Preclinical and clinical evidence of the association of colibactinproducing Escherichia coli with anxiety and depression in colon cancer

BACKGROUND The association between the intestinal microbiota and psychiatric disorders is becoming increasingly *** gut microbiota contributes to colorectal carcinogenesis(CRC),as demonstrated with colibactin-producing Escherichia coli(CoPEC).AIM To evaluate the association between CoPEC prevalence and anxiety-and depressive-like behaviors with both preclinical and clinical *** Patients followed after a CRC surgery and for whom the prevalence of CoPEC has been investigated underwent a psychiatric *** were compared according to the CoPEC *** parallel C57BL6/J wild type mice and mice with a CRC susceptibility were chronically infected with a CoPEC *** behavior was assessed using the Elevated Plus Maze test,the Forced Swimming Test and the Behavior recognition system PhenoTyper®.RESULTS In a limited cohort,all patients with CoPEC colonization presented with psychiatric disorders several years before cancer diagnosis,whereas only one patient(17%)without CoPEC *** result was confirmed in C57BL6/J wildtype mice and in a CRC susceptibility mouse model(adenomatous polyposis colimultiple intestinal neoplasia/+).Mice exhibited a significant increase in anxiety-and depressive-like behaviors after chronic infection with a CoPEC *** This finding provides the first evidence that CoPEC infection can induce microbiota-gut-brain axis disturbances in addition to its procarcinogenic properties.

Among the impacts of climate change,there is the intensification of phenomena such as the El Niño South-ern Oscillation(ENSO)responsible for El Niño and La Niñ***,understanding their effects on the functional pro-cesses of forests is ***,this study evaluated the effects of ENSO on litter stock and water holding capac-ity(WHC)in a successional forest in eastern *** occurred in periods with the most rainfall in El Niño(2019)and least in La Niña(2021)*** permanent plots were used to sample *** effects were evident for WHC,higher during El Niñ***,this influence was not clear for litter,as only in the rainy season effects were *** was a positive correlation of WHC with precipitation and humidity,while litter stocks were negatively correlated with temperature and wind *** the subject of this study requires long-term assessments,preliminary results suggests that,depending on the intensity of ENSO,forest functional processes can be strongly impacted and *** conclusion reinforces warnings by the scientific community about the impacts of climate change on the maintenance of litter stocks,decomposition and,consequently,the biogeochemical cycle and essential ecosystem services for the maintenance of Amazonia *** need to develop long-term research to understand the effects of climatic change on litter stocks and water holding capacity is highlighted,especially in Amazonia.

Combined evaluation of biomarkers as predictor of maintained remission in Crohn's disease

BACKGROUND The individual performances and the complementarity of Crohn's disease(CD)activity index(CDAI), C-reactive protein(CRP) and faecal calprotectin(Fcal) to monitor patients with CD remain poorly inves-tigated in the era of "tight control"and "treat to target" *** To assess CDAI, CRP and Fcal variation, alone or combined, after 12 wk(W12) of anti-tumor necrosis factor(TNF) therapy to predict corticosteroids-free remission(CFREM = CDAI 150 and either CRP >2.9 mg/L or Fcal > 250 μg/g were prospectively *** Among the 40 included patients, 13 patients(32.5%) achieved CFREM at W52. In univariable analysis, CDAI < 150 at W12(P = 0.012), CRP level < 2.9 mg/L at W12(P = 0.001) and Fcal improvement at W12(Fcal < 300 μg/g; or, for patients with initial Fcal < 300 μg/g, at least 50% decrease of Fcal or normalization of Fcal(< 100 μg/g)(P = 0.001) were predictive of CFREM at W52. Combined endpoint(CDAI < 150 and CRP ≤ 2.9 mg/L and FCal improvement) at W12 was the best predictor of CFREM at W52 with positive predictive value = 100.0%(100.0-100.0)and negative predictive value = 87.1%(75.3-98.9). In multivariable analysis, Fcal improvement at W12 [odd ratio(OR) = 45.1(2.96-687.9); P = 0.03] was a better predictor of CFREM at W52 than CDAI < 150 [OR = 9.3(0.36-237.1); P = 0.145]and CRP < 2.9 mg/L(0.77-278.0; P = 0.073).CONCLUSION The combined monitoring of CDAI, CRP and Fcal after anti-TNF induction therapy is able to predict favorable outcome within one year in patients with CD.

3D-Parallel Simulation of Contaminant in Waste Disposals

In this work the authors simulate a contaminant transport problem in three dimensions that takes place in the soil of waste disposals. Such problem is modeled by a diffusion-dominated equation. The solution of this equation is addressed by using mixed finite element method for the spatial discretization of the equation. The resulting linear algebraic system is handled by an iterative domain decomposition procedure. This procedure is naturally parallelizable, and permits to implement computational codes in distributed memory machines in order to save on CPU time. Numerical results of the serial and parallel codes were compared with experimental results, and their performance measures were evaluated. The results indicate that the parallelizable procedure is an efficient tool for performing simulations of the problem.

Faecal calprotectin and magnetic resonance imaging in detecting Crohn's disease endoscopic postoperative recurrence

AIM To assess magnetic resonance imaging(MRI) and faecal calprotectin to detect endoscopic postoperative recurrence in patients with Crohn's disease(CD).METHODS From two tertiary centers, all patients with CD who underwent ileocolonic resection were consecutively and prospectively included. All the patients underwent MRI and endoscopy within the first year after surgery or after the restoration of intestinal continuity [median = 6 mo(5.0-9.3)]. The stools were collected the day before the colonoscopy to evaluate faecal calprotectin level. Endoscopic postoperative recurrence(POR) was defined as Rutgeerts' index ≥ i2b. The MRI was analyzed independently by two radiologists blinded from clinical *** Apparent diffusion coefficient(ADC) was lower in patients with endoscopic POR compared to those with no recurrence(2.03 ± 0.32 vs 2.27 ± 0.38 × 10^(-3) mm^2/s, P = 0.032). Clermont score(10.4 ± 5.8 vs 7.4 ± 4.5, P = 0.038) and relative contrast enhancement(RCE)(129.4% ± 62.8% vs 76.4% ± 32.6%, P = 0.007) were significantly associated with endoscopic POR contrary to the magnetic resonance index of activity(Ma RIA)(7.3 ± 4.5 vs 4.8 ± 3.7; P = 0.15) and MR scoring system(P = 0.056). ADC 100%(sensitivity = 0.75, specificity = 0.81, PPV = 0.75, NPV = 0.81) were the best cutoff values to identify endoscopic POR. Clermont score > 6.4(sensitivity = 0.61, specificity = 0.82, PPV = 0.73, NPV = 0.74), Ma RIA > 3.76(sensitivity = 0.61, specificity = 0.82, PPV = 0.73, NPV = 0.74) and a MR scoring system ≥ MR1(sensitivity = 0.54, specificity = 0.82, PPV = 0.70, and NPV = 0.70) demonstrated interesting performances to detect endoscopic POR. Evaluation of Physicochemical and Functional Properties of Protein Recovered Obtaining from Whitemouth Croaker (<i>Micropogonias furnieri</i>) Byproducts

Proteins recovered were obtained from Whitemouth Croaker (Micropogonias furnieri) byproducts and their physico-chemical and functional properties were evaluated. The proximate composition, presented 8.64% ± 0.10% of moisture, 85.33% ± 0.12% of protein, 2.69% ± 0.09% of ashes and 2.16% ± 0.12% of lipids, in dry basis. The highest solubility was obtained in pH 11 (93.24%), the maximum water holding capacity was presented at pH 11 (25.71 mL H2O/gprotein), the oil holding capacity was 13.71 mL/gprotein and the digestibility in vitro was 91.32% ± 0.15%. The electro-phoretic profile was observed typical of the myofibrillar proteins, with the appearance of the heavy chain of myosin (220 kDa) and actin (50 kDa). The results show that the products of low commercial value of fish that are usually used for the production of animal feed or simply discarded, contributing to environmental pollution, may be used to produce products with a greater added value.

Analysis of the Liver of Fish Species <i>Prochilodus lineatus</i>Altered Environments, Analyzed with ImageJ

The fish Prochilodus lineatus (Characiformes, Prochilodontidae), in addition to being a good bioindicator, is also of economic and ecological importance with a broad distribution in the neotropics. Ecotoxicology examines the interaction between environmental chemistry and biota;and in this study we assess alterations of bile and glycogen levels in the fish liver, organ responsible for detoxification, biotransformation and storing nutrients, such as glycogen, and for secreting bile. Fish were separated in three groups to examine the damage caused by the exposure to waters from Lago Azul-Rio Claro-SP and containing diluted biodegradable detergents in comparison to a control group (chlorinated water from an artesian well of UNESP-Campus Rio Claro). A histological analysis was performed on HE and PAS stained sections. The identification of structural changes and the assessment of the area occupied by bile and glycogen were carried out with the software ImageJ, showing that the liver was affected morphologically (cell vacuolization, peripherals nuclei, for example) and problems in bile release and production and storage of glycogen.

Effects of Ingested Baccharis dracunculifolia D.C.(Asteraceae)Extract in the Liver of Prochilodus lineatus Fish

Baccharis dracunculifolia, popularly known in Brazil as “alecrim-do-campo”, is widely recognized for its therapeutic potential. The extract of its leaves is used for liver problems, stomach disorders and others. The objective of the present study was to perform a histochemical analysis of curimbata fish livers to evaluate the potential effects and risks of the ingestion of B. dracunculifolia. Thirty-two animals were divided into two experimental groups in duplicate: Control group (regular food) and B. dracunculifolia Treated group (food added with B. dracunculifolia). The fishes were collected on the 14th and 21st days after the treatment period of 21 days. The histological alterations were evaluated using the semiquantitative methods Mean Value of Alterations (MVA), Histopathological Alteration Index (HAI) and Image J®. HAI and MAV showed that the extract caused slight but statistically significant damages, widely distributed throughout the organ. The results showed significant hepatic alterations caused by the ingestion of B. dracunculifolia extract.

Fish populations are sensitive to environmental impact resulting from many factors, as an introduction of exotic species, industrial or residual waste, oil spills, pesticides or other agents that can directly affect the ecology and species survival. Astyanax altiparanae is pelagic and active swimmer specie and can have serious implications on its way of life as changes in muscle structure of fish may occur since the presence of pollutants in the water. In this study we analyze the alterations in Muscle and collagen fibers of the pectoral muscles and pollutants exposed flows contained in water with biodegradable detergents, and water coming from the Blue Lake, River clear-SP (2o24'39"S, 47o33'39"E) diluted. Effects of these exposures were analyzed during the period of seven and thirty days. Compared with acclimatized fish (Astyanax altiparanae) with pure water from an artesian well, the muscle fibers morphology and collagen concentration on them have changed due to the exposure to urban pollutants and biodegradable detergents.

Clinical outcomes of tricuspid valve repair accompanying left-sided heart disease

AIM To determine whether the need for additional tricuspid valve repair is an independent risk factor when surgery is required for a left-sided heart *** One hundred and eighty patients(68 ± 12 years,79 males) underwent tricuspid *** proportionalhazards regression model for multivariate analysis was performed for variables found significant in univariate *** Tricuspid regurgitation etiology was functional in 154 cases(86%),organic in 16 cases(9%),and mixed in10 cases(6%),*** mortality at 30 days was 11.7%.Mean follow-up was 51.7 mo with survival at 5 years of 73.5%.Risk factors for mortality were acute endocarditis [hazard ratio(HR) = 9.22(95%CI:2.87-29.62),P < 0.001],ischemic heart disease requiring myocardial revascularization [HR = 2.79(1.26-6.20),P = 0.012],and aortic valve stenosis [HR = 2.6(1.15-5.85),P = 0.021].Significant predictive factors from univariate analyses were double-valve replacement combined with tricuspid annuloplasty [HR = 2.21(1.11-4.39),P = 0.003] and preoperatively impaired ejection fraction [HR = 1.98(1.04-3.92),P = 0.044].However,successful mitral valve repair showed a protective effect [HR = 0.32(0.10-0.98),P = 0.046].Additionally,in instances where tricuspid regurgitation required the need for concomitant tricuspid valve repair,mortality predictor scores such as Euroscore 2 could be shortened to a simple Euroscore-tricuspid comprised of only 7 *** explanation may lie in the fact that significant tricuspid regurgitation following leftsided heart disease represents an independent risk factor encompassing several other factors such as pulmonary arterial hypertension and *** Tricuspid annuloplasty should be used more often as a concomitant procedure in the presence of relevant tricuspid regurgitation,although it usually reveals an overly delayed correction of a left-sided heart disease.
