional aspects of the sperm and semen sample. These tests also include male endocrine profile, biochemical evaluation of the semen, detection of antisperm antibodies in serum, the use of computer-aided sperm analysis （cASA）, sperm DNA integrity, and its damage due to oxidative stress. Assisted reproductive techniques （e.g., IVF, IcSI） have shown great success but are too expensive. Further development in this field with newer techniques and extensive training/instructions can improve accuracy and reduce variability, thus maintaining the quality and standards of such an evaluation. There is an urgent need to have standardized training centers and increased awareness in this area of men＇s health for reproductive success.

Genes and genetics in eye diseases: a genomic medicine approach for investigating hereditary and inflammatory ocular disorders

Past 25 y have witnessed an exponential increase in knowledge and understanding of ocular diseases and their respective genetic underpinnings. As a result, scientists have mapped many genes and their variants that can influence vision and health of our eyes. Based on these findings, it is becoming clear that an early diagnosis employing genetic testing can help evaluate patients＇ conditions for instituting treatment plan（s） and follow-up care to avoid vision complications later. For example, knowing family history becomes crucial for inherited eye diseases as it can benefit members in family who may have similar eye diseases or predispositions. Therefore, gathering information from an elaborate examination along with complete assessment of past medical illness by ophthalmologists followed by consultation with geneticists can help create a roadmap for making diagnosis and treatment precise and beneficial. In this review, we present an update on ocular genomic medicine that we believe has tremendous potential towards unraveling genetic implications in ocular diseases and patients＇ susceptibilities. We also discuss translational aspects of genetic ophthalmology and genome engineering that may help advance molecular diagnostics and therapeutics.

Homocysteine mediates transcriptional changes of the inflammatory pathway signature genes in human retinal pigment epithelial cells

AIM：To test whether homocysteine（Hcy） can influence the transcriptional profile, we hypothesized that Hcy can lead to the induction of proinflammatory molecules in the retinal cells of aging ***：An unbiased in vitro inflammatory pathway focused study was designed employing retinal pigment epithelial（RPE） cell line, ARPE-19. cells were cultured in the presence or absence of Hcy to capture target genes＇ expression profile. Three different concentrations of Hcy were added in the culture medium of confluent monolayers. c RNAs were made from the isolated total RNAs and the labeled c RNA probes were hybridized to microarrays specific for human disease pathway inflammatory cytokines, chemokines and their receptor gene micro-array panels as per manufacture＇s recommendations. Two Hcy up-regulated molecules：IL6 and cEBPB were further validated via Western blot analysis. Hcy＇s effect on ARPE-19 cellular morphology and genomic DNA integrity were also ***：Gene microarray analyses of RPE cells in response to Hcy treatment revealed alterations in the expressions of several inflammatory gene transcripts such as ccL5, cEBPB, IL13RA2, IL15 RA, IL6, IL8 and cXcL3 that were up-regulated. The transcripts for c3, ccL2, IL11 RA and IL18 genes exhibited down-regulation. The IL6 and cEBPB expressions were subsequently validated at the protein levels. Treatment of the retinal cells with increasing Hcy concentration influenced their density in culture however their morphology and DNA integrity remained unaffected. cONcLUSION：These findings suggest that Hcy can potentially mediate the expression of chemokines,cytokines and interleukins receptors in the retinal cells without having any debilitating effects on their morphology and the genomic DNA integrity.

There are many vision threatening diseases of the eye affecting millions of people worldwide. In this article, we are summarizing potential role of various matrix metalloproteinases （MMPs）; the Zn （2＋）-dependent endoproteases in eye health along with pathogenesis of prominent ocular diseases such as macular degeneration, diabetic retinopathy, and glaucoma via understanding MMPs regulation in affected patients, interactions of MMPs with their substrate molecules, and key regulatory functions of tissue inhibitor of metalloproteinases （TIMPs） towards maintaining overall homeostasis.

Biomass Industrial Effluent Effect on carbohydrates,Aminoacids,Nitrite and Nitrite Enzyme Activities of Arachis hypogaea L.

The objectives of the present investigations are to see the effects of biomass power plant effluent on the carbohydrates, aminoacids, nitrite and nitrite enzyme activitiesand proline of Arachis hypogaea L. var TcGS 320 under controlled pot culture methods. Plants were cultivated with 25, 50, 75 and 100% of the effluent and a control without the effluent. The treatment of the crop with 25% of the effluent has shown stimulatory effect on all the biochemical parameters studied. carbohydrates, starch, aminoacids, protein, nitrate and nitrite reductase enzymatic activities have increased in 10, 15, 20 DAS （days after sowing）. In 25 and 30 DAS all biochemical parameters have decreased, due to environmental factors （rainfall, temperature, humidity, etc.）. The present investigation clearly indicated that the biomass power plant effluent has stimulatory effect on all the biochemical contents at lower concentration, and at higher concentration they have deleterious effects.

comparative evaluation of semen parameters in pre-and post-Hurricane Katrina human population

A natural disaster leading to accumulation of environmental contaminants may have substantial effects on the male reproductive system. Our aim was to compare and assess semen parameters in a normospermic population residing in the Southern Louisiana, USA area pre- and post-Hurricane Katrina. We retrospectively evaluated semen analyses data （n = 3452） of 1855 patients who attended the Tulane University Andrology/Fertility clinic between 1999 and 2013. The study inclusion criteria were men whose semen analyses showed -〉 1.5 ml volume; _〉15 million ml1 sperm concentration; _〉39 million total sperm count; _〉40% motility; 〉30% morphology, with an abstinence interval of 2-7 days. After the inclusion criteria applied to the population, 367 normospermic patients were included in the study. Descriptive statistics and group-based analyses were performed to interpret the differences between the pre-Katrina （Group 1, 1999-2005） and the post-Katrina （Group 2, 2006-2013） populations. There were significant differences in motility, morphology, number of white blood cell, immature germ cell count, pH and presence of sperm agglutination, but surprisingly there were no significant differences in sperm count between the two populations. This long-term comparative analysis further documents that a major natural disaster with its accompanied environmental issues can influence certain semen oarameters （e.g.. motility and morphology） and, bv extension, fertility potential of the population of such areas.

Age-related macular degeneration（AMD） is a leading cause of blindness and is becoming a global crisis since affected people will increase to 288 million by ***,age,diabetes,gender,obesity,hypertension,race,hyperopia,iris-color,smoking,sun-light and pyroptosis have varying roles in AMD,but oxidative stress-induced inflammation remains a significant driver of *** is a unique organ as it contains a remarkable oxygengradient that generates reactive oxygen species（ROS） which upregulates inflammatory *** becomes a source of functional and morphological impairments in retinal pigment epithelium（RPE）,endothelial cells and retinal ganglion *** demonstrated that hydrogen sulfide（H2S） acts as a signaling molecule and that it may treat ***,we propose a novel hypothesis that H2S may restore homeostasis in the eyes thereby reducing damage caused by oxidative injury and *** H_2S has been shown to be a powerful antioxidant because of its free-radicals＇ inhibition properties in addition to its beneficial effects in age-relatedconditions,therefore,patients may benefit from H2S salubrious effects not only by minimizing their oxidant and inflammatory injuries to retina but also by lowering retinal glutamate excitotoxicity.

Systemic treatment strategies for triple-negative breast cancer

Triple-negative breast cancer(TNBc) is defined by the lack of immunohistochemical expression of the estrogen and progesterone receptors and human epidermal growth factor receptor 2(EGFR2). Most TNBc has a basal-like molecular phenotype by gene expression profiling and shares clinical and pathological features with hereditary BRcA1 related breast cancers. This review evaluates the activity of available chemotherapy and targeted agents in TNBc. A systematic review of PubM ed and conference databases was carried out to identify randomised clinical trials reporting outcomes in women with TNBc treated with chemotherapy and targeted agents. Our review identified TNBc studies of chemotherapy and targeted agents with different mechanisms of action, including induction of synthetic lethality and inhibition of angiogenesis, growth and survival pathways. TNBc is sensitive to taxanes and anthracyclins. Platinum agents are effective in TNBc patients with BRcA1 mutation, either alone or in combination with poly adenosine diphosphate polymerase 1 inhibitors. combinations of ixabepilone and capecitabine have added to progression-free survival(PFS) without survival benefit in metastatic TNBc. Antiangiogenic agents, tyrosine kinase inhibitors and EGFR inhibitorsin combination with chemotherapy produced only modest gains in PFS and had little impact on survival. TNBc subgroups respond differentially to specific targeted agents. In future, the treatment needs to be tailored for a specific patient, depending on the molecular characteristics of their malignancy. TNBc being a chemosensitive entity, combination with targeted agents have not produced substantial improvements in outcomes. Appropriate patient selection with rationale combinations of targeted agents is needed for success.

The significant rise of ultra-violet（UV） radiation and pathogenic infectious bacteria poses a serious threat to global *** researchers' interests are attracted by novel materials with strong UVblocking ability,antibacterial activity and low toxicity to other *** this case,a simple wet chemical method with different annealing temperatures（400,500,and 600℃） was employed to create highly effective rare earth（Sm）-doped ZnO ***（101） plane of wurtzite ZnO shifts towards a lower angle with increasing annealing temperature,according to the X-ray diffraction（XRD） study findings,which additionally establishes the consequence of lattice *** of doublet peaks of Sm 3d(Sm 3d5/2and Sm 3d3/2) in the X-ray photoelectron spectroscopy（XPS） spectrum clearly validates the substitution of Sm3+ions in the 500℃-annealed *** 500℃-annealed nanorods exhibit combined performances of the wide band gap,improved UV absorbance,and vivid green luminescent emission（563 nm）.Additio nally,the nanorods have favorable UV-blocking execution of 96% for UVA at 360 nm,92% for UVB at 320 nm,and 57% for UVc at 225 nm,which is greater than the majority of ZnO-related materials that have been reported up to this *** doping is also appropriate for improving bacterial inhibition against the two studied strains（Escherichia coli and Staphylococcus aureus）,in addition to the intriguing features discussed ***,with maximum inhibition zone diameters of 20±0.72 and 18±0.57 mm,respectively,these nanorods exhibit high inhibitory action against *** and *** bacterial *** rare earth-doped material developed during the current experiment,which was annealed at 500℃,could potentially serve as an effective replacement for UV-blocking and antibacterial material,especially for biomedical applications.
