Analysis of the Arabidopsis Floral Proteome:Detection of over 2000 Proteins and evidence for Posttranslational Modifications

The proteome of the Arabidopsis flower has not been extensively studied previously. Here, we report a proteomic analysis of the wild type Arabidopsis flower. Using both two-dimensional electrophoresis/mass spectrometry （2-DGeIMS） and multi-dimensional protein identification technology （MudPIT） approaches, we identified 2 446 proteins. Although a single experiment or analysis uncovered only a subset of the proteins we identified, a combination of multiple experiments and analyses facilitated the detection of a greater number of proteins. When proteins are grouped according to RNA expression levels revealed by microarray experiments, we found that proteins encoded by genes with relatively high levels of expression were detected with greater frequencies. On the other hand, at the level of the individual gene/protein, there was not a good correlation between protein spot intensity and microarray values. We also obtained strong evidence for post-translational modification from 2-DGe and MudPIT data. We detected proteins that are annotated to function in protein synthesis, folding, modification, and degradation, as well as the presence of regulatory proteins such as transcription factors and protein kinases. Finally, sequence and evolutionary analysis of genes for active methyl group metabolisms suggests that these genes are highly conserved. Our results allow the formulation of hypotheses regarding post-translational regulation of proteins in the flower, providing new understanding about Arabidopsis flower development and physiology.

Correlation between Microstructures and Yield Strength of a High Strength enameling Steel

Numerical estimates of the components of yield strength of a high strength Fe-C-Mn-P-N-Si enameling steel were determined using empirical relationships between microstructure and yield strength. Results are reported for both the hot rolled （HR） and cold rolled （CR） forms before and after simulating an enamel-fire anneal （eFA）. To determine the solubilities of alloying elements, thermodynamic calculations were performed in combination with the considerations of process conditions and the element diffusivities. The results show that the main solid solution strengtheners were the elements Mn, Si, and P, while the elements C and N were nearly completely tied up as precipitates. The yield strength reduction, due to the eFA, resulted primarily from an increase in grain size and a decrease in dislocation density, and the eFA appeared to have a negligible effect on the element solubilities.

Objective: The prevalence of sport-related concussion(SRC) is high and results in a number of serious health consequences. One area that has received minimal research is the relationship between SRC and sleep. The literature shows that sleep deficiency is a frequent negative consequence of SRC. At the same time, sleep deficiency delays recovery from SRC and contributes added risk of symptom recurrence.A 2014 study of chronic pain patients who learned to apply the complementary and alternative medicine intervention hand self-shiatsu(HSS) had promising, sleep-promoting results that warrant further investigation with other populations. This proof-of-concept study explored the feasibility of HSS as an intervention to promote sleep onset and continuity for young adults with ***: This study employed a prospective case-series design, where participants act as their own controls. Baseline and follow-up data included standardized self-reported assessment tools and sleep ***: Seven athletes, aged between 18 and 25 years, participated in the study. Although statistically significant improvement in actigraphy sleep scores between baseline and follow-up was not achieved,metrics for sleep quality and daytime fatigue showed significant ***: These findings support the hypothesis that HSS has the potential to improve sleep and reduce daytime fatigue in young postconcussion athletes. This pilot study provides guidance to refine research protocols and lays a foundation for further, large-sample, controlled studies.

Abstract High altitude test facilities are required to test the high area ratio nozzles operating at the upper stages of rocket in the nozzle full flow conditions. It is typically achieved by creating the ambient pressure equal or less than the nozzle exit pressure. On average, air/GN2 is used as active gas for ejector system that is stored in the high pressure cylinders. The wind tunnel facilities are used for conducting aerodynamic simulation experiments at/under various flow velocities and operating conditions. However, constructing both of these facilities require more laboratory space and expensive instruments. Because of this demerit, a novel scheme is implemented for conducting wind tunnel experiments by using the existing infrastructure available in the high altitude testing （HAT） facility. This article presents the details about the methods implemented for suitably modifying the sub-scale HAT facility to conduct wind tunnel experiments. Hence, the design of nozzle for required area ratio A/A＊, realization of test section and the optimized configuration are focused in the present analysis. Specific insights into various rocket models including high thrust cryogenic engines and their holding mechanisms to conduct wind tunnel experiments in the HAT facility are analyzed. A detailed CFD analysis is done to propose this conversion without affecting the existing functional requirements of the HAT facility.

AIM: To construct a global "metabolic phenotype" of pancreatic ductal adenocarcinoma(PDAC) reflecting tumour-related metabolic enzyme ***: A systematic review of the literature was performed using Ovid SP and Pub Med databases using keywords "pancreatic cancer" and individual glycolytic and mitochondrial oxidative phosphorylation(MOP) enzymes. Both human and animal studies investigating the oncological effect of enzyme expression changes and inhibitors in both an in vitro and in vivo setting were included in the review. Data reporting changes in enzyme expression and the effects on PDAC cells, such as survival and metastatic potential, were extracted to construct a metabolic phenotype. ReSULTS: Seven hundred and ten papers were initially retrieved, and were screened to meet the review inclusion criteria. 107 unique articles were identified as reporting data involving glycolytic enzymes, and 28 articles involving MOP enzymes in PDAC. Data extraction followed a pre-defined protocol. There is consistent over-expression of glycolytic enzymes and lactate dehydrogenase in keeping with the Warburg effect to facilitate rapid adenosine-triphosphate production from glycolysis. Certain isoforms of these enzymes were over-expressed specifically in PDAC. Altering expression levels of HK, PGI, FBA, enolase, PK-M2 and LDA-A with metabolic inhibitors have shown a favourable effect on PDAC, thus identifying these as potential therapeutic targets. However, the Warburg effect on MOP enzymes is less clear, with different expression levels at different points in the Krebs cycle resulting in a fundamental change of metabolite levels, suggesting that other essential anabolic pathways are being stimulated. CONCLUSION: Further characterisation of the PDAC metabolic phenotype is necessary as currently there are few clinical studies and no successful clinical trials targeting metabolic enzymes.
