A dominant Allele of Arabidopsis Pectin-Binding Wall-Associated Kinase Induces a Stress Response Suppressed by MPK6 but Not MPK3 Mutations

The plant cell wall is composed of a matrix of cellulose fibers, flexible pectin polymers, and an array of as- sorted carbohydrates and proteins. The receptor-like Wall-Associated Kinases （WAKs） of Arabidopsis bind pectin in the wall, and are necessary both for cell expansion during development and for a response to pathogens and wounding. Mitogen Activated Protein Kinases （MPKs） form a major signaling link between cell surface receptors and both transcrip- tional and enzyme regulation in eukaryotes, and Arabidopsis MPK6 and MPK3 indeed have important roles in develop- ment and the response to stress and pathogens. A dominant allele of WAK2 requires kinase activity and activates a stress response that includes an increased ROS accumulation and the up-regulation of numerous genes involved in pathogen resistance, wounding, and cell wall biogenesis. This dominant allele requires a functional pectin binding and kinase domain, indicating that it is engaged in a WAK signaling pathway. A null mutant of the major plasma membrane ROS-producing enzyme complex, rbohd/f does not suppress the WAK2cTAP-induced phenotype. A mpk6, but not a mpk3, null allele is able to suppress the effects of this dominant WAK2 mutation, thus distinguishing MPK3 and MPK6, whose activity previously was thought to be redundant. Pectin activation of gene expression is abated in a wak2-null, but is tempered by the WAK-dominant allele that induces elevated basal stress-related transcript levels. The results suggest a mechanism in which changes to the cell wall can lead to a large change in cellular responses and help to explain how pathogens and wounding can have general effects on growth.

A series of 60~nm thick indium oxide thin-films,all amorphous as determined by x-ray diffraction,were found to have physical and electrical properties that depended on the temperature of *** carrier mobility and flm conductivity decreased with decreasing deposition temperature;the best electrical properties of high mobility and conductivity were observed at a deposition temperature just below the temperature at which crystalline films *** density of the flm also decreased with deposition temperature from 7.2g/cm3 at+50℃ to 5.3g/cm3 at 100℃.

New insights into the vitamin d requirements during pregnancy

Pregnancy represents a dynamic period with physical and physiological changes in both the mother and her developing fetus. The dramatic 2-3 fold increase in the active hormone 1,25（OH）2d concentrations during the early weeks of pregnancy despite minimal increased calcium demands during that time of gestation and which are sustained throughout pregnancy in both the mother and fetus suggests an immunomodulatory role in preventing fetal rejection by the mother. While there have been numerous observational studies that support the premise of vitamin d＇s role in maintaining maternal and fetal well-being, until recently, there have been few randomized clinical trials with vitamin d supplementation. One has to exhibit caution, however, even with RCTs, whose results can be problematic when analyzed on an intent-to-treat basis and when there is high non-adherence to protocol （as if often the case）, thereby diluting the potential good or harm of a given treatment at higher doses. As such, a biomarker of a drug or in this case ＂vitamin＂ or pre-prohormone is better served. For these reasons, the effect of vitamin d therapies using the biomarker circulating 25（OH）d is a far better indicator of true ＂effect.＂ When pregnancy outcomes are analyzed using the biomarker 25（OH）d instead of treatment dose, there are notable differences in maternal and fetal outcomes across diverse racial/ethnic groups, with improved health in those women who attain a circulating 25（OH）d concentration of at least 100 nmol. L-1 （40 ng. mL-1）. Because ,an important issue is the timing or initiation of vitamin d treatment/supplementation, and given the potential effect of vitamin d on placental gene expression and its effects on inflammation within the placenta, it appears crucial to start vitamin d treatment before placentation （and trophoblast invasion）; however, this question remains unanswered. Metabolic Engineering of Plant-derived (E)-β-farnesene Synthase Genes for a Novel Type of Aphid-resistant Genetically Modified Crop Plants

Aphids are major agricultural pests that cause significant yield losses of crop plants each year. Excessive dependence on insec- ticides for long-term aphid control is undesirable because of the development of insecticide resistance, the potential negative effects on non-target organisms and environmental pollution. Transgenic crops engineered for resistance to aphids via a non-toxic mode of action could be an efficient alternative strategy. （E）-β-Farnesene （EβF） synthases catalyze the formation of EβF, which for many pest aphids is the main component of the alarm pheromone involved in the chemical communication within these species. EβF can also be synthesized by certain plants but is then normally contaminated with inhibitory compounds. Engineering of crop plants capable ofsynthesizing and emitting EβF could cause repulsion of aphids and also the attraction of natural enemies that use EβF as a foraging cue, thus minimizing aphid infestation. In this review, the effects of aphids on host plants, plants＇ defenses against aphid herbivory and the recruitment of natural enemies for aphid control in an agricultural setting are briefly introduced. Furthermore, the plant-derived EβF synthase genes cloned to date along with their potential roles in generating novel aphid resistance via genetically modified approaches are discussed.

Quantification of soluble epoxide hydrolase inhibitors in experimental and clinical samples using the nanobody-based ELISA

To ensure proper dosage of a drug,analytical quantification of it in biofluid is *** chromatography mass spectrometry(LC-MS)is the conventional method of choice as it permits accurate identification and ***,it requires expensive instrumentation and is not appropriate for bedside *** soluble epoxide hydrolase(sEH)inhibitors(EC5026 and TPPU)as examples,we report development of a nanobody-based enzyme-linked immunosorbent assay(ELISA)for such small molecules and its use to accurately quantify the drug chemicals in human *** optimized conditions,two nanobody-based ELISAs were successfully established for EC5026 and TPPU with low limits of detection of 0.085 ng/mL and 0.31 ng/mL,respectively,and two order of magnitude linear ranges with high precision and *** assay was designed to detect parent and two biologically active metabolites in the investigation of a new drug candidate *** addition,the ELISAs displayed excellent correlation with LC-MS analysis and evaluation of inhibitory *** results indicate that nanobody-based ELISA methods can efficiently analyze drug like *** methods could be easily implemented by the bedside,in the field in remote areas or in veterinary *** work illustrates that nanobody based assays offer alternative and supplementary analytical tools to mass spectrometry for monitoring small molecule medicines during clinical development and *** of nanobody based pharmaceutical assays are discussed.

Assessment of Risk of Establishment of Wheat dwarf Bunt (Tilletia controversa) in China

Wheat dwarf bunt, caused by Tilletia controversa Kühn （TCK）, is an important quarantine wheat disease throughout the world. Based on published research results of the biology and the epidemiology of the disease, the main factors including temperature, humidity, snow cover, and their parameters relating to teliospore germination, infection and epidemics of TCK were determined. The geophytopathological models for the risk analysis of wheat dwarf bunt establishment were modified. Fifty-year meteorologic data from about 500 weather stations in China were used to calculate the probabilities of TCK establishment in different geographic sites with the models. The map that displays the establishment risk of TCK in winter wheat growing regions in China was generated by using geographical information system （GIS）. The zones showing high, moderate, low, and very low, including no risk, of TCK establishment accounted for 27.33, 27.69, 38.12, and 6.86% of total winter wheat growing areas in China, respectively. These results will provide useful information to formulate quarantine regulations and wheat importation policy in China.

Performance enhancement of futuristic airplanes by nature inspired biomimetic fish scale arrays--A design approach

Biomimetics has an immense potential to drive the next generation of technologies forward by propounding competent solutions from *** decades,the nonsmooth topography of most living creatures has been critical to their existence and *** to human-made smooth surfaces,when adapted for fluid flow applications,nonsmooth surfaces can enhance the overall aerodynamic performance by reducing the drag ***,the bioinspired scale structure of fish has been identified as a key biomimetic derivative for improving the aerodynamic efficiency in various cross-domain *** study investigates the aerodynamics of a fish scale array(FSA)NACA 0021 model at a specific Reynolds number(Re)of approximately 2.46×105 that is meant for laminar flow conditions using Computational Fluid dynamics(CFd)tools and a subsonic wind tunnel facility.A 3d printed biomimetic FSA film is developed and affixed on the NACA 0021 wing profile for the experimental investigation,followed by flow visualization through the smoke tunnel *** proved qualitatively under specific aerodynamic conditions,the creation of velocity streaks has a major role in the drag reduction *** obtain a clear perspective of flow across the overlapping fish scale structures,the results are focused on the central and overlapping regions of the FSA *** experiment has proved the existence of a combined formation of low and high velocity streaks in central and overlapping *** FSA 0021 model showed a maximum drag reduction of 9.57%,which was attributed to the streak formation phenomenon observed in the overlapping FSA configuration.

Analysis of Bio-inspired Fishbone Based Corrugated Rib for Adaptive Camber Morphing

Bioinspired active camber morphing is an innovative solution for the aerodynamic performance enhancement of flight vehicles such as Micro Aerial Vehicles(MAVs)and Unmanned Aerial Vehicles(UAVs).In the present article,a bio-inspired Fish Bone Active Camber(FishBAC)corrugated rib design concept with and without a spine for an Unmanned Aerial Vehicle(UAV)is *** wing model is composed of multiple corrugated ribs and a splitted spar for connecting each *** rib geometry is subjected to static structural analysis using ANSYS Finite Element Analysis(FEA)module under the unit load *** deformation modes are first extracted from the solution of the wind tunnel test and the elastically deformed NACA 4412 profile will be used to perform Fluid-Structure Interaction(FSI)studies as a partly coupled *** corrugated rib with spine design is a stable structure as inspired from nature species that can withstand elastic and inertial *** fluid dynamics(CFd)simulation is performed to compute the coefficient of lift(Cl),coefficient of drag(Cd)at different Angle of Attack(AoA)for a modified NACA 4412 airfoil and the pressure loads acting on the deformed shape is *** Cl obtained for the modified airfoil at lower AoA is about 60–80%higher and the maximum Cl(Clmax)is 62%higher than the baseline *** Cd of modified airfoil is reduced about 20%at the AoAα=3°as compared with the baseline *** proposed corrugated rib structure has been successfully 3d printed with Polylactic Acid plus(PLA+)material and the wind tunnel testing is done to validate the Cl,Cd values obtained through CFd simulations and the results are presented.
